1microRNAs and ceRNAs: RNA networks in pathogenesis of cancer显示文摘microRNAs (miRNAs) are a class of endogenous, single-stranded non-coding RNAs of 2023 nucleotides in length, functioning as negative regulators of gene expression at the post-transcriptional level. The dysregulation of miRNAs has been demonstrated to play critical roles in tumorigenesis, either through inhibiting tumor suppressor genes or activating oncogenes inappropriately. Besides their promising clinical applications in cancer diagnosis and treatment, recent studies have uncovered that miRNAs could act as a regulatory language, through which messenger RNAs, transcribed pseudogenes, and long noncoding RNAs crosstalk with each other and form a novel regulatory network. RNA transcripts involved in this network have been termed as competing endogenous RNAs (ceRNAs), since they influence each other's level by competing for the same pool of miRNAs through miRNA response elements (MREs) on their target transcripts. 2Biodegradation of geosmin in drinking water by novel bacteria isolated from biologically active carbon显示文摘Three strains of Gram-negative bacteria capable of removing geosmin from drinking water were isolated from biologically active carbon and identified to be Chryseobacterium sp., Sinorhizobium sp. and Stenotrophomonas sp. based on physio-biochemistry analysis and 16S rRNA gene sequence analysis. Removal efficiencies of 2 mg/L geosmin in mineral salts medium were 84.0%, 80.2% and 74.4% for Chryseobacterium sp., Sinorhizobium sp. and Stenotrophomonas sp., respectively, while removal efficiencies of 560 ng/L geosmin in filter influent were 84.8%, 82.3% and 82.5%, respectively. The biodegradation of geosmin was determined to be a pseudo first-order reaction, with rate constants at 2 mg/L and 560 ng/L being 0.097 and 0.086 day 1 , 0.089 and 0.084 day 1 , 0.074 and 0.098 day 1 for the above mentioned degraders, respectively. 5Blind signal separation of underdetermined mixtures based on clustering algorithms on planes显示文摘Based on clustering method on planes, blind signal separation (BSS) of underdetermined mixtures with three observed signals is discussed. The condition of sufficient sparsity of the source signals is not necessary when clustering method on planes is used. In other words, it needs not that only one source signal plays the main role among others at one time. The proposed method uses normal line clustering of planes first. Then, the mixing matrix can be identified via deciding the intersection lines of the planes. This method is an effective implement of the new theory presented by Georgiev. 7Optimal Groundwater Development in Coastal Aquifers Near Beihai, China显示文摘Groundwater resources occur in a multi aquifer system in the alluvial coastal plain near Beihai, China. The aquifers receive recharge from precipitation, canal and reservoir infiltration, and discharge through subterranean drainage into the sea and through artificial pumping. A quasi three dimensional finite element model has been used to simulate the spatial and temporal distribution of groundwater levels in the aquifers. Various input parameters were considered in the simulation model. A linear optimization model has been developed for groundwater development within the coastal aquifers. The objective function of the model is to maximize the total groundwater pumpage from the confined aquifer. The control of sea water intrusion is examined by the restriction of the water levels at points along the coast and of the pumping rates in coastal management cells. The response matrix used in the optimization model was generated from the simulation model by forecasting drawdown produced by pumping at a unit impulse discharge. 8Source number estimation and separation algorithms of underdetermined blind separation显示文摘Recently,sparse component analysis (SCA) has become a hot spot in BSS re-search. Instead of independent component analysis (ICA),SCA can be used to solve underdetermined mixture efficiently. Two-step approach (TSA) is one of the typical methods to solve SCA based BSS problems. It estimates the mixing matrix before the separation of the sources. K-means clustering is often used to estimate the mixing matrix. It relies on the prior knowledge of the source number strongly. However,the estimation of the source number is an obstacle. In this paper,a fuzzy clustering method is proposed to estimate the source number and mixing matrix simultaneously. After that,the sources are recovered by the shortest path method (SPM). 11Clinical Pathologic Study on Effect of Qianggan Capsule (强肝胶囊) in Treating Patients of Chronic Hepatitis B with Liver Cirrhosis显示文摘Objective: To explore the clinical therapeutic effect of Qianggan Capsule (QGC) in treating chronic hepatitis B with liver fibrosis from the pathological aspect. Methods: Sixty three patients of chronic hepatitis B with liver fibrosis were randomly divided into the treated group (n=45) and the control group (n=18). Both groups were treated with general liver protective drugs, such as Glucurone and vitamins B complex for 6 months. To the treated group, QGC was used additionally. The levels of serum alanine transaminase and liver fibrosis indexes including hyaluronic acid (HA), collagen type Ⅳ (C Ⅳ) and laminin (LN) as well as the pathological examination of liver biopsy were observed before and after treatment. Results: The liver cirrhosis indexes, HA, C Ⅳ and LN, were improved significantly in the treated group after treatment, P <0.05. The liver function improvement rate in the treated group and the control group was 90.3% and 66.7% respectively, comparison of the two groups showed insignificant difference, P >0.05. Pathological examination showed that the effective rate of treatment on liver inflammatory necrosis activity grade in the treated group was 57.8% and that on liver fibrosis stage was 75.6%, which were significantly improved as compared with those before treatment ( P <0.05 and P <0.01), while in the control group, no significant improvement was found after treatment ( P >0.05). 15A peptide fusion protein inhibits angiogenesis and tumor growth by blocking VEGF binding to KDR显示文摘Vascular endothelial growth factor (VEGF) binding to its tyrosine kinase receptors (KDR/FLK1, Flt-1) induces angiogenesis. In search of the peptides blocking VEGF binding to its receptor KDR/FLK1 to inhibit tumor-angiogenesis and growth, we screened a phage display pep-tide library with KDR as target protein, and some candidate peptides were isolated. In this study, we cloned the DNA fragment coding the peptide K237 from the library, into a vector pQE42 to express fusion protein DHFR-K237 in E. coli M15. The affection of fusion protein DHFR-K237 on endothelial cell proliferation and angiogenesis was investigated. In vitro, DHFR-K237 could completely block VEGF binding to KDR and significantly inhibit the VEGF-medi-ated proliferation of the human vascular endothelial cells. In vivo, DHFR-K237 inhibited angiogenesis in chick embryo chorioa- llantoric membrane and tumor growth in nude mice.
