1Underexpression of LATS1 TSG in colorectal cancer is associated with promoter hypermethylation显示文摘AIM:To investigate large tumor suppressor 1 (LATS1 ) expression, promoter hypermethylation, and microsatellite instability in colorectal cancer (CRC).METHODS:RNA was isolated from tumor tissue of 142 CRC patients and 40 colon mucosal biopsies of healthy controls. After reverse transcription, quantitative polymerase chain reaction (PCR) was performed, and LATS1 expression was normalized to expression of the ACTB and RPL32 housekeeping genes. To analyze hypermethylation, genomic DNA was isolated from 44 tumor CRC biopsies, and methylation-specific PCR was performed. Microsatellite instability (MSI) status was checked with PCR using BAT26, BAT25, and BAT40 markers in the genomic DNA of 84 CRC patients, followed by denaturing gel electrophoresis. RESULTS:Decreased LATS1 expression was found in 127/142 (89.4%) CRC cases with the average ratio of the LATS1 level 10.33 ± 32.64 in CRC patients vs 32.85 ± 33.56 in healthy controls. The lowest expression was found in Dukes' B stage tumors and G1 (welldifferentiated) cells. Hypermethylation of the LATS1 promoter was present in 25/44 (57%) CRC cases analyzed. LATS1 promoter hypermethylation was strongly associated with decreased gene expression; methylated cases showed 162× lower expression of LATS1 than unmethylated cases. Although high-grade MSI (mutation in all three markers) was found in 14/84 (17%) cases and low-grade MSI (mutation in 1-2 markers) was found in 30/84 (36%) cases, we found no association with LATS1 expression. CONCLUSION:Decreased expression of LATS1 in CRC was associated with promoter hypermethylation, but not MSI status. 2Laser-induced white-light emission from graphene ceramics–opening a band gap in graphene显示文摘Recent theoretical and experimental studies have indicated the existence of a new stable phase of carbon with mixed sp^(2) and sp^(3) hybridized bonds—diaphite.Such a two-layered structure with sp^(2)/sp^(3) bonds may be observed after the photostimulation of highly oriented pyrolytic graphene with femtosecond laser pulses.This hidden multistability of graphene may be used to create a semiconducting phase immersed in the semimetallic continuum,resulting in bandgap opening.We demonstrate that bandgap opening and light emission from graphene is possible using continuous-wave laser beams with wavelengths from the visible(405 nm)to the near-infrared range(975 nm).We demonstrate that without the application of cooling,the effective temperature of the emitting sample remains lower than 900 K,which is far below the value predicted by the theory of black-body radiation.Moreover,light emission from a graphene sample may be observed at temperatures as low as 10 K.Wieslaw Strek Bartlomiej Cichy Lukasz Radosinski Pawel Gluchowski Lukasz Marciniak Mikolaj Lukaszewicz Dariusz Hreniak 2015Light(Science & Applications)2015,4,1:6

4Coherent supercontinuum generation in soft glass photonic crystal fibers显示文摘An overview of the progress on pulse-preserving, coherent, nonlinear fiber-based supercontinuum generation is presented. The context encompasses various wavelength ranges and pump sources, starting with silica photonic crystal fibers pumped with 1.0 μm femtosecond lasers up to chalcogenide step-index and microstructured fibers pumped from optical parametric amplifiers tuned to mid-infrared wavelengths. In particular, silica and silicatebased all-normal dispersion(ANDi) photonic crystal fibers have been demonstrated for pumping with femtosecond lasers operating at 1.56 μm with the recorded spectra covering 0.9–2.3 μm. This matches amplification bands of robust fiber amplifiers and femtosecond lasers. The review therefore focuses specifically on this wavelength range, discussing glass and nonlinear fiber designs, experimental results on supercontinuum generation up to the fundamental limit of oxide glass fiber transmission around 2.8 μm, and various limitations of supercontinuum bandwidth and coherence. Specifically, the role of nonlinear response against the role of dispersion profile shape is analyzed for two different soft glass ANDi fibers pumped at more than 2.0 μm. A spatio-temporal interaction of the fundamental fiber mode with modes propagating in the photonic lattice of the discussed ANDi fibers is shown to have positive effects on the coherence of the supercontinuum at pump pulse durations of 400 fs. 6Sphincter-preserving R0 total mesorectal excision with resection of internal genitalia combined with pre-or postoperative chemoradiation for T4 rectal cancer in females显示文摘AIM: To evaluate the impact of chemoradiation admi- nistered pre- or postoperatively on prognosis in females following R0 extended resection with sphincter- preserving total mesorectal excision (TME) for locally advanced rectal cancer and to assess the association between chemoradiation and intra- and postoperative variables. METHODS: Twenty-one females were treated for locally advanced but preoperatively assessed as primarily resectable rectal cancer involving reproductive organs. Anterior resection with TME and excision of internal genitalia was combined with neo- or adjuvant chemoradiation. Two-year disease-free survival analysis was performed with the Kaplan-Meier method and log- rank test. The association between chemoradiation and other variables was evaluated with the Fisher’s exact test and Mann-Whitney test. RESULTS: Survival rate decreased in anaemic females (51.5% vs 57.4%), in patients older than 60 years (41.8% vs 66.7%) with poorly differentiated cancers (50.0% vs 55.6%) and tumors located ≤ 7 cm from the anal verge (42.9% vs 68.1%) but with the lack of importance. Patients with negative lymph nodes and women chemoradiated preoperatively had significantly favourable prognosis (85.7% vs 35.7%; P= 0.03 and 80.0% vs 27.3%; P = 0.01, respectively). Preoperative chemoradiation compared to adjuvant radiochemotherapy was not significantly associated with the duration of surgery, incidence of intraoperative bowel perforation and blood loss ≥ 1 L, rate of postoperative bladder and anorectal dysfunction, and minimal distal resection margin. It significantly influenced minimal radial margin (mean 4.2 mm vs 1.1 mm; P < 0.01). CONCLUSION: Despite involving internal genitalia, long-term disease-free survival and sphincter preservation may be achieved with combined-modality therapy for females with T4 locally advanced rectal carcinoma. 14Applicability of surface directional wells for upper Silesia Basin coal seams' drainage ahead of mining显示文摘Methods of exploitation drainage, which is presently applied in polish hard coal mines in Upper Silesian Coal Basin(Poland), are not effective enough, high risk of methane hazard can be observed, and production capacity of the mining plant is not fully used. Methane hazard, which may occur during planned coal exploitation, is presented in this paper. Following parameters are taken into consideration in the forecasts: coal extraction parameters, geological and mining conditions, deposit's methane saturation degree and impact of coal exploitation on the degasification coefficient of the seams, which are under the influence of relaxation zone. This paper presents the results of the analysis aiming to verify applicability of drainage ahead of mining of the coal seams by using surface directional wells. Based on the collected data(coal seams' structural maps, profiles of the exploratory wells, geological cross-sections), the lab tests of drilling cores and direct wells' tests, static model of the deposit was constructed and suitable grid of directional wells from the surface was designed. Comparison of forecasted methane emission volume between the two methods is investigated.
