| 1 | From contraction to gene expression:nanojunctions of the sarco/endoplasmic reticulum deliver site-and function-specific calcium signals显示文摘Calcium signals determine, for example, smooth muscle contraction and changes in gene expression. How calcium signals select for these processes is enigmatic. We build on the 'panjunctional sarcoplasmic reticulum' hypothesis, describing our view that different calcium pumps and release channels, with different kinetics and affinities for calcium, are strategically positioned within nanojunctions of the SR and help demarcate their respective cytoplasmic nanodomains. SERCA2 b and Ry R1 are preferentially targeted to the sarcoplasmic reticulum(SR) proximal to the plasma membrane(PM), i.e., to the superficial buffer barrier formed by PM-SR nanojunctions, and support vasodilation. In marked contrast, SERCA2 a may be entirely restricted to the deep, perinuclear SR and may supply calcium to this sub-compartment in support of vasoconstriction. Ry R3 is also preferentially targeted to the perinuclear SR, where its clusters associate with lysosome-SR nanojunctions. The distribution of Ry R2 is more widespread and extends from this region to the wider cell. Therefore, perinuclear Ry R3 s most likely support the initiation of global calcium waves at L-SR junctions, which subsequently propagate by calcium-induced calcium release via Ry R2 in order to elicit contraction. Data also suggest that unique SERCA and Ry R are preferentially targeted to invaginations of the nuclear membrane. | 2 | Quantitative trait loci associated with soybean seed weight and composition under different phosphorus levels显示文摘Seed size and composition are important traits in food crops and can be affected by nutrient availability in the soil. Phosphorus(P) is a non-renewable,essential macronutrient, and P deficiency limits soybean(Glycine max) yield and quality. To investigate the associations of seed traits in low-and high-P environments, soybean recombinant inbred lines(RILs) from a cross of cultivars Fiskeby III and Mandarin(Ottawa) were grown under contrasting P availability environments.Traits including individual seed weight, seed number, and intact mature pod weight were significantly affected by soil P levels and showed transgressive segregation among the RILs. Surprisingly, P treatments did not affect seed composition or weight, suggesting that soybeanmaintains sufficient P in seeds even in low-P soil.Quantitative trait loci(QTLs) were detected for seed weight, intact pods, seed volume, and seed protein,with five significant QTLs identified in low-P environments and one significant QTL found in the optimal-P environment. Broad-sense heritability estimates were0.78(individual seed weight), 0.90(seed protein), 0.34(seed oil), and 0.98(seed number). | 3 | Metallurgy at the Crossroads: New Analyses of Copper-based Objects at Tianshanbeilu, Eastern Xinjiang, China显示文摘Tianshanbeilu is the largest Bronze Age site in eastern Xinjiang, China. Stretching across the entire second millennium BC, it performed a prominent role in connecting the Hexi corridor, Central China and the steppe. A further insight into the metallurgical tradition and the metal supply network is of vital importance to improve our understanding of its multi-connected nature. This paper offers a new set of chemical and isotopic data on the copper-based objects at Tianshanbeilu, including alloying elements, trace elements(impurities) and lead isotopes. Combining the concentrations of arsenic and antimony reveals that arsenic was introduced to copper partially due to the use of specific minerals tethrahedrite-tennantite. Lead isotopes demonstrate that multiple sources of copper were employed at Tianshanbeilu and a majority of them are characterized by common lead, which appears rather different from those of the Central Plains and the Hexi corridor, but highly consistent with local ores. Surprisingly, one object at Tianshanbeilu contains the well-known highly radiogenic lead. This object undoubtedly marks the westernmost boundary of the distribution of the highly radiogenic lead. | 9 | Every Cloud has a Silver Lining: Using Silver Concentration to Identify the Number of Sources of Lead used in Shang Dynasty Bronzes显示文摘The use of lead, some of which is characterized by a highly radiogenic signature, sharply distinguishes Bronze Age China from the rest of Eurasia. Scholars have long hypothesized that silver can offer an independent proxy to characterize lead minerals. The summary of silver distribution associated with Shang and Western Zhou bronzes in this paper reveals an important difference between the south(Sanxingdui, Hanzhong, Jinsha, Panlongcheng, Xin’gan) and the Central Plains.
