1Dietary advanced glycation end-products aggravate non-alcoholic fatty liver disease显示文摘AIM To determine if manipulation of dietary advanced glycation end product(AGE), intake affects nonalcoholic fatty liver disease(NAFLD) progression and whether these effects are mediated via RAGE. METHODS Male C57Bl6 mice were fed a high fat, high fructose, high cholesterol(HFHC) diet for 33 wk and compared with animals on normal chow. A third group were given a HFHC diet that was high in AGEs. Another group was given a HFHC diet that was marinated in vinegar to prevent the formation of AGEs. In a second experiment, RAGE KO animals were fed a HFHC diet or a high AGE HFHC diet and compared with wildtype controls. Hepatic biochemistry, histology, picrosirius red morphometry and hepatic mR NA were determined. RESULTS Long-term consumption of the HFHC diet generated significant steatohepatitis and fibrosis after 33 wk. In this model, hepatic 4-hydroxynonenal content(a marker of chronic oxidative stress), hepatocyte ballooning, picrosirius red staining, α-smooth muscle actin and collagen type 1A gene expression were all significantly increased. Increasing the AGE content of the HFHC diet by baking further increased these markers of liver damage, but this was abrogated by pre-marination in acetic acid. In response to the HFHC diet, RAGE-/-animals developed NASH of similar severity to RAGE+/+ animals but were protected from the additional harmful effects of the high AGE containing diet. Studies in isolated Kupffer cells showed that AGEs increase cell proliferation and oxidative stress, providing a likely mechanism through which these compounds contribute to liver injury. CONCLUSION In the HFHC model of NAFLD, manipulation of dietary AGEs modulates liver injury, inflammation, and liver fibrosis via a RAGE dependent pathway. 4Comparative effectiveness of Biobrane^(■),RECELL^(■)Autologous skin Cell suspension and Silver dressings in partial thickness paediatric burns:BRACS randomised trial protocol显示文摘Background:Mixed partial thickness burns are the most common depth of burn injury managed at a large Australian paediatric hospital specialty burns unit.Prolonged time until re-epithelialisation is associated with increased burn depth and scar formation.Whilst current wound management approaches have benefits such as anti-microbial cover,these are not without inherent limitations including multiple dressing changes.The Biobrane^(■)RECELL^(■)Autologous skin Cell suspension and Silver dressings(BRACS)trial aims to identify the most effective wound management approach for mixed partial thickness injuries in children.Methods:All children presenting with an acute burn injury to the study site will be screened for eligibility.This is a single-centre,three-arm,parallel group,randomised trial.Children younger than 16 years,with burns≥5%total body surface area involving any anatomical location,up to 48 h after the burn injury,and of a superficial partial to mid-dermal depth,will be included.A sample size of 84 participants will be randomised to standard silver dressing or a Regenerative Epithelial Suspension(RES^(TM))with Biobrane^(■)or Biobrane^(■)alone.The first dressing will be applied under general anaesthesia and subsequent dressings will be changed every 3 to 5 days until the wound is≥95%re-epithelialised,with re-epithelialisation time the primary outcome.Secondary outcomes of acute pain,acute itch,scar severity,health-related quality of life,treatment satisfaction,dressing application ease and healthcare resource use will be assessed at each dressing change and 3,6 and 12 months post-burn injury.Discussion:The findings of this study can potentially change the wound management approach for superficial partial to mid-dermal burns in children locally and worldwide.Trial registration:The Australian New Zealand Clinical Trials Registry(ACTRN12618000245291)approved prospective registration on 15 February 2018.Registration details can be viewed at http://gffzzf8a84f62798d4facs666uofk6wc0q6kpc.ffgz.tsg.suse.edu.cn/Trial/Registration/TrialReview.aspx?id=374272&isReview=true.Anjana Bairagi Bronwyn Griffin Zephanie Tyack Dimitrios Vagenas Steven M.McPhail Roy Kimble 2019Burns & Trauma2019,7,1:3
5Pregnancy and neonatal outcomes in Indigenous Australians with diabetes in pregnancy显示文摘AIM: To perform a systematic review of reported neonatal and pregnancy outcomes of Indigenous Australians with diabetes in pregnancy(DIP).METHODS: Electronic searches of Pub Med and Web of Science were carried out. Articles were selected if they contained original data on DIP outcomes in Indigenous Australians. There were no specific exclusion criteria.RESULTS: A total of eight articles, predominantly from Queensland and Western Australia were identified once inclusion criteria were applied. Birth data from midwifery registries or paper charts encompassing years 1985-2008 were used. A total of 465591 pregnant women with and without DIP were included in the eight studies, with 1363 being Indigenous women with DIP. Indigenous Australians experienced increased rates of many known adverse outcomes of DIP including: macrosomia, caesarean section, congenital deformities, low birth weight, hypoglycaemia, and neonatal trauma. There were regional differences among Indigenous Australians, particularly regional/remote vs metropolitan populations where the regional/remote data showed worse outcomes. Two of the articles did not note a difference between Aboriginals and Caucasians in the rates of measured adverse outcome. Studies varied significantly in size, measured outcomes, and subsequent analysis.CONCLUSION: The health disparities between Indigenous Australians and non-Indigenous Australians are further evidenced by poorer outcomes in DIP. 11Tailoring specific properties of polymer-based composites by using graphene and its associated compounds显示文摘Graphene and its associated compounds have been identified as extraordinary structural nano-fillers that can tailor the properties of new polymer-based composites with specific functionalities.Graphene possesses perfect 2D atomic architecture and has a large surface area that enhances the bonding,with tailored functional groups on its surface with polymer chains to form high strength composites.Recently,a lot of research has focused on developing composites with high specific strength,high electrical and thermal conductivities,high impact resistance,excellent energy storage capability and ability to maintain their strength at low-temperature environments by using graphene,graphene oxide(GO),reduced graphene oxide(rGO)or carbon nanotube/graphene.Ultimate goals are to design composites that can meet specific requirements for different engineering applications.In this paper,the discussion on all aspects in relation to the use of graphene and its associated compounds for advanced composites will be given in detail.The potentiality of using these materials for high-tech applications will be explored.Up to date,it has been proved that the use of graphene-based nanofillers does improve the mechanical,interfacial bonding,electrical,thermal and electromagnetic interference shield properties of polymer-based materials.Appropriately adding a small amount of these nanofillers do make a big difference to the properties of host materials.Pui-yan Hung Kin Tak Lau Qiaoshi Guo Baohua Jia Bronwyn Fox 2020International Journal of Smart and Nano Materials2020,11,2:2
