1三峡工程对生态环境与生态系统的影响及政策分析模型研究显示文摘三峡工程作为一项世界举世瞩目的特大水利水电工程,一直备受公众的关注和争论,其利弊之争一直没有停止。其中,关于三峡工程对生态环境与生态系统的影响也是众说纷纭,但从总体上看,目前关于三峡工程对生态环境与生态系统影响的研究主要侧重于微观的、局部的、分学科的定量研究,而宏观的政府政策层面的研究比较少,尤其是关于三峡工程对生态环境和生态系统影响的政策分析模型研究更少,几乎没有。基于此,本文通过尽可能全面地收集、整理近些年来公开发表的权威资料和数据,在考虑经济、社会、环境和生态等多种因素影响下,通过分析和计算,运用环境影响经济损益的分析方法,从三峡水库对生态环境的影响因子中,筛选出起主导作用的几个因素,从经济价值的角度解释三峡工程对生态环境与生态系统的影响:即有利的影响主要是节能减排与防洪抗旱带来的经济效益,不利的影响主要是泥沙淤积、水污染和地质灾害。并通过进一步比较分析,结合权威专家的利弊观点梳理,得出三峡工程对生态环境与生态系统的总体影响是利大于弊的客观结论。在此基础上构建了改善三峡生态环境和生态系统影响政策分析体系模型,具体构建分析了五个方面的政策模型,并对政府政策进行了分析与建议,这五个方面的政策分析模型是:健全监督管理体制的政策分析模型;联动调度协调机制的科学管理的政策分析模型;采取'蓄清排浑'的政策分析模型;转变集中管理的水污染问题政策分析模型;建立多元主体防治与保护体系的地质灾害问题的政策分析模型。王树文 祁源莉 Farhed A.Shah 2015中国人口·资源与环境2015,25,5:9

3Fast and sensitive LC–MS/MS method for the simultaneous determination of lisinopril and hydrochlorothiazide in human plasma显示文摘A sensitive and rapid liquid chromatography-tandem mass spectrometry(LC–MS/MS) method has been developed for the simultaneous determination of lisinopril(LIS) and hydrochlorothiazide(HCTZ) in human plasma using their labeled internal standards(ISs). Sample pre-treatment involved solid phase extraction on Waters Oasis HLB cartridges using 100 μL of plasma, followed by liquid chromatography on Hypersil Gold C_(18)(50 mm×3.0 mm, 5 μm) column. The analytes were eluted within 2.0 min using acetonitrile-5.0 m M ammonium formate, p H 4.5(85:15, v/v) as the mobile phase. The analytes and ISs were analyzed in the negative ionization mode and quantified using multiple reaction monitoring. The method showed excellent linearity over the concentration range of 0.50–250.0 ng/m L for both the analytes. The intra-batch and inter-batch precision(% CV) was ≤5.26% and their extraction recoveries were in the range of 96.6%–103.1%. Matrix effect evaluated in terms of IS-normalized matrix factors ranged from 0.97 to 1.03 for both the analytes. 4The role of cofilin in age-related neuroinflammation显示文摘Aging brain becomes susceptible to neurodegenerative diseases due to the shifting of microglia and astrocyte phenotypes to an active“pro-inflammatory”state,causing chronic low-grade neuroinflammation.Despite the fact that the role of neuroinflammation during aging has been extensively studied in recent years,the underlying causes remain unclear.The identification of relevant proteins and understanding their potential roles in neuroinflammation can help explain their potential of becoming biomarkers in the aging brain and as drug targets for prevention and treatment.This will eventually reduce the chances of developing neurodegenerative diseases and promote healthier lives in the elderly.In this review,we have summarized the morphological and cellular changes in the aging brain,the effects of age-related neuroinflammation,and the potential role of cofilin-1 during neuroinflammation.We also discuss other factors contributing to brain aging and neuroinflammation.Amsha S.Alsegiani Zahoor A.Shah 2020Neural Regeneration Research2020,15,8:3
5Application of an LC–MS/MS method for the analysis of amlodipine,valsartan and hydrochlorothiazide in polypill for a bioequivalence study显示文摘A sensitive and selective method has been proposed for the simultaneous determination of amlodipine(AML),valsartan(VAL) and hydrochlorothiazide(HCTZ) in human plasma by liquid chromatography–tandem mass spectrometry(LC–MS/MS). The analytes and their deuterated analogs were quantitatively extracted from100 μL human plasma by solid phase extraction on Oasis HLB cartridges. The chromatographic separation of the analytes was achieved on a Chromolith RP18 e(100 mm × 4.6 mm) analytical column within 2.5 min. The resolution factor between AML and VAL, AML and HCTZ, and VAL and HCTZ was 2.9, 1.5 and 1.4, respectively,under isocratic conditions. The method was validated over a dynamic concentration range of 0.02–20.0 ng/m L for AML, 5.00–10,000 ng/m L for VAL and 0.20–200 ng/m L for HCTZ. Ion-suppression/enhancement effects were investigated by post-column infusion technique. The mean IS-normalized matrix factors for AML, VAL and HCTZ were 0.992, 0.994 and 0.998, respectively. The intra-batch and inter-batch precision(% CV) across quality control levels was ≤ 5.56% and the recovery was in the range of 93.4%–99.6% for all the analytes. The method was successfully applied to a bioequivalence study of 5 mg AML + 160 mg VAL + 12.5 mg HCTZ tablet formulation(test and reference) in 18 healthy Indian males under fasting. The mean log-transformed ratios of C max, AUC0–120 h and AUC0-inf and their 90% CIs were within 90.2%–102.1%. 850 years of scanning electron microscopy of bone——a comprehensive overview of the important discoveries made and insights gained into bone material properties in health,disease,and taphonomy显示文摘Bone is an architecturally complex system that constantly undergoes structural and functional optimisation through renewal and repair.The scanning electron microscope (SEM) is among the most frequently used instruments for examining bone.It offers the key advantage of very high spatial resolution coupled with a large depth of field and wide field of view.Interactions between incident electrons and atoms on the sample surface generate backscattered electrons,secondary electrons,and various other signals including X-rays that relay compositional and topographical information.Through selective removal or preservation of specific tissue components (organic,inorganic,cellular,vascular),their individual contribution(s) to the overall functional competence can be elucidated.With few restrictions on sample geometry and a variety of applicable sample-processing routes,a given sample may be conveniently adapted for multiple analytical methods.While a conventional SEM operates at high vacuum conditions that demand clean,dry,and electrically conductive samples,non-conductive materials (e.g.,bone) can be imaged without significant modification from the natural state using an environmental scanning electron microscope.This review highlights important insights gained into bone microstructure and pathophysiology,bone response to implanted biomaterials,elemental analysis,SEM in paleoarchaeology,3D imaging using focused ion beam techniques,correlative microscopy and in situ experiments.The capacity to image seamlessly across multiple length scales within the meso-micro-nano-continuum,the SEM lends itself to many unique and diverse applications,which attest to the versatility and user-friendly nature of this instrument for studying bone.Significant technological developments are anticipated for analysing bone using the SEM.Furqan A.Shah Krisztina Ruscsák Anders Palmquist 2019Bone Research2019,7,2:2
9Determination of lercanidipine in human plasma by an improved UPLC–MS/MS method for a bioequivalence study显示文摘An improved and reliable ultra-performance liquid chromatography/tandem mass spectrometry(UPLC–MS/MS) method has been developed and validated for the determination of lercanidipine in human plasma. Plasma samples with lercanidipine-d3 as an internal standard(IS) were prepared by solid phase extraction on Phenomenex Strata-X cartridges using 100 μL of human plasma. Chromatographic analysis was performed on UPLC BEH C_(18)(50 mm*2.1 mm, 1.7μm) column under isocratic conditions. Linear calibration curves were obtained over a wide dynamic concentration range of 0.010–20.0 ng/mL. Matrix effect was assessed by post-column infusion, post-extraction spiking and standard-line slope methods.The mean extraction recovery was >94% for the analyte and IS. Inter-batch and intra-batch precision(%CV) across five quality controls was <5.8%. 11Anthelmintic, antimicrobial, antioxidant and cytotoxic activity of Caltha palustris var. alba Kashmir, India显示文摘The methanolic extract obtained from the root portion of Caltha palustris var. alba was evaluated for its anthelmintic efficacy against gastrointestinal nematodes of sheep under both in vitro and in vivo conditions using worm motility inhibition(WMI) assay and fecal egg count reduction(FECR) assay, respectively. The extract was subjected to antimicrobial activity using agar-well diffusion method against different bacterial strains. In addition the extract was evaluated for cytotoxic and antioxidant activity against cultured THP-1(Leukemia), A-549(Lung), HCT-15(Colon), Cervix(HeLa) and PC-3(Prostrate) cell lines by SRB and DPPH radical scavenging assays. The extract used resulted in mean %WMI of 94.44%, as observed when the worms were put in lukewarm buffer for 30 min after exposure to different treatments. The mean mortality index of the sample was 0.95. The lethal concentration(LC50) was 0.11 mg·mL-1. Cell lines were exposed to concentration of 100 μg·mL-1 of extract for 48 h, which reduced the viability of these cell lines. Finding general and verifiable conditions which imply that the wavelet systems are wavelet frames is among the core problems in time-frequency analysis. In this article, we establish some new inequalities for wavelet frames on local fields of positive characteristic by means of the Fourier transform. 14Invasion by Conyza sumatrensis alters soil microbial community structure in urban ecosystems显示文摘Introduction:Whether invasive plants stimulate or inhibit the soil microbial diversity is still an open question.Despite large-scale invasion by Conyza sumatrensis(Retz.)E.Walker in the urban ecosystems of the Srinagar city of the Kashmir Himalayan region,limited information exists on its impact,particularly,on the belowground microbial diversity.The present study was thus conducted to compare the soil microbial(bacterial and ascomycetous fungal)diversity between the sites invaded by C.sumatrensis and un-invaded(control)sites.Methods:Soil metagenome was extracted from C.sumatrensis invaded and un-invaded plots at the three study sites.A total of six plots(5×5 m each in size),including three invaded by C.sumatrensis and three un-invaded plots were nested within each study site.DNA after amplification was subject to denaturing gradient gel electrophoresis(DGGE);the bands were extracted from the DGGE gel,re-amplified,and sequenced for identification of the species.Results:The number of bacterial species was reduced in the invaded plots at two out of the three sites while as it was relatively higher in the un-invaded plots with many species exclusively found in these plots.Fungal species richness was higher in the invaded plots compared to the un-invaded plots at all the three sites.Also,more fungal species were found to occur exclusively in the invaded plots without being represented in the un-invaded plots.Conclusions:Invasion by C.sumatrensis alters soil microbial community structure in the urban ecosystems in the Kashmir Himalaya.How this species does so and what benefits does it draw from such alteration promise to be an interesting future discourse.Nazima Rasool Zafar A.Reshi Damase P.Khasa Mudasir Roshan Manzoor A.Shah 2016Ecological Processes2016,5,1:1
15Predicting plant invasiveness from native range size: clues from the Kashmir Himalaya显示文摘Aims In view of the growing interest in modelling the potential spread of invasive species,prediction of plant invasiveness on the basis of native range size holds considerable promise.Our objective was to use a simple model to evaluate whether a wider native range predisposes plant species to become invasive in non-native regions and to easily identify potential invaders on this basis.The Kashmir Himalayan alien flora,of which a large proportion is native to Europe,was used to test this model.Methods The Kashmir Himalayan alien flora comprises 436 species of vascular plants at different stages of invasion.We focussed on plant species at two critical invasion stages(sensu Colautti and MacIsaac 2004),i.e.Stage II(species that are just at the earliest phase of introduction)and Stage V(species that are widespread and dominant in the invaded region and are thus considered invasive).We used the territorial distribution in Europe(number of countries)as a surrogate for the native range size of plants of European origin.Important Findings Using a subset of 88 species,for which information on the native European range was available,we showed that a large proportion(68%)of Stage II species growing in the Kashmir Valley had a relatively restricted European range(present in<20 countries);on the other hand,77%of Stage V species had an extensive native range(present in>20 countries).We consequently hypothesized that 14 Kashmir Himalayan Stage II species of European origin that are distributed in>20 European countries are at risk of becoming future invaders in Kashmir.On the other hand,those Kashmir Himalayan Stage II species of European origin distributed in<20 European countries are less likely to become invasive.Although this analysis is quite simple,the data suggest that a wider native range is a good predictor of plant invasiveness and could be used as a simple and low-cost early warning tool in predicting potential invasive species.Manzoor A.Shah Zafar A.Reshi Claude Lavoie 2012Journal of Plant Ecology2012,5,2:1

18Simultaneous quantification of amiloride and hydrochlorothiazide in human plasma by liquid chromatography–tandem mass spectrometry显示文摘A selective, sensitive and precise assay based on solid phase extraction and liquid chromatography–tandem mass spectrometry(LC–MS/MS) was developed for the simultaneous determination of amiloride(AMI) and hydrochlorothiazide(HCTZ) in human plasma. Sample clean-up with 250 μL of plasma was done on Phenomenex Strata^(?)-X extraction cartridges using their labeled internal standards(AMI-15 N3 and HCTZ-13 C,d2). Chromatography was performed on Hypersil Gold C18(50 mm×3.0 mm, 5 μm) column using acetonitrile with 4.0 m M ammonium formate(p H 4.0, adjusted with 0.1% formic acid)(80:20, v/v) as the mobile phase. Detection was carried out on a triple quadrupole API 5500 mass spectrometer utilizing an electrospray ionization interface and operating in the positive ionization mode for AMI and negative ionization mode for HCTZ. Multiple reaction monitoring was used following the transitions at m/z 230.6/116.0, m/z233.6/116.0, m/z 296.0/204.9 and m/z 299.0/205.9 for AMI, AMI-15 N3, HCTZ and HCTZ-13 C,d2,respectively. Calibration curves were linear(r2≥0.9997) over the concentration range of 0.050–50.0 and0.50–500 ng/m L for AMI and HCTZ, respectively, with acceptable accuracy and precision. The signal-to-noise ratio at the limit of quantitation was ≥14 for both the analytes. The mean recovery of AMI and HCTZ from plasma was 89.0% and 98.7%, respectively. The IS-normalized matrix factors determined for matrix effect ranged from 0.971 to 1.024 for both the analytes.
