| 1 | Methionine adenosyltransferases in liver cancer显示文摘Methionine adenosyltransferases(MATs)are essential enzymes for life as they produce S-adenosylmethionine(SAMe),the biological methyl donor required for a plethora of reactions within the cell.Mammalian systems express two genes,MAT1A and MAT2A,which encode for MATα1 and MATα2,the catalytic subunits of the MAT isoenzymes,respectively.A third gene MAT2B,encodes a regulatory subunit known as MATβwhich controls the activity of MATα2.MAT1A,which is mainly expressed in hepatocytes,maintains the differentiated state of these cells,whilst MAT2A and MAT2B are expressed in extrahepatic tissues as well as non-parenchymal cells of the liver(e.g.,hepatic stellate and Kupffer cells).The biosynthesis of SAMe is impaired in patients with chronic liver disease and liver cancer due to decreased expression and inactivation of MATα1.A switch from MAT1A to MAT2A/MAT2B occurs in multiple liver diseases and during liver growth and dedifferentiation,but this change in the expression pattern of MATs results in reduced hepatic SAMe level.Decades of study have utilized the Mat1a-knockout(KO)mouse that spontaneously develops non-alcoholic steatohepatitis(NASH)and hepatocellular carcinoma(HCC)to elucidate a variety of mechanisms by which MAT proteins dysregulation contributes to liver carcinogenesis.An increasing volume of work indicates that MATs have SAMe-independent functions,distinct interactomes and multiple subcellular localizations.Here we aim to provide an overview of MAT biology including genes,isoenzymes and their regulation to provide the context for understanding consequences of their dysregulation.We will highlight recent breakthroughs in the field and underscore the importance of MAT’s in liver tumorigenesis as well as their potential as targets for cancer therapy. | Ben Murray Lucia Barbier-Torres Wei Fan JoséM Mato Shelly C Lu | 2019 | World Journal of Gastroenterology2019,25,31: | 10 |
| 2 | Phytochemical constituents and antibacterial efficacy of the flowers of Peltophomm pterocarpum(DC.) Baker ex Heyne显示文摘Objective:To investigate the preliminary phytochemistry and antibacterial activity of the flower extract of Peltophorum pterocarpum.Methods:Phytochemical analysis was done by using the standard methods given by Harbone.The methanolic flower extract were tested against Escherichia coli,Klebsiella pneumonia,Pseudomonas aeruginosa.Staphylococcus aureus. | 4 | Gastrointestinal perforation in metastatic colorectal cancer patients with peritoneal metastases receiving bevacizumab显示文摘AIM:To investigate the safety and efficacy of adding bevacizumab to first-line chemotherapy in metastatic colorectal cancer patients with peritoneal disease.METHODS:We compared rates of gastrointestinal perforation in patients with metastatic colorectal cancer and peritoneal disease receiving first-line chemotherapy with and without bevacizumab in three distinct cohorts:(1) the AGITG MAX trial(Phase Ⅲ randomised clinical trial comparing capecitabine vs capecitabine and bevacizumab vs capecitabine,bevacizumab and mitomycin C);(2) the prospective Treatment of Recurrent and Advanced Colorectal Cancer(TRACC) registry(any first-line regimen ± bevacizumab);and(3) two cancer centres in New South Wales,Australia [Macarthur Cancer Therapy Centre and Liverpool Cancer Therapy Centre(NSWCC) from January 2005 to Decenber 2012,(any first-line regimen ± bevacizumab).For the AGITG MAX trial capecitabine was compared to the other two arms(capecitabine/bevacizumab and capecitabine/bevacizumab/mitomycin C).In the AGITG MAX trial and the TRACC registry rates of gastrointestinal perforation were also collected in patients who did not have peritoneal metastases.Secondary endpoints included progression-free survival,chemotherapy duration,and overall survival.Time-toevent outcomes were estimated using the Kaplan-Meier method and compared using the log-rank test.RESULTS:Eighty-four MAX,179 TRACC and 69 NSWCC patients had peritoneal disease.There were no gastrointestinal perforations recorded in either the MAX subgroup or the NSWCC cohorts.Of the patients without peritoneal disease in the MAX trial,4/300(1.3%) in the bevacizumab arms had gastrointestinal perforations compared to 1/123(0.8%) in the capecitabine alone arm.In the TRACC registry 3/126(2.4%) patients who had received bevacizumab had a gastrointestinal perforation compared to 1/53(1.9%) in the chemotherapy alone arm.In a further analysis of patients without peritoneal metastases in the TRACC registry,the rate of gastrointestinal perforations was 9/369(2.4%) in the chemotherapy/bevacizumab group and 5/177(2.8%) in the chemotherapy alone group.The addition of bevacizumab to chemotherapy was associated with improved progression-free survival in all three cohorts:MAX 6.9 m vs 4.9 m,HR = 0.64(95%CI:0.42-1.02);P = 0.063;TRACC 9.1 m vs 5.5 m,HR = 0.61(95%CI:0.37-0.86);P = 0.009;NSWCC 8.7 m vs 6.8 m,HR = 0.75(95%CI:0.43-1.32);P = 0.32.Chemotherapy duration was similar across the groups.CONCLUSION:Patients with peritoneal disease do not appear to have an increased risk of gastrointestinal perforations when receiving first-line therapy with bevacizumab compared to systemic therapy alone. | Aflah Roohullah Hui-Li Wong Katrin M Sjoquist Peter Gibbs Kathryn Field Ben Tran Jeremy Shapiro Joe Mckendrick Desmond Yip Louise Nott Val Gebski Weng Ng Wei Chua Timothy Price Niall Tebbutt Lorraine Chantrill | 2015 | World Journal of Gastroenterology2015,21,17: | 3 |

| 7 | Use of human pluripotent stem cells to study and treat retinopathies显示文摘Human cell types affected by retinal diseases(such as age-related macular degeneration or retinitis pimentosa) are limited in cell number and of reduced accessibility. As a consequence, their isolation for in vitro studies of disease mechanisms or for drug screening efforts is fastidious. Human pluripotent stem cells(h PSCs), either of embryonic origin or through reprogramming of adult somatic cells,represent a new promising way to generate models of human retinopathies, explore the physiopathological mechanisms and develop novel therapeutic strategies. Disease-specific human embryonic stem cells were the first source of material to be used to study certain disease states. The recent demonstration that human somatic cells, such as fibroblasts or blood cells, can be genetically converted to induce pluripotent stem cells together with the continuous improvement of methods to differentiate these cells into disease-affected cellular subtypes opens new perspectives to model and understand a large number of human pathologies, including retinopathies. This review focuses on the added value of h PSCs for the disease modeling of human retinopathies and the study of their molecular pathological mechanisms. | 9 | Radiolarian and Planktic Foraminifera Biostratigraphy of the Early Albian Organic Rich Beds of Fahdene Formation,Northern Tunisia显示文摘The lower part of Fahdene Formation outcropping in northeastern Tunisia is represented by pelagic sequences and characterized by an important siliciclastic fraction that includes radiolarian and organic-rich beds of Allam Member.?Litho-biostratigraphic analysis based on planktic foraminifera and radiolaria shows that deposition of organic-rich beds of Allam Member is confined to the Microhedbergella rischi Zone through the lower part of Ticinella primula Zone.?Age diagnostic radiolaria recovered from these beds has been identified to constrain a direct age of black shale deposition.The assemblages can be correlated with the radiolarian biochronozone U.A.10–11 indicating an Early Albian to early Middle Albian in age.Radiolarian assemblages composed of species characteristic of U.A.10 biochronozone(A.montisserei,D.gracilis)are associated with radiolarian taxa belonging to Costata Zone(U.A.9 Pseudoeucyrtis hanni and Thanarla pseudodecora),which can be correlated with Dercourt Member in the Ionian Zone and Paquier level of the Vocontian Basin(southeast of France).Biostratigraphic and abundance curves analysis has demonstrated significant diversification of cryptocephalic Nassellaria and Archaeodictyomitrae,probably adapted to eutrophic conditions that characterized the Aptian-Albian transition.Ecological conditions may have governed the stratigraphic range of many cosmopolitan taxa(i.e.,Pseudodictyomitrae lodogaensis)compared with stratigraphic distributions schemes reported from other domains.The timing of black shales deposition was diachronic due to local geodynamic conditions and upwelling currents distribution.The Allam black shales are correlative with the oceanic anoxic event OAE1b which is characterized by the widespread of supraregionnally organic-rich beds in the Mediterranean Tethys basins across the Aptian-Albian transition. | M Ben Fadhel T Zouaghi A Amri M Ben Youssef | 2014 | Journal of Earth Science2014,25,1: | 2 |
