8Do astrocytes respond to light,sound,or electrical stimulation?显示文摘Astrocytes are not only the most populous cell type in the human brain,but they also have the most extensive and dive rse sets of connections,across synapses,axons,blood vessels,as well as having their own internal network.Unsurprisingly,they are associated with many brain functions;from the synaptic transmission to energy metabolism and fluid homeostasis,and from cerebral blood flow and blood-brain barrier maintenance to neuroprotection,memory,immune defenses and detoxification,slee p,and early development.And yet,notwithstanding these key roles,so many current therapeutic approaches to a range of brain disorders have largely neglected their potential involvement.In this review,we consider the role of astrocytes in three brain therapies;two are emerging treatments(photobiomodulation and ultrasound),while the other is well-established(deep brain stimulation).In essence,we explore the issue of whether external sources,such as light,sound,or electricity,can influence the function of astrocytes,as they do neurons.We find that,when taken all together,each of these external sources can influence many,if not,all of the functions associated with astrocytes.These include influencing neuronal activity,prompting neuroprotection,reducing inflammation(astrogliosis) and potentially increasing cerebral blood flow and stimulating the glymphatic system.We suggest that astrocytes,just like neurons,can respond positively to each of these external applications and that their activation co uld each impart many beneficial outcomes on brain function;they are likely to be key playe rs underpinning the mechanisms behind many therapeutic strategies.Audrey Le Naour Emilien Beziat Jaimie Hoh Kam Pierre Magistretti Alim-Louis Benabid John Mitrofanis 2023Neural Regeneration Research2023,18,11:0
9Added value of molecular karyotype in childhood acute lymphoblastic leukemia显示文摘Background:Thanks to an improved therapeutic regimen in childhood B-cell precursor acute lymphoblastic leukemia(BCP-ALL),5 year-overall survival now exceeds 90%.Unfortunately,the 25%of children who relapse have an initial poor prognosis,potentially driven by pre-existing or emerging molecular anomalies.The latter are initially and essentially identified by cytogenetics.However,some subtle alterations are not visible through karyotyping.Methods:Single nucleotide polymorphisms(SNP)array is an alternative way of chromosomal analysis allowing for a more in-depth evaluation of chromosomal modifications such as the assessment of copy number alterations(CNA)and loss of heterozygosity(LOH).This method was applied here in retrospective diagnosis/relapse paired samples from seven children with BCP-ALL and in a prospective cohort of 38 newly diagnosed childhood cases.Results:In the matched study,compared to the initial karyotype,SNP array analysis reclassified two patients as poor prognosis cases.Modulation during relapse was seen for 4 CNA and 0.9 LOH.In the prospective study,SNP reclassified the 10 patients with intermediate karyotype as 7 good prognosis and 3 poor prognosis.Ultimately,in all the children tested,SNP array allowed to identify additional anomalies compared to conventional karyotype,refine its prognostic value and identify some druggable anomalies that could be used for precision medicine.Overall,the anomalies detected could be segregated in four groups respectively involved in B-cell development,cell proliferation,transcription and molecular pathways.Conclusion:SNP therefore appears to be a method of choice in the integrated diagnosis of BCP ALL,especially for patients initially classified as intermediate prognosis.This complementary method of both cytogenetics and high throughput sequencing allows to obtain further classified information and can be useful in case of failure of these techniques.Margaux Camuset Baptiste Le Calvez Olivier Theisen Catherine Godon Audrey Grain Caroline Thomas Marie-Laure Couec Marie C.Béné Fanny Rialland Marion Eveillard 2023Cancer Innovation2023,2,6:0
10Endothelin increases the proliferation of rat olfactory mucosa cells显示文摘The olfactory mucosa holds olfactory sensory neurons directly in contact with an aggressive environment. In order to maintain its integrity, it is one of the few neural zones which are continuously renewed during the whole animal life. Among several factors regulating this renewal, endothelin acts as an anti-apoptotic factor in the rat olfactory epithelium. In the present study, we explored whether endothelin could also act as a proliferative factor. Using primary culture of the olfactory mucosa, we found that an early treatment with endothelin increased its growth. Consistently, a treatment with a mixture of BQ123 and BQ788(endothelin receptor antagonists) decreased the primary culture growth without affecting the cellular death level. We then used combined approaches of calcium imaging, reverse transcriptase-quantitative polymerase chain reaction and protein level measurements to show that endothelin was locally synthetized by the primary culture until it reached confluency. Furthermore, in vivo intranasal instillation of endothelin receptor antagonists led to a decrease of olfactory mucosa cell expressing proliferating cell nuclear antigen(PCNA), a marker of proliferation. Only short-term treatment reduced the PCNA level in the olfactory mucosa cells. When the treatment was prolonged, the PCNA level was not statistically affected but the expression level of endothelin was increased. Overall, our results show that endothelin plays a proliferative role in the olfactory mucosa and that its level is dynamically regulated. 11High-elevation hypoxia impacts perinatal physiology and performance in a potential montane colonizer显示文摘Climate change is generating range shifts in many organisms,notably along the elevational gradient in mountainous environments.However,moving up in elevation exposes organisms to lower oxygen availability,which may reduce the successful reproduction and development of oviparous organisms.To test this possibility in an upwardcolonizing species,we artificially incubated developing embryos of the viperine snake(Natrix maura)using a split-clutch design,in conditions of extreme high elevation(hypoxia at 2877 m above sea level;72%sea-level equivalent O2 availability)or low elevation(control group;i.e.normoxia at 436 m above sea level).Hatching success did not differ between the two treatments.Embryos developing at extreme high elevation had higher heart rates and hatched earlier,resulting in hatchlings that were smaller in body size and slower swimmers compared to their siblings incubated at lower elevation.Furthermore,post-hatching reciprocal transplant of juveniles showed that snakes which developed at extreme high elevation,when transferred back to low elevation,did not recover full performance compared to their siblings from the low elevation incubation treatment.These results suggest that incubation at extreme high elevation,including the effects of hypoxia,will not prevent oviparous ectotherms from producing viable young,but may pose significant physiological challenges on developing offspring in ovo.These early-life performance limitations imposed by extreme high elevation could have negative consequences on adult phenotypes,including on fitness-related traits.Jérémie SOUCHET Eric J.GANGLOFF Gaëlle MICHELI Coralie BOSSU Audrey TROCHET Romain BERTRAND Jean CLOBERT Olivier CALVEZ Albert MARTINEZ-SILVESTRE Elodie DARNET Hugo LE CHEVALIER Olivier GUILLAUME Marc MOSSOLL-TORRES Laurent BARTHE Gilles POTTIER HervéPHILIPPE Fabien AUBRET 2020Integrative Zoology2020,15,6:0
12Use of a specific questionnaire and perineal electromyography to assess neuropathic pain after radical retropubic prostatectomy显示文摘Objective:Prostate cancer is the most frequent cancer in men and radical retropubic prostatectomy(RRP)is one of the first-line treatment.However,RRP has some side effects and can lead to chronic perineal pain.The objective of the study was to determine in patients suffering from perineal pain after RRP the possibility of a neurogenic damage by means of a specific questionnaire dedicated to track down neuropathic pain.Methods:Forty patients were explored by a specific and validated questionnaire,the Neuropathic Pain Symptom Inventory(NPSI).Patients were divided into two groups:Group A with an NSPI score≥4 was considered as suffering from neuropathic pain,and Group B was considered as a control group without neuropathic pain(NSPI score<4).All patients had a perineal electrophysiological testing to confirm the possibility of a neurogenic damage.Results:Group A was composed by 13 men and Group B by 27 men,with mean age 72.45 years and mean duration of pain 2.7 years.In Group A,the most frequent symptoms were burning sensation,electrical shock and numbness.Location of the pain was global perineal area(8/13),anus(10/13),penis(5/13)and glans penis(2/13).Electromyography(EMG)findings confirmed the presence of denervation and neurogenic damages compared with controls(p<0.001).Conclusion:One third of the patients consulting for chronic pain following RRP had probably a neuropathic lesion leading to a chronic perineal pain as suggested by an NSPI score≥4 and EMG alterations.Nicolas Turmel Samer Sheikh Ismael Camille Chesnel Audrey Charlanes Claire Hentzen Frédérique Le Breton Gérard Amarenco 2019Asian Journal of Urology2019,6,4:0
