| 1 | Ethanol extract from Gynostemma pentaphyllum ameliorates dopaminergic neuronal cell death in transgenic mice expressing mutant A53T human alpha-synuclein显示文摘Gynostemma(G.) pentaphyllum(Cucurbitaceae) contains various bioactive gypenosides. Ethanol extract from G. pentaphyllum(GP-EX) has been shown to have ameliorative effects on the death of dopaminergic neurons in animal models of Parkinson’s disease(PD) induced by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine-and 6-hydroxydopamine. PD patients exhibit multiple symptoms, so PD-related research should combine neurotoxin models with genetic models. In the present study, we investigated the ameliorative effects of GP-EX, including gypenosides, on the cell death of dopaminergic neurons in the midbrain of A53 T α-synuclein transgenic mouse models of PD(A53 T). Both GP-EX and gypenosides at 50 mg/kg per day were orally administered to the A53 T mice for 20 weeks.α-Synuclein-immunopositive cells and α-synuclein phosphorylation were increased in the midbrain of A53 T mice, which was reduced following treatment with GP-EX. Treatment with GP-EX modulated the reduced phosphorylation of tyrosine hydroxylase, extracellular signal-regulated kinase(ERK1/2), Bcl-2-associated death promoter(Bad) at Ser112, and c-Jun N-terminal kinase(JNK1/2) due to α-synuclein overexpression. In the A53 T group, GP-EX treatment prolonged the latency of the step-through passive avoidance test and shortened the transfer latency of the elevated plus maze test. Gypenosides treatment exhibited the effects and efficacy similar to those of GP-EX. Taken together, GP-EX, including gypenosides, has ameliorative effects on dopaminergic neuronal cell death due to the overexpression of α-synuclein by modulating ERK1/2, Bad at Ser112, and JNK1/2 signaling in the midbrain of A53 T mouse model of PD. Further studies are needed to investigate GP-EX as a treatment for neurodegenerative synucleinopathies, including PD. | 2 | Assignment and expression patterns of porcine muscle-specific isoform of phosphoglycerate mutase gene显示文摘It has been reported that the muscle-specific isoform (type M, PGAM2) of phosphoglycerate mutase (PGAM) is a housekeeping en- zyme; it catalyzes the conversion of 3-phosphoglycerate into 2-phosphoglycerate in the glycolysis process to release energy. It is encoded by the Pgam2 gene. In this study, the cDNA of the porcine Pgam2 was cloned. This gene contains an open reading frame of 765 bp en- coding a protein of 253 residues, and the predicted protein sequences share high similarity with other mammalians, 96% identity with humans, and 94% identity with mouse and rats. Pgam2 was mapped to SSC18q13-q21 by the RH panel. In this region, there are several QTLs, such as fat ratio, lean percentage, and diameter of muslce fiber, which affect meat production and quality. The reverse transcrip- tase-polymerase chain reaction revealed that the porcine Pgam2 gene was mainly expressed in the muscle tissue (skeletal muscle and cardiac muscle), and was expressed highly at skeletal muscle development stages (embryonic periods: 33, 65, and 90 days post-conception (dpc); postnatal pigs: 4 days and adult). This indicates that the Pgam2 gene plays an important role in muscle growth and development. | 7 | Simultaneous enhancement of coercivity and electric resistivity of Nd-Fe-B magnets by Pr-Tb-Al-Cu synergistic grain boundary diffusion toward high-temperature motor rotors显示文摘To high-power permanent magnetic motors,it is critical for Nd-Fe-B magnets to maintain the desirable coercivity at high-temperature operating conditions.To address this,two approaches have been proven effective:(1)enhancing the room temperature coercivity;(2)reducing the eddy current loss.However,these two items are difficult to be simultaneously achieved.Here,the grain boundary diffusion(GBD)of the Pr-Tb-Al-Cu-based source is applied to enhance the coercivity and electric resistivity at room temperature from 1101 kA m-1 and 2.13×10–6Ωm to 1917 kA m-1 and 2.60×10–6Ωm,and those at 120°C from 384 kA m-1 and 4.31×10–6Ωm to 783 kA m-1 and 4.86×10–6Ωm,respectively.Such optimization is ascribed to the improved formation depth of Tb-rich 2:14:1 shells with large magnetocrystalline anisotropy and the increased intergranular Pr-based oxides with high electric resistivity,induced by the coordination effects of Tb and Pr,as proven by the atomic-scale observations and the first principles calculations.It thus results in the simultaneously improved output power and energy efficiency of the motor because of the combination of magnetic thermal stability enhancement and eddy current loss reduction,as theoretically confirmed by electromagnetic simulation. | Jiayi He Jinwen Hu Bang Zhou Haoyang Jia Xiaolian Liu Zhenhua Zhang Lin Wen Lizhong Zhao Hongya Yu Xichun Zhong Xuefeng Zhang Zhongwu Liu | 2023 | Journal of Materials Science & Technology2023,,23: | 1 |
