| 1 | The m^(6)A methylome of SARS-CoV-2 in host cells显示文摘The newly identified Severe Acute Respiratory Syndrome Coronavirus 2(SARS-CoV-2)has resulted in a global health emergency because of its rapid spread and high mortality.The molecular mechanism of interaction between host and viral genomic RNA is yet unclear.We demonstrate herein that SARS-CoV-2 genomic RNA,as well as the negative-sense RNA,is dynamically N6-methyladenosine(m^(6)A)-modified in human and monkey cells.Combined RIP-seq and miCLIP analyses identified a total of 8 m^(6)A sites at single-base resolution in the genome.Especially,epidemic strains with mutations at these identified m^(6)A sites have emerged worldwide,and formed a unique cluster in the US as indicated by phylogenetic analysis.Further functional experiments showed that m^(6)A methylation negatively regulates SARS-CoV-2 infection.SARS-CoV-2 infection also triggered a global increase in host m^(6)A methylome,exhibiting altered localization and motifs of m^(6)A methylation in mRNAs.Altogether,our results identify m^(6)A as a dynamic epitranscriptomic mark mediating the virus–host interaction. | Jun’e Liu Yan-Peng Xu Kai Li Qing Ye Hang-Yu Zhou Hanxiao Sun Xiaoyu Li Liu Yu Yong-Qiang Deng Rui-Ting Li Meng-Li Cheng Bo He Jia Zhou Xiao-Feng Li Aiping Wu Chengqi Yi Cheng-Feng Qin | 2021 | Cell Research2021,31,4: | 5 |
| 2 | Correlation of vascular endothelial growth factor to permeability of blood-brain barrier and brain edema during high-altitude exposure显示文摘BACKGROUND:Many studies have evaluated the role of vascular endothelial growth factor(VEGF) in traumatic brain edema and hemorrhagic brain edema. OBJECTIVE:To observe the effects of VEGF expression on permeability of the blood-brain barrier (BBB) during high-altitude and hypoxia exposure,and to investigate the correlation between VEGF expression and BBB permeability with regard to Evans blue staining and brain edema during high-altitude exposure. DESIGN,TIME AND SETTING:The randomized,controlled,animal study was performed at the Tanggula Etape,Central Laboratory of Chengdu Medical College,and Central Laboratory of General Hospital of Chengdu Military Area Command of Chinese PLA,China,from July 2003 to November 2004. MATERIALS:Quantitative RT-PCR kit(Sigma,USA),VEGF ELISA kit(Biosource,USA),and Evans blue(Jingchun,China) were acquired for this study. METHODS:A total of 180 Wistar rats were equally and randomly assigned to 15 groups:low-altitude (500 m),middle-altitude(2 880 m),high-altitude(4 200 m),super-high-altitude(5 000 m),1,3,5,7,9, 11,13,15,17,19,and 21 days of super high-altitude exposure.Wistar rats were exposed to various altitude gradients to establish a hypoxia model. MAIN OUTCOME MEASURES:Brain water content was calculated according to the wet-to-dry weight ratio.BBB permeability to Evans blue was determined by colorimetric method.VEGF mRNA and protein levels in brain tissues were detected using RT-PCR and double-antibody sandwich ELISA. RESULTS:Brain water content,BBB permeability to Evans blue,and VEGF mRNA and protein levels in brain tissues increased with increasing altitude and prolonged exposure to altitude.The greatest increase was determined on day 9 upon ascending 5 000 m.Simultaneously,VEGF expression positively correlated to BBB permeability of Evans blue and brain water content(r = 0.975,0.917,P<0.01). | 3 | Development and calibration of the Moon-based EUV camera for Chang'e-3显示文摘The process of development and calibration for the first Moon-based extreme ultraviolet(EUV) camera to observe Earth's plasmasphere is introduced and the design, test and calibration results are presented. The EUV camera is composed of a multilayer film mirror, a thin film filter, a photon-counting imaging detector, a mechanism that can adjust the direction in two dimensions, a protective cover, an electronic unit and a thermal control unit. The center wavelength of the EUV camera is 30.2 nm with a bandwidth of 4.6 nm. The field of view is 14.7° with an angular resolution of 0.08°, and the sensitivity of the camera is 0.11 count s-1Rayleigh-1. | 4 | Effect of endothelial progenitor cells derived from human umbilical cord blood on oxygen-induced retinopathy in mice by intravitreal transplantation显示文摘·AIM: To investigate the effect of endothelial progenitor cells(EPCs) labeled by carboxy fluorescein diacetate succinimidyl ester(CFSE) on murine oxygen-induced retinopathy(OIR) by intravitreal transplantation.·METHODS: After isolated from human umbilical cord blood mononuclear cells, EPCs were cultivated and then labeled with CFSE in vitro. C57BL/6J mice were placed to75% hyperoxia chamber from P7 to P12 to establish OIR model. At P12, OIR mice were intravitreally injected with1 μL suspension contained 2×105EPCs(EPCs group) or isometric phosphate buffered saline(PBS group). The contralateral eye of each mice received no injection(OIR group). Evans blue angiography and frozen section were examined to track the labeled cells in OIR group at P15 and P19. Using retina paraffin sections and adenosinediphos phatase staining at P12 and P19, the effect of EPCs on OIR mice was evaluated quantitatively and qualitatively.· RESULTS: The retinas from EPCs group with less non-perfusion area and fewer peripheral tufts wereobserved at P19, comparing with that from PBS or OIR group. The retinopathy in EPCs group receded earlier with less non-ganglion cells and neovascular nuclei,together with relatively regular distribution. The counts of the neovascular nuclei at P19 were reduced by 44% or45%, compared with those of OIR group or PBS group respectively. Three days after EPCs injection, a large number of EPCs appeared in the vitreous cavity and adhered to the retinal surface.
