| 2 | MicroRNA-548 down-regulates host antiviral response via direct targeting of IFN-λ1显示文摘Interferon(IFN)-mediated pathways are a crucial part of the cellular response against viral infection.Type III IFNs,which include IFN-λ1,2 and 3,mediate antiviral responses similar to Type I IFNs via a distinct receptor complex.IFN-λ1 is more effective than the other two members.Transcription of IFN-λ1 requires activation of IRF3/7 and nuclear factor-kappa B(NF-κB),similar to the transcriptional mechanism of Type I IFNs.Using reporter assays,we discovered that viral infection in-duced both IFN-λ1 promoter activity and that of the 3′-untranslated region(UTR),indicating that IFN-λ1 expression is also regulated at the post-transcriptional level.After analysis with microRNA(miRNA)prediction programs and 3′UTR targeting site assays,the miR-NA-548 family,including miR-548b-5p,miR-548c-5p,miR-548i,miR-548j,and miR-548n,was identified to target the 3′UTR of IFN-λ1.Further study demonstrated that miRNA-548 mimics down-regulated the expression of IFN-λ1.In contrast,their inhibitors,the complemen-tary RNAs,enhanced the expression of IFN-λ1 and IFN-stimulated genes.Furthermore,miRNA-548 mimics promoted infection by enterovirus-71(EV71)and ve-sicular stomatitis virus(VSV),whereas their inhibitors significantly suppressed the replication of EV71 and VSV.Endogenous miRNA-548 levels were suppressed during viral infection.In conclusion,our results sug-gest that miRNA-548 regulates host antiviral response via direct targeting of IFN-λ1,which may offer a poten-tial candidate for antiviral therapy. | Yongkui Li Jiajia Xie Xiupeng Xu Jun Wang Fang Ao Yushun Wan Ying Zhu | 2013 | Protein & Cell2013,4,2: | 22 |
| 3 | Human umbilical cord blood stem cell transplantation for the treatment of chronic spinal cord injury Electrophysiological changes and long-term efficacy显示文摘Stem cell transplantation can promote functional restoration following acute spinal cord injury (injury time < 3 months), but the safety and long-term efficacy of this treatment need further exploration. In this study, 25 patients with traumatic spinal cord injury (injury time > 6 months) were treated with human umbilical cord blood stem cells via intravenous and intrathecal injection. The follow-up period was 12 months after transplantation. Results found that autonomic nerve functions were restored and the latent period of somatosensory evoked potentials was reduced. There were no severe adverse reactions in patients following stem cell transplantation. | 4 | Water regime-nitrogen fertilizer incorporation interaction:Field study on methane and nitrous oxide emissions from a rice agroecosystem in Harbin, China显示文摘Water regime and nitrogen(N) fertilizer are two important factors impacting greenhouse gases(GHG) emission from paddy field, whereas their effects have not been well studied in cold region. In this study, we conducted a two-year field experiment to study the impacts of water regime and N fertilizer on rice yields and GHG emissions in Harbin, China, a cold region located in high latitudes. Our results showed that intermittent irrigation significantly decreased methane(CH_4) emission compared with continuous flooding, however,the decrement was far lower than the global average level. The N_2O emissions were very small when flooded but peaked at the beginning of the disappearance of floodwater. The N fertilizer treatments increased CH_4 emissions at low level(75 kg N/ha). But both CH_4 and N_2O emissions were uninfluenced at the levels of 150 kg N/ha and 225 kg N/ha. Rice yields increased under intermittent irrigation and were highest at the level of 150 kg N/ha. | 5 | Invasion biology of the cotton mealybug, Phenacoccus solenopsis Tinsley: Current knowledge and future directions显示文摘The cotton mealybug Phenacoccus solenopsis Tinsley(Hemiptera: Pseudococcidae), which is native to North America, emerged as a major invasive insect pest of multiple crops in Asia at the beginning of the 21 st century. Considering the economic significance of this insect and its rapid worldwide spread, we examined the possible factors driving its invasions. In this paper, we summarize the life history traits of P. solenopsis conceivably related to population development in invaded regions; these traits include its use of diverse host plants, reproductive capacity and mode, adaptation to temperature, response to food shortage, and insecticidal resistance. Then, focusing on the multiple trophic interactions that may promote or hinder invasion, we review the mutualistic relationship of this mealybug with ants and predation and parasitism by natural enemies. Finally, we suggest topics for future research and provide our perspective on the biological invasions of this mealybug. | 6 | Cytocompatibility with osteogenic cells and enhanced in vivo anti-infection potential of quaternized chitosan-loaded titania nanotubes显示文摘Infection is one of the major causes of failure of orthopedic implants. Our previous study demonstrated that nanotube modification of the implant surface, together with nanotubes loaded with quaternized chitosan(hydroxypropyltrimethyl ammonium chloride chitosan, HACC), could effectively inhibit bacterial adherence and biofilm formation in vitro. Therefore, the aim of this study was to further investigate the in vitro cytocompatibility with osteogenic cells and the in vivo anti-infection activity of titanium implants with HACC-loaded nanotubes(NT-H). The titanium implant(Ti), nanotubes without polymer loading(NT), and nanotubes loaded with chitosan(NT-C) were fabricated and served as controls. Firstly, we evaluated the cytocompatibility of these specimens with human bone marrow-derived mesenchymal stem cells in vitro.The observation of cell attachment, proliferation, spreading, and viability in vitro showed that NT-H has improved osteogenic activity compared with Ti and NT-C. A prophylaxis rat model with implantation in the femoral medullary cavity and inoculation with methicillin-resistant Staphylococcus aureus was established and evaluated by radiographical, microbiological, and histopathological assessments. Our in vivo study demonstrated that NT-H coatings exhibited significant anti-infection capability compared with the Ti and NT-C groups. | 11 | An Inverse Power Generation Mechanism Based Fruit Fly Algorithm for Function Optimization显示文摘As a novel population-based optimization algorithm, fruit fly optimization(FFO) algorithm is inspired by the foraging behavior of fruit flies and possesses the advantages of simple search operations and easy implementation. Just like most population-based evolutionary algorithms, the basic FFO also suffers from being trapped in local optima for function optimization due to premature convergence.In this paper, an improved FFO, named IPGS-FFO, is proposed in which two novel strategies are incorporated into the conventional FFO. Specifically, a smell sensitivity parameter together with an inverse power generation mechanism(IPGS) is introduced to enhance local exploitation. Moreover,a dynamic shrinking search radius strategy is incorporated so as to enhance the global exploration over search space by adaptively adjusting the searching area in the problem domain. The statistical performance of FFO, the proposed IPGS-FFO, three state-of-the-art FFO variants, and six metaheuristics are tested on twenty-six well-known unimodal and multimodal benchmark functions with dimension 30, respectively. | 12 | Celastrol suppresses colorectal cancer via covalent targeting peroxiredoxin 1显示文摘As a terpenoids natural product isolated from the plant Thunder God Vine,Celastrol is widely studied for its pharmacological activities,including anti-tumor activities.The clinical application of Celastrol is strictly limited due to its severe side effects,whereas previously revealed targets and mechanism of Celastrol seldom reduce its in vivo toxicity via structural optimization.Target identification has a far-reaching influence on the development of innovative drugs,and omics data has been widely used for unbiased target prediction.However,it is difficult to enrich target of specific phenotype from thousands of genes or proteins,especially for natural products with broad promising activities.Here,we developed a text-mining-based web-server tool to enrich targets from omics data of inquired compounds.Then peroxiredoxin 1(PRDX1)was identified as the ROS-manipulating target protein of Celastrol in colorectal cancer.Our solved high-resolution crystal structure revealed the unique covalent binding mode of Celastrol with PRDX1.New derivative compound 19-048 with improved potency against PRDX1 and selectivity towards PRDX2~PRDX6 were synthesized based on crystal structure analysis.Both Celastrol and 19-048 effectively suppressed the proliferation of colorectal cancer cells.The anti-tumor efficacy of Celastrol and 19-048 was significantly diminished on xenograft nude mice bearing PRDX1 knock-down colorectal cancer cells.Several downstream genes of p53 signaling pathway were dramatically up-regulated with Celastrol or 19-048 treatment.Our findings reveal that the side effects of Celastrol could be reduced via structural modification,and PRDX1 inhibition is promising for the treatment of colorectal cancer. | Heng Xu Hongfang Zhao Chunyong Ding Defang Jiang Zijie Zhao Yang Li Xiaoyu Ding Jing Gao Hu Zhou Cheng Luo Guoqiang Chen Ao Zhang Ying Xu Hao Zhang | 2023 | Signal Transduction and Targeted Therapy2023,8,3: | 3 |
| 13 | Effects of infusing milk precursors into the artery on rumen fermentation in lactating cows显示文摘This experiment was conducted to investigate the effects of infusing milk precursors into the external pudic artery on rumen fermentation in lactating dairy cows. Eight multiparous Holstein cows were randomly assigned to Group A(experimental group) and Group B(control group) with 4 cows each. A2×4 complex factor crossover design was used. Cows in Group A were fed corn straw as the only roughage, and cows in Group B were fed mixed roughage. The experiment was divided into two periods.In the first period, cows in Group A, received treatments: 1) a basal infusate as a control(CSC); 2) a milk fat precursor infusion including C16:0, C18:0, C18:1 c9, C18:2 c6, C18:3 n3, acetic acid(CSF); 3) a milk protein precursor infusion including 16 amino acids(CSA); 4) the mixed infusion of milk fat and protein precursors(CSFA). And meanwhile, cows in Group B were infused the basal infusate as a control group. In the second period, the cows in both Groups A and B were crossed over, which cows in Group A were named as Group B and the cows originally in Group B were in Group A. The experimental results showed that cows in experimental group had higher ruminal pH compared with the control, and ruminal pH in CSC, CSF, CSA were significantly higher than those in their respective control group(P < 0.05). The concentration of ammonia nitrogen(NH_3-N) was significantly higher in CSA and CSFA compared with Group B(P < 0.05). We also observed that the infusion of mixed amino acids significantly increased the bacterial protein(BCP) content in rumen(P < 0.05) and influenced the rumen acetic acid concentration as well as the acetic to propionic ratio(P < 0.05). Milk fat precursors infusion significantly affected butyric acid concentration(P < 0.05). In addition, the content of lipopolysaccharide(LPS) in CSA was significantly higher than that in the control group(P < 0.05). It is concluded that the milk precursors infused into external pudic artery caused feedback effects on ruminal fermentation under the corn straw roughage conditions. | 14 | The clinical relevance of MBL2 gene polymorphism and sepsis显示文摘Objective:To detect the clinical relevance of mannose-binding lectin 2(MBL2) gene polymorphism and sepsis in Chinese lived in Hainan island.Methods: Blood samples from 57 patients with sepsis and 69 patients without sepsis were collected in the ICU of several large hospitals in Hainan province. Genomic DNA was extracted from whole blood and then PCR purification product was sequenced and typed by 3730 sequencing analyzer. The concentration of MBIL2 in serum was detected by ELISA.Results: We found that genotype and allele distributions in two groups were in accordance with the Hardy-Weinberg Equilibrium. The frequency of GA genotype was significantly higher than that in non-sepsis group(P=0.013). A allele frequency in sepsis group was also much higher than that in non-sepsis group(P=0.028).Logister regression analysis showed that the patients who carried A allele were more prone to get sepsis than G allele carrier(P=0.014, OR=2,550, 95%CI=1.207-5.386). The MBL2 level in serum of sepsis patients with genotype GG and GA was significantly lower than that in non-sepsis group(P<0.05). Variations in microstructures for ascast SDSS2607 during heat preservation at 1 220 ℃ indicated two kinds of transformations from γ phase to δ phase.In-situ observations of microstructural changes during the tensile process at 1 050 ℃ showed a mutual coordination betw een γ and δ phases. When the true strain increased,the mutual coordination between γ and δ phases was damaged. Subsequently,cracks nucleated at the γ/δ interface. With the increase in temperature,the strength of ascast SDSS2607 decreased while its plasticity increased. Its thermoplasticity was poor,and the reduction in area of tensile specimens was less than 80%. When the deformation strain of hot compression increased,the stable deformation zone in the heat processing maps enlarged gradually. | 17 | Microstructure evolution of A380 aluminum alloy during rheological process under applied pressure显示文摘In squeeze casting process,the essence of mold-filling and feeding is rheological solidification of the alloy melt under pressure.Microstructure evolution is inevitable in this process,which affects the mold-filling and feeding in turn.In this work,the Archimedes spiral sample prepared by indirect squeeze casting was applied to investigate the microstructure evolution during the rheological process under pressure.The results showed that the primaryα-Al phase was transformed from fine rosette-like or granular structure to coarse platelet-like structure with the increase of spiral length.However,the primaryα-Al grain size of the starting point had a slight growing trend compared with that at the position of 140 mm away from the starting point.The volume fraction of the primaryα-Al phase increased from 45.57%to 70.35%along the spiral length direction,demonstrating that the experimental pressure improved the rheological ability of the alloy melt to some extent.Furthermore,the eutectic Si phase was varied from a fine granular or wormlike structure to a coarse platelet-like or needle-like structure,and the dispersion of eutectic Si particles was also varied along the spiral length direction.This microstructure evolution was mainly owing to the comprehensive action of rheological solidification and pressurized solidification.More specifically,the microstructure evolution strongly depended on the pressure and rheological velocity during the rheological process,however,the effect of rheological distance was relatively small. | Ying Wang Shu-ming Xing Xiao-hui Ao Ting-yue Wang | 2019 | China Foundry2019,16,6: | 2 |
| 18 | Exosomes secreted from cardiomyocytes suppress the sensitivity of tumor ferroptosis in ischemic heart failure显示文摘Heart failure(HF)patients in general have a higher risk of developing cancer.Several animal studies have indicated that cardiac remodeling and HF remarkably accelerate tumor progression,highlighting a cause-and-effect relationship between these two disease entities.Targeting ferroptosis,a prevailing form of non-apoptotic cell death,has been considered a promising therapeutic strategy for human cancers.Exosomes critically contribute to proximal and distant organ-organ communications and play crucial roles in regulating diseases in a paracrine manner.However,whether exosomes control the sensitivity of cancer to ferroptosis via regulating the cardiomyocyte-tumor cell crosstalk in ischemic HF has not yet been explored.Here,we demonstrate that myocardial infarction(MI)decreased the sensitivity of cancer cells to the canonical ferroptosis activator erastin or imidazole ketone erastin in a mouse model of xenograft tumor.Post-MI plasma exosomes potently blunted the sensitivity of tumor cells to ferroptosis inducers both in vitro in mouse Lewis lung carcinoma cell line LLC and osteosarcoma cell line K7M2 and in vivo with xenograft tumorigenesis model.The expression of miR-22-3p in cardiomyocytes and plasma-exosomes was significantly upregulated in the failing hearts of mice with chronic MI and of HF patients as well.Incubation of tumor cells with the exosomes isolated from post-MI mouse plasma or overexpression of miR-22-3p alone abrogated erastin-induced ferroptotic cell death in vitro.Cardiomyocyte-enriched miR-22-3p was packaged in exosomes and transferred into tumor cells.Inhibition of cardiomyocyte-specific miR-22-3p by AAV9 sponge increased the sensitivity of cancer cells to ferroptosis.ACSL4,a pro-ferroptotic gene,was experimentally established as a target of miR-22-3p in tumor cells.Taken together,our findings uncovered for the first time that MI suppresses erastin-induced ferroptosis through releasing miR-22-3p-enriched exosomes derived from cardiomyocytes.Therefore,targeting exosome-mediated cardiomyocyte/tumor pathological communication may offer a novel approach for the ferroptosis-based antitumor therapy. | Ye Yuan Zhongting Mei Zhezhe Qu Guanghui Li Shuting Yu Yingqi Liu Kuiwu Liu Zhihua Shen Jiaying Pu Yanquan Wang Changhao Wang Zhiyong Sun Qian Liu Xiaochen Pang Ao Wang Zijing Ren Tong Wang Ying Liu Jinhuan Hong Jiajie Xie Xin Li Zhonghua Wang Weijie Du Baofeng Yang | 2023 | Signal Transduction and Targeted Therapy2023,8,4: | 2 |

| 20 | NANOG Alleviates the Damage of Human Hair Follicle Mesenchymal Stem Cells Caused by H_2O_2 through Activation of AKT Pathway显示文摘Objective To explore the protective effect of NANOG against hydrogen peroxide(H_2O_2)-induced cell damage in the human hair follicle mesenchymal stem cells(hHF-MSCs). Methods NANOG was expressed from a lentiviral vector, pLVX-IRES-ZsGreen. NANOG hHF-MSCs and vector hHF-MSCs were treated with 400 μmol/L hydrogen peroxide(H_2O_2) for 2 h, the cell survival rate, cell morphology, ROS production, apoptosis and expression of AKT, ERK, and p21 were determined and compared. Results Our results showed that NANOG could activate AKT and upregulate the expression of p-AKT, but not p-ERK. When treated with 400 μmol/L H_2O_2, NANOG hHF-MSCs showed higher cell survival rate, lower ROS production and apoptosis, higher expression of p-AKT, higher ratio of p-AKT/AKT.
