Application of microfluidic chip technology in pharmaceutical analysis:A review显示文摘The development of pharmaceutical analytical methods represents one of the most significant aspects of drug development. Recent advances in microfabrication and microfluidics could provide new approaches for drug analysis, including drug screening, active testing and the study of metabolism. Microfluidic chip technologies, such as lab-on-a-chip technology, three-dimensional (3D) cell culture, organs-on-chip and droplet techniques, have all been developed rapidly. Microfluidic chips coupled with various kinds of detection techniques are suitable for the high-throughput screening, detection and mechanistic study of drugs. This review highlights the latest (2010–2018) microfluidic technology for drug analysis and discusses the potential future development in this field. | Ping Cui Sicen Wang | 2019 | Journal of Pharmaceutical Analysis2019,9,4: | 8
Imitation of drug metabolism in cell co-culture microcapsule model using a microfluidic chip platform coupled to mass spectrometry显示文摘In this work,a multi-functional analysis platform by coupling a microfluidic chip to a mass spectrometry(MS) detector was described. We constructed a three-dimensional tumor-endothelial co-culture model for simulating drug resistance during tumor treatment. On this specially designed integrated platform, the first step was to prepare heterogeneous cell-encapsulated alginate microcapsules for threedimensional co-culture,and the second step was to achieve on-line perfusion culture and continuous drug stimulation on chip.It facilitates cell proliferation analysis and the collection of metabolism medium. After micro solid phase extraction column(SPE) pretreatment, subsequent mass spectrometry could detect drug metabolism. The high activity of two kinds of cells(A549 and HUVEC) shows the biocompatibility of the platform.Paclitaxel was used as a model drug, the distinctions of drug absorption between the mono-culture group and co-culture group were clearly observed by electrospray ionization quadrupole time-of-flight mass spectrometry(ESI-Q-TOF MS).Therefore, the integrated platform has shown promise as a high throughput, low cost for cell metabolism research and drug screening processes. | Yajing Zheng Zengnan Wu Jin-Ming Lin Ling Lin | 2020 | Chinese Chemical Letters2020,31,2: | 4
In situ fabrication of organic electrochemical transistors on a microfluidic chip显示文摘Microfluid chips integrating with organic electrochemical transistors (OECTs) are useful for manufacturing biosensors with high throughput and large-scale analyses. We report here the utilization of alternating current (AC) electrodeposition to fabricate OECTs in situ on a microfiuid chip. With this method, the organic semiconductor (OS) layer with a channel length of 8μm was readily prepared without requiring the postbonding process in the conventional construction of microfluidic chips. Poly(3,4-ethylenedioxythiophene):poly(4-styrenesulfonate)/graphene quantum dots (PEDOT:PSS/GQDs) composites with different morphologies, such as microfilms, nanodendrites and nanowires were electropolymerized. The mass transfer process of the electropolymerization reaction was evidenced to be diffusion limited. Morphologies, growth directions, and chemical structures of OS layers could be tuned by the amplitude and the frequency of the AC voltage. Transfer and output characteristic curves of OECTs were measured on the microfluidic chip. The maximum transconductance, on/off current ratio and threshold voltage measured in the experiment was 1.58 mS, 246, and 0.120 V, respectively. | Jianlong Ji Mangmang Li Zhaowei Chen Hongwang Wang Xiaoning Jiang Kai Zhuo Ying Liu Xing Yang Zhen Gu Shengbo Sang Yang Shu | 2019 | Nano Research2019,12,8: | 3
Microfluidic Chip Method for Multi-SNPs Genotyping in Individual Risk Assessment of Micronutrient Deficiency显示文摘Research on nutrigenomics has accumulated sufficient data in the past two decades that have dem on strated phe no types of single n ucleotide polymorphisms (SNPs) betwee n healthy and micronutrient-deficient populations. For instance, Zhang et al. showed that the genes MTHFR C677T, MTRR A66G, and MTR A2756G were the genetic factors resp on sible for low absorptio n and bioavailability of vitamins such as folate, B6/ and B12. It has also been reported that these nutrients are closely associated with the prevale nee of neural tube defects in newborn infants。 | ZHANG Chun Hong HUO Jun Sheng CHEN Shan XU You Chun SUN Jing HUANG Jian | 2019 | Biomedical and Environmental Sciences2019,32,6: | 0
Impacts of Wettability on Immiscible Fluid Flow Pattern-Microfluidic Chip Experiment显示文摘Immiscible fluid flow is frequently found in resource recovery or soil remediation. The flow pattern in the porous media is affected by the wettability of pore surface. In this study, quartz substrates and microfluidic chips are treated by silica nanoparticles(SNP) and triethoxy(octyl)silane(TES) to fabricate the water-wet and oil-wet surfaces. The wettability of the treated-surface is measured in term of contact angle for several combinations of surrounding fluid and a liquid droplet. The effect of the wettability on the fluid flow pattern is explored by injecting oil and water alternately into the surface-treated microfluidic chips. The results reveal that the SNP-coated quartz substrate shows strong water-wet property and the TES coating makes water-repellent/oil-wet surfaces. In addition, it is found that the maximum and minimum oil and water saturation during alternate injection of oil and water depends on the surface wettability of the microfluidic chips. The characteristics of the pore-scale fluid flow pattern are also described. | Zheng Xianglei Jang Jaewon | 2019 | China Petroleum Processing & Petrochemical Technology2019,21,1: | 0
Development of a Miniature Permanent Magnetic Circuit for Nuclear Magnetic Resonance Chip显示文摘The existing researches of miniature magnetic circuits focus on the single-sided permanent magnetic circuits and the Halbach permanent magnetic circuits. In the single-sided permanent magnetic circuits, the magnetic flux density is always very low in the work region. In the Halbach permanent magnetic circuits, there are always great difficulties in the manufacturing and assembly process. The static magnetic flux density required for nuclear magnetic resonance(NMR) chip is analyzed based on the signal noise ratio(SNR) calculation model, and then a miniature C-shaped permanent magnetic circuit is designed as the required magnetic flux density. Based on Kirchhoff's law and magnetic flux refraction principle, the concept of a single shimming ring is proposed to improve the performance of the designed magnetic circuit. Using the finite element method, a comparative calculation is conducted. The calculation results demonstrate that the magnetic circuit improved with a single shimming has higher magnetic flux density and better magnetic field homogeneity than the one improved with no shimming ring or double shimming rings. The proposed magnetic circuit is manufactured and its experimental test platform is also built. The magnetic flux density measured in the work region is 0.7 T, which is well coincided with the theoretical design. The spatial variation of the magnetic field is within the range of the instrument error. At last, the temperature dependence of the magnetic flux density produced by the proposed magnetic circuit is investigated through both theoretical analysis and experimental study, and a linear functional model is obtained. The proposed research is crucial for solving the problem in the application of NMR-chip under different environmental temperatures. | LU Rongsheng YI Hong WU Weiping NI Zhonghua | 2013 | Chinese Journal of Mechanical Engineering2013,26,4: | 0
