1Pinacidil,a K_(atp) channel opener,identified as a novel agonist for TRPA1显示文摘The transient receptor potential Ankyrin 1(TRPA1) cation channel is activated by various pungent and irritant compounds,and it also mediates the perception of noxious cold.Identification of different agonists for this channel is important for understanding its activation mechanism.Therefore,a screen for novel TRPA1 agonists was performed using an agonist-induced calcium influx assay.Out of 90 compounds screened,pinacidil was identified as a novel agonist for this channel.Pinacidil is a known opener of the K atp channel,for which it has an EC50 value of 1-3 μmol/L.In comparison,the EC50 value of pinacidil for TRPA1 is relatively high(260 μmol/L).Recombinant HEK-TRPA1 cells did not respond to P1075,another K atp channel opener,suggesting that the effect of pinacidil on TRPA1 was highly specific.Further studies revealed that the agonist activity of pinacidil could be blocked by the TRP channel inhibitors,ruthenium red and HC-030031.Using glutathione(GSH) and site-specific mutagenesis,we demonstrated that pinacidil could activate TRPA1 by covalent modification of the critical amino acids C619,C639 and C663 in the N-terminus of TRPA1.MA LiangHui DENG Ying ZHANG Bi BAI YanQiu CAO Jing LI ShiYou LIU JianFeng 2012Chinese Science Bulletin2012,57,15:1
2Building a Highly Stable Ultrathin Nanoporous Layer Assisted by Glucose for Desalination显示文摘Although nanoporous membranes are of great interest in desalination,it is still challenging to construct highly permeable nanoporous membranes with excellent rejections for an efficient desalination process.In this work,highly permeable nanoporous membranes were built from renewable resources,assisted by the versatile functions of glucose and dopamine,with coupling reactive groups via interfacial reaction with 1,3,5-benzenetricarbonyl trichloride(TMC).The small molecules(0.66 nm)of glucose,which have high hydrophilicity,can diffuse into the membrane for an effective reaction to ensure structural integration.Our novel ultrathin(~44 nm)nanofiltration(NF)membrane exhibits ultra-high Na_(2)SO_(4)flux and excellent rejection of Na_(2)SO_(4)(66.5 L·m^(-2)·h^(-1),97.3%)and MgSO4(63.0 L·m^(-2)·h^(-1),92.1%)under a pressure of 5 bar(1 bar=10^(5)Pa)which is much superior to the performance of natural-product NF membranes.The membrane demonstrates excellent long-term stability,as well as tremendous acid-base and alkali-base stability and high anti-pollution capacity.The designed membrane materials and architecture open a new door to biopolymer-based separation membranes beyond existing membrane materials.Yanqiu Zhang Fan Yang Hongguang Sun Yongping Bai Songwei Li Lu Shao 2022Engineering2022,,9:0
3Dynamics of excited m-dichlorobenzene显示文摘Dynamics of excited m-dichlorobenzene is investigated in real time by femtosecond pump-probe method, combined with time-of-flight mass spectrometric detection in a supersonic molecular beam. The yields of the parent ion and daughter ion C6H4Cl+ are examined as a func-tion of the delay between the 270 and 810 nm femtosecond laser pulses, respectively. The life-time of the first singlet excited state S1 of m-dichlorobenzene is measured. The origin of this daughter ion C6H4Cl+ is discussed. The ladder mechanism is proposed to form the fragment ion. 4Development of a functional cell-based HTS assay for identification of NKCC1-negative modulators显示文摘Na–K–Cl cotransporter 1(NKCC1) cotransports Na+, K+, and Cl-ions across the plasma membrane into cells. Accumulation of Cl-ions in dorsal root ganglion neurons induces depolarizing GABAA receptors, which mediate presynaptic inhibition and filtration of sensory noise. The activity of the Na–K–Cl cotransporter is modulated by high-dose loop diuretics, such as furosemide and bumetanide. To identify NKCC1 modulators, we developed a functional cell-based assay feasible for highthroughput screening(HTS), in which the activity of NKCC1 was detected by a BTC-AM dye-based thallium transportation assay. We demonstrated that the influx of Tl?was mediated by NKCC1, which required the existence of Cl-ions and could be inhibited by bumetanide and furosemide.
