| 1 | Comparative study of CEA and CA19-9 in esophageal,gastric and colon cancers individually and in combination(ROC curve analysis)显示文摘Objective:To determine the clinical serum levels of carcinoembryonic antigen(CEA) and carbohydrate antigen 19-9(CA19-9),individually and in combination,for the diagnosis of 50 healthy subjects and 150 cases of esophageal,gastric,and colon cancers.Methods:The sensitivities of the two markers were compared individually and in combination,with specificity set at 100%.Receiver operating characteristic(ROC) curves were plotted.Results:Serum CEA levels were significantly higher in cancer patients than in the control group.The sensitivity of CEA was determined:in esophageal cancer,sensitivity=28%,negative predictive value(NPV)=61.72%,and AUC=0.742(SE=0.05),with a significance level of P<0.0001;in gastric cancer,sensitivity=30%,NPV=58.82%,and AUC=0.734(SE=0.05),with a significance level of P<0.0001;in colon cancer,sensitivity=74%,NPV=79.36%,and AUC=0.856(SE=0.04),with a significance level of P<0.0001.The sensitivity of CA19-9 was also evaluated:in esophageal cancer,sensitivity=18%,NPV=54.94%,and AUC=0.573(SE=0.05),with a significance level of P=0.2054.In gastric cancer,sensitivity=42%,NPV=63.29%,and AUC=0.679(SE=0.05),with a significance level of P<0.0011.In colon cancer,sensitivity=26%,NPV=57.47%,and AUC=0.580(SE=0.05),with a significance level of P=0.1670.The following were the sensitivities of CEA/CA19-9 combined:in esophageal cancer,sensitivity=42%,NPV=63.29%,SE=0.078(95% CI:0.0159-0.322);gastric cancer,sensitivity=58%,NPV=70.42%,SE=0.072(95% CI:-0.0866-0.198);and colon cancer,sensitivity=72%,NPV=78.12%,SE=0.070(95% CI:0.137-0.415).Conclusion:CEA exhibited the highest sensitivity for colon cancer,and CA19-9 exhibited the highest sensitivity for gastric cancer.Combined analysis indicated an increase in diagnostic sensitivity in esophageal and gastric cancer compared with that in colon cancer. | Bhawna Bagaria Sadhna Sood Rameshwaram Sharma Soniya Lalwani | 2013 | Cancer Biology & Medicine2013，10，3： | 53 |

Our understanding of insulin resistance has improved tremendously over the years,but certain aspects of its estimation still remain elusive to researchers and clinicians.The quantitative assessment of insulin sensitivity is not routinely used during biochemical investigations for diagnostic purposes,but the emerging importance of insulin resistance has led to its wider application research studies.Evaluation of a number of clinical states where insulin sensitivity is compromised calls for assessment of insulin resistance. Insulin resistance is increasingly being assessed in various disease conditions where it aids in examining their pathogenesis,etiology and consequences. | 6 | Cell-type specificity of β-actin expression and its clinicopathological correlation in gastric adenocarcinoma显示文摘AIM:To investigate cell type specific distribution ofβ-actin expression in gastric adenocarcinoma and its correlation with clinicopathological parameters.METHODS:β-actin is a housekeeping gene,frequently used as loading control,but,differentially expresses in cancer.In gastric cancer,an overall increased expression ofβ-actin has been reported using tissue disruptive techniques.At present,no histological data is available to indicate its cell type-specific expression and distribution pattern.In the present study,we analyzedβ-actin expression and distribution in paired normal and tumor tissue samples of gastric adenocarcinoma patients using immunohistochemistry(IHC),a tissue non-disruptive technique as well as tissue disruptive techniques like reverse transcriptase-polymerase chain reaction(RT-PCR)and western blotting.Correlation ofβ-actin level with clinicopathological parameters was done using univariate analysis.RESULTS:The results of this study showed significant overexpression,at both mRNA and protein level in tumor tissues as confirmed by RT-PCR(1.47±0.13 vs2.36±0.16;P<0.001)and western blotting(1.92±0.26 vs 2.88±0.32;P<0.01).IHC revealed thatβ-actin expression is majorly distributed between epithelial and inflammatory cells of the tissues.Inflammatory cells showed a significantly higher expression compared to epithelial cells in normal(2.46±0.13 vs 5.92±0.23,P<0.001),as well as,in tumor tissues(2.79±0.24 vs6.71±0.14,P<0.001).Further,comparison of immunostaining between normal and tumor tissues revealed that both epithelial and inflammatory cells overexpressβ-actin in tumor tissues,however,significant difference was observed only in inflammatory cells(5.92±0.23vs 6.71±0.14,P<0.01).Moreover,combined expression in epithelial and inflammatory cells also showed significant increase(4.19±0.15 vs 4.75±0.14,P<0.05)in tumor tissues.In addition,univariate analysis showed a positive correlation ofβ-actin level of inflammatory cells with tumor grade(P<0.05)while epithelial cells exhibited negative correlation(P>0.05).CONCLUSION:In gastric cancer,β-actin showed an overall higher expression predominantly contributed by inflammatory or tumor infiltrating immune cells of the tissue microenvironment and correlates with tumor grade. | Shafqat A Khan Monica Tyagi Ajit K Sharma Savio G Barreto Bhawna Sirohi Mukta Ramadwar Shailesh V Shrikhande Sanjay Gupta | 2014 | World Journal of Gastroenterology2014，20，34： | 2 |
