| 1 | Protective Roles of α-lipoic Acid in Rat Model of Mitochondrial DNA4834bp Deletion in Inner Ear显示文摘The protective roles of α-lipoic acid in the rat model of mitochondrial DNA (mtDNA) 4834bp deletion in inner ear were investigated. Forty female Wistar rats at 4 weeks of age were divided into four groups: group A (D-galactose group, n=10), group B (D-galactose+α-lipoic acid group, n=10), group C (α-lipoic acid group, n=10), and group D (control group, n=10). Auditory brainstem response (ABR) was used to detect the hearing threshold. Colorimetry was used to analyze activity of superoxide dismutase (SOD) and concentration of malondialdehyde (MDA). The percentage of mtDNA4834bp deletion in inner ear was identified by real-time PCR. There was no significant difference in ABR threshold shift among all groups. The percentage of mtDNA4834bp deletion in group A was higher than that in other groups, but there was no significant difference in percentage of mtDNA4834bp deletion among groups B, C, and D. The activity of SOD in group A was lower than that in other groups. The concentration of MDA in group A was higher than that in other groups. | 2 | Downregulation of Cav3.1 T-type Calcium Channel Expression in Age-related Hearing Loss Model显示文摘Objective Age-related hearing loss(AHL),characterized by degeneration of cochlea structures,is the most common sensory disorder among the elderly worldwide.The calcium channel is considered to contribute to normal hearing.However,the role of the T-type voltage-activated calcium channel,Cav3.1,remains unclear in AHL.Here,we investigate the age-related change of Cav3.1 expression in the cochlea and D-gal-induced senescent HEI-OC1 cells.Methods Cochleae from C57BL/6 mice at 2 months and 12 months of age were assessed.Senescence in House Ear Institute-Organ of Corti 1(HEI-OC1)cells was induced by D-gal treatment.The immunofluorescence technique was employed to investigate the distribution of Cav3.1 in vivo and in vitro.Quantitative assessment was achieved by Western blotting and real-time PCR.Results In comparison with 2-month-old animals,12-month old C57BL/6 mice exhibited great loss of hair cells and elevated auditory brainstem threshold.The Cav3.1 was located in hair cells,spiral ganglion cells,lateral walls,and the expression of Cav3.1 protein and mRNA decreased in the aged cochleae.D-gal-induced senescence assay confirmed the down-regulation of Cav3.1 expression in senescent HEI-OC1 cells.Conclusion Our results show that age-related down-regulated expression of Cav3.1 in the cochleae is associated with AHL and may contribute to the pathogenesis of AHL. | Chun-chen PAN Zhi-hui DU Yi ZHAO Han-qi CHU Jin-wu SUN | 2021 | Current Medical Science2021,41,4: | 0 |
| 3 | AUDITORY BRAINSTEM RESPONSES IN SENILE PRESBYCUSIS PATIENTS OVER 90 YEARS显示文摘Objective To analyze the characteristics of auditory brainstem response(ABR) in presbycusis patients elder than 90 years. Methods Fourteen presbycusis patients elder than 90 years(presbycusis group, 91.1.4±1.3 years, 26 ears) and 9 normal-hearing young adults(control group, 22.7±1.2 years, 18 ears) participated in the study. Alternative click-evoked ABRs were recorded in both groups. The peak latency(PL) of peak I,Ⅲ, and V, and the inter-peak latency(IPI) of I-Ⅲ, Ⅲ-V, and I-V were compared between groups. Results In elder presbycusis patients, the occurrence rate of peak I and Ⅲ were both 76.9%, and that of peak V was84.6%. In presbycusis group, the peak latencies of I, Ⅲ, V were significantly longer than that of control group(P<0.001). There was no significant difference between groups in the IPI of peak I-I Ⅲ(P=0.298,peak Ⅲ-V(P=0.254) and peak I-V(P=0.364). | 4 | Vestibular and cochlear dysfunction in aging:Two sides of the same coin?显示文摘Objective:Nonspecific complaints of hearing loss,vertigo,imbalance,and instability,without a defined etiology,are very prevalent in the elderly population,with a great impact on morbidity and mortality in this age group.The objectives of this study were to verify whether there is age-related vestibular dysfunction and to test the association of vestibular dysfunction with presbycusis in the elderly population.Methods:Original retrospective analytical cross-sectional study,carried out with 80 patients who underwent a videonystagmography and complete audiometric evaluation due to nonspecific vestibular complaints,without a specific vestibular disorder diagnosis.Patients were selected and divided into two distinct age groups(group A:>60 years;group B:18-50 years)and,in both groups,we analyzed the caloric tests and the pure-tone audiometry.Results:In the vestibular evaluation,we found that there was a statistically significant difference(P<0.05)between groups in the prevalence of bilateral vestibular weakness(group A:22.5%;group B:5%),and that the increase in age,above 60 years,is negatively correlated with the mean total caloric response.Additionally,we obtained a reasonable negative and statistically significant correlation(r=-0.320,P<0.05)between the mean bone conduction thresholds at high frequencies and total caloric responses in group A.Conclusions:In patients with hearing loss,it is essential to perform a complete vestibular study to diagnose vestibular disorders and,consequently,prevent adverse outcomes that may result from these alterations. | Catia Azevedo Sergio Vilarinho Ana Sousa Menezes Fernando Milhazes Mar Luis Dias | 2022 | World Journal of Otorhinolaryngology-Head and Neck Surgery2022,8,4: | 0 |
| 5 | Medical Therapy of Hearing Impairment and Tinnitus with Chinese Medicine:An Overview显示文摘The current review gives a comprehensive overview of the recent development in Chinese medicine(CM)for treating several kinds of acquired nerve deafness and tinnitus,as well as links the traditional principle to well-established pharmacological mechanisms for future research.To date,about 24 herbal species and 40 related ingredients used in CM to treat hearing loss and tinnitus are reported for the treatment of endocochlear potential,endolymph growth,lowering toxic and provocative substance aggregation,inhibiting sensory cell death,and retaining sensory transfer.However,there are a few herbal species that can be used for medicinal purposes.Nevertheless,clinical studies have been hampered by a limited population sample,a deficiency of a suitable control research group,or contradictory results.Enhanced cochlear blood flow,antiinflammatory antioxidant,neuroprotective effects,and anti-apoptotic,as well as multi-target approach on different auditory sections of the inner ear,are all possible benefits of CM medications.There are numerous unknown natural products for aural ailment and tinnitus identified in CM that are expected to be examined in the future utilizing various aural ailment models and processes. | ZHANG Ying XIE Hui HE Zhong-mei ZHANG Feng LI Ling-long WANG Na MAO De-hong | 2023 | Chinese Journal of Integrative Medicine2023,29,8: | 0 |
