2Single-cell transcriptomes and runx2b^(-/-)mutants reveal the genetic signatures of intermuscular bone formation in zebrafish显示文摘Intermuscular bones(IBs)are mineralized spicules,present in the myosepta of many,but not all,teleost species.IBs are often small and sharp,and they consequently limit how the fish can be processed;the IBs may cause injury or trauma if lodged in consumers’throats or mouths,and therefore affect the appeal of the fish to many consumers.The development of IBs in teleosts is still not fully understood and the molecular basis of IB development remains to be established.Here,the characteristics of IB tissue are evaluated based on single-cell transcriptomics in wild-type zebrafish.The analysis defined 18 distinct cell types.Differentiation trajectories showed that IBs are derived from tendons and that a core tendon-osteoblast cell lineage is related to IB formation.In particular,the functions of 10 candidate genes were evaluated via CRISPR-Cas9 mutants.Among those,runx2b^(-/-)mutants completely lost IBs,while swimming performance,growth and bone mineral density were not significantly different from runx2b^(+/+)zebrafish.Comparative single-cell RNA sequencing(sc RNA-seq)analysis in runx2b^(-/-)and runx2b^(+/+)zebrafish revealed the role of osteoblasts in IB formation.In addition,differentially expressed genes were enriched in the transforming growth factorβ/bone morphogenetic protein(TGF-β/BMP)pathway after runx2b deletion.This study provides evidence for the crucial role of runx2b regulation in IB formation.Genetic breeding can target runx2b regulation and generate strains of commercial fish species without IBs,which can improve the safe consumption and economic value of many farmed fish species.Chun-Hong Nie Shi-Ming Wan Yu-Long Chen Ann Huysseune Ya-Ming Wu Jia-Jia Zhou Alexandre Wagner Silva Hilsdorf Wei-Min Wang Paul Eckhard Witten Qiang Lin Ze-Xia Gao 2022National Science Review2022,9,11:6
3(La0.8A0.2)MnO3(A=Sr,K) perovskite catalysts for NO and C10H22 oxidation and selective reduction of NO by C(10)H(22)显示文摘In this work, we studied the catalytic activity of LaMnO3 and(La0.8A0.2)MnO3(A = Sr, K) perovskite catalysts for oxidation of NO and C10H22 and selective reduction of NO by C10H22. The catalytic per‐formances of these perovskites were compared with that of a 2 wt% Pt/SiO2 catalyst. The La site substitution increased the catalytic properties for NO or C10H22 oxidation compared with the non‐substituted LaMnO3 sample. For the most efficient perovskite catalyst,(La0.8Sr0.2)MnO3, the results showed the presence of two temperature domains for NO adsorption:(1) a domain corre‐sponding to weakly adsorbed NO, desorbing at temperatures lower than 270 °C and(2) a second domain corresponding to NO adsorbed on the surface as nitrate species, desorbing at temperatures higher than 330 °C. For the Sr‐substituted perovskite, the maximum NO2 yield of 80% was observed in the intermediate temperature domain (around 285 °C). In the reactant mixture of NO/C10H22/O2/H2O/He,(La0.8Sr0.2)MnO3 perovskite showed better performance than the 2 wt% Pt/SiO2 catalyst: NO2 yields reaching 50% and 36% at 290 and 370 °C, respectively. This activity improvement was found to be because of atomic scale interactions between the A and B active sites, Sr2+ cation and Mn4+/Mn3+ redox couple. 4Effect of micro-algae Schizochytrium sp.supplementation in plant diet on reproduction of female rainbow trout(Oncorhynchus mykiss):maternal programming impact of progeny显示文摘Background:The broodstock diet,and in particular the lipid and fatty acid composition of the diet,is known to play a key role in reproductive efficiency and survival of the progeny in fish.A major problem when replacing both fish meal and fish oil by plant sources is the lack of n-3 long chain polyunsaturated fatty acids,such as eicosapentaenoic acid(EPA)and docosahexaenoic acid(DHA).To address this problem,we studied the effect of the plant-based diet supplemented with Schizochytrium sp.microalgae,source of DHA,compared to a conventional commercial diet rich in fish meal and fish oil on reproductive performance and egg quality and the consequences on progeny,in female rainbow trout broodstock.Results:The results demonstrated that DHA-rich microalgae supplementation in a plant-based diet allowed for the maintenance of reproductive performance and egg quality comparable to a conventional commercial feed rich in fish meal and fish oil and led to an increased significant fry survival after resorption.Moreover,when females were fed a plant-based diet supplemented with micro-algae,the 4-month-old progenies showed a significant higher growth when they were challenged with a similar diet as broodstock during 1 month.We provide evidence for metabolic programming in which the maternal dietary induced significant protracted effects on lipid metabolism of progeny.Conclusions:The present study demonstrates that supplementation of a plant-based diet with DHA-rich microalgae can be an effective alternative to fish meal and fish oil in rainbow trout broodstock aquafeed.Emilie Cardona Emilien Segret Yoann Cachelou Thibaut Vanderesse Laurence Larroquet Alexandre Hermann Anne Surget Geneviève Corraze Frederic Cachelou Julien Bobe Sandrine Skiba-Cassy 2022Journal of Animal Science and Biotechnology2022,13,4:1
