3Two-dimensional optical spatial differentiation and high-contrast imaging显示文摘Optical analog signal processing technology has been widely studied and applied in a variety of science and engineering fields, with the advantages of overcoming the low-speed and high-power consumption associated with its digital counterparts. Much attention has been given to emerging metasurface technology in the field of optical imaging and processing systems. Here, we demonstrate, for the first time, broadband two-dimensional spatial differentiation and high-contrast edge imaging based on a dielectric metasurface across the whole visible spectrum. This edge detection method works for both intensity and phase objects simply by inserting the metasurface into a commercial optical microscope. 6In situ fabrication of organic electrochemical transistors on a microfluidic chip显示文摘Microfluid chips integrating with organic electrochemical transistors (OECTs) are useful for manufacturing biosensors with high throughput and large-scale analyses. We report here the utilization of alternating current (AC) electrodeposition to fabricate OECTs in situ on a microfiuid chip. With this method, the organic semiconductor (OS) layer with a channel length of 8μm was readily prepared without requiring the postbonding process in the conventional construction of microfluidic chips. Poly(3,4-ethylenedioxythiophene):poly(4-styrenesulfonate)/graphene quantum dots (PEDOT:PSS/GQDs) composites with different morphologies, such as microfilms, nanodendrites and nanowires were electropolymerized. The mass transfer process of the electropolymerization reaction was evidenced to be diffusion limited. Morphologies, growth directions, and chemical structures of OS layers could be tuned by the amplitude and the frequency of the AC voltage. Transfer and output characteristic curves of OECTs were measured on the microfluidic chip. 7Scattered seeding of CAR T cells in solid tumors augments anticancer efficacy显示文摘Chimeric antigen receptor T cell(CAR T) therapy was a milestone in the treatment of relapsed and refractory B cell malignancies. However, beneficial effects of CAR T cells have not been obtained in solid tumors yet. Herein, we implement a porous microneedle patch that accommodates CAR T cells and allows in situ penetration-mediated seeding of CAR T cells when implanted in the tumor bed or in the post-surgical resection cavity. CAR T cells loaded in the pores of the microneedle tips were readily escorted to the tumor in an evenly scattered manner without losing their activity. Such microneedle-mediated local delivery enhanced infiltration and immunostimulation of CAR T cells as compared to direct intratumoral injection. 8OsbZIP72 Is Involved in Transcriptional Gene-Regulation Pathway of Abscisic Acid Signal Transduction by Activating Rice High-Affinity Potassium Transporter OsHKT1;1显示文摘We created CRISPR-Cas9 knock-out and overexpressing OsbZIP72 transgenic rice plants to gain a better understanding of the role and molecular mechanism of OsbZIP72 gene in stress tolerance,which has remained largely elusive.OsbZIP72 was expressed and integrated into rice transgenic plant genomes,and the OsbZIP72 transcript in overexpression lines was elicited by salinity,abscisic acid(ABA)and drought stresses.OsbZIP72 overexpressing plants showed higher tolerance to drought and salinity stresses,while knock-out transgenic lines showed higher sensitivity to these stresses.The differentially expressed genes(DEGs)from RNA-sequencing data encompassed several abiotic stress genes,and the functional classification of these DEGs demonstrated the robust transcriptome diversity in OsbZIP72.Yeast one-hybrid,along with luciferase assay,indicated that OsbZIP72 acted as a transcriptional initiator.Remarkably,electrophoresis mobility assay revealed that OsbZIP72 bound directly to the ABAresponsive element in the OsHKT1;1 promoter region and activated its transcription.Overall,our findings revealed that OsbZIP72 can act as a transcriptional modulator with the ability to induce the expression of OsHKT1;1 in response to environmental stress through an ABA-dependent regulatory pathway,indicating that OsbZIP72 can play a crucial role in the ABA-mediated salt and drought tolerance pathway in rice.WANG Baoxiang LIU Yan WANG Yifeng LI Jingfang SUN Zhiguang CHI Ming XING Yungao XU Bo YANG Bo LI Jian LIU Jinbo CHEN Tingmu FANG Zhaowei LU Baiguan XU Dayong Babatunde Kazeem BELLO 2021Rice science2021,28,3:2

13Immunotherapy in leukaemia显示文摘Leukaemia is the common name for a group of malignant diseases of the haematopoietic system with complex classifications and characteristics.Remarkable progress has been made in basic research and preclinical studies for acute leukaemia compared to that of the many other types/subtypes of leukaemia,especially the exploration of the biological basis and application of immunotherapy in acute myeloid leukaemia(AML)and B-cell acute lympho-blastic leukaemia(B-ALL).In this review,we summarize the basic approaches to immunotherapy for leukaemia and focus on the research progress made in immunotherapy development for AML and ALL.Importantly,despite the advances made to date,big challenges still exist in the effectiveness of leukaemia immunotherapy,especially in AML.Therefore,we use AML as an example and summarize the mechanisms of tumour cell immune evasion,describe recently reported data and known therapeutic targets,and discuss the obstacles in finding suitabie treatment targets and the results obtained in recent clinical trials for several types of single and combination immunotherapies,such as bispecific antibodies,cell therapies(CAR-T-cell treatment),and checkpoint blockade.Finally,we summarize novel immunotherapy strategies for treating lymphocytic leukaemia and clinical trial results.Xingmei Mu Chumao Chen Loujie Dong Zhaowei Kang Zhixian Sun Xijie Chen Junke Zheng Yaping Zhang 2023Acta Biochimica et Biophysica Sinica2023,55,6:1
14An analytical poroelastic model for laboratorial mechanical testing of the articular cartilage (AC)显示文摘The articular cartilage(AC) can be seen as a biphasic poroelastic material.The cartilage deformation under compression mainly leads to an interstitial fluid flow in the porous solid phase.In this paper,an analytical poroelastic model for the AC under laboratorial mechanical testing is developed.The solutions of interstitial fluid pressure and velocity are obtained.The results show the following facts.(i) Both the pressure and fluid velocity amplitudes are proportional to the strain loading amplitude.(ii) Both the amplitudes of pore fluid pressure and velocity in the AC depend more on the loading amplitude than on the frequency.Thus,in order to obtain the considerable fluid stimulus for the AC cell responses,the most effective way is to increase the loading amplitude rather than the frequency.(iii) Both the interstitial fluid pressure and velocity are strongly affected by permeability variations.This model can be used in experimental tests of the parameters of AC or other poroelastic materials,and in research of mechanotransduction and injury mechanism involved interstitial fluid flow.Xiaogang WU Kuijun CHEN Zhaowei WANG Ningning WANG Teng ZHAO Yanan XUE Yanqin WANG Weiyi CHEN 2018Applied Mathematics and Mechanics(English Edition)2018,39,6:1

17Full-scale multi-functional test platform for investigating mechanical performance of track–subgrade systems of high-speed railways显示文摘Motivated by the huge practical engineering demand for the fundamental understanding of mechanical characteristics of high-speed railway infrastructure,a fullscale multi-functional test platform for high-speed railway track–subgrade system is developed in this paper,and its main functions for investigating the mechanical performance of track–subgrade systems are elaborated with three typical experimental examples.Comprising the full-scale subgrade structure and all the five types of track structures adopted in Chinese high-speed railways,namely the CRTS I,the CRTS II and the CRTS III ballastless tracks,the double-block ballastless track and the ballasted track,the test platform is established strictly according to the construction standard of Chinese high-speed railways.Three kinds of effective loading methods are employed,including the real bogie loading,multi-point loading and the impact loading.Various types of sensors are adopted in different components of the five types of track–subgrade systems to measure the displacement,acceleration,pressure,structural strain and deformation,etc.Utilizing this test platform,both dynamic characteristics and long-term performance evolution of high-speed railway track–subgrade systems can be investigated,being able to satisfy the actual demand for large-scale operation of Chinese high-speed railways.As examples,three typical experimental studies are presented to elucidate the comprehensive functionalities of the full-scale multi-functional test platform for exploring the dynamic performance and its long-term evolution of ballastless track systems and for studying the long-term accumulative settlement of the ballasted track–subgrade system in high-speed railways.Some interesting phenomena and meaningful results are captured by the developed test platform,which provide a useful guidance for the scientific operation and maintenance of high-speed railway infrastructure.Wanming Zhai Kaiyun Wang Zhaowei Chen Shengyang Zhu Chengbiao Cai Gang Liu 2020Railway Engineering Science2020,28,3:1

19Aqueous Synthesis of Covalent Organic Frameworks as Photocatalysts for Hydrogen Peroxide Production显示文摘Covalent organic frameworks(COFs)are crystalline porous polymers with designable structures and properties.Their crystallization typically relies on trialand-error experimentation involving harsh conditions,including organic solvents,presenting significant obstacles for rational design and large-scale production.Herein,we present a liquid crystal-directed synthesis methodology and its implementation for up to gram-scale production of highly crystalline COFs in water and air.It is compatible with monomers of different structures,shape,size,length of side chains,and electron-donating,electron-accepting,and heterocyclic substitutions near reactive sites.Seventeen types of donor-acceptor two-dimensional COFs including four types of new ones and a three-dimensional COF with a yield of up to 94%were demonstrated,showing great generality of the method.The as-synthesized donor-acceptor COFs are organic semiconductors and contain macropores besides intrinsic mesopores which make them attractive catalysts.The production of H_(2)O_(2)under visible light in water was studied and the structure-property relationships were revealed.The production rate reached 4347μmol h^(−1)gcat^(−1),which is about 467%better than that of the benchmark photocatalyst g-C_(3)N_(4).This study will inspire the mild synthesis and scale-up of a wide spectrum of COFs and organic semiconductors as efficient catalysts,promote their structure-property investigation,and boost their applications.Fanglin Tan Yuanyuan Zheng Zhipeng Zhou Honglei Wang Xin Dong Jing Yang Zhaowei Ou Haoyuan Qi Wei Liu Zhikun Zheng Xudong Chen 2022CCS Chemistry2022,4,12:1
