2Side Effects during Treatment of Advanced Gastric Carcinoma by Chemotherapy Combined with CIK-cell Transfusion in Elderly People显示文摘OBJECTIVE To study the side effects and therapeutic results of autologous cytokine-induced killer(CIK)cell treatment in elderly patients with advanced gastric cancer. METHODS CIK cells were induced and cultured using biotechnics in vitro,and then the cells were infused back into the patients.Sixty elderly gastric cancer patients treated by chemotherapy(FOLFOX4 protocol)were followed-up.Among them,29 patients were treated with CIK cells during application of chemotherapy.Short-term curative effects and adverse events from the CIK transfusion and chemotherapy were observed. RESULTS Eight cases developed partial remission(PR),9 cases moderate remission(MR),7 cases stable disease(SD)and 5 cases progressive disease(PD).Out of a total of 29 patients who received chemotherapy combined with autologous CIK therapy, the total remission rate(PR+MR)was 58.6%.The total remission rate following chemotherapy alone was 45.2%,including 5 PR cases,9 MR cases,7 SD cases,and 10 PD cases.There was a relatively lower rate of severe chemotherapic toxicities in the CIK- cell transfusion group.Side e?ects of autologous CIK transfusion included chills(13 cases),fever(9 cases),nausea and vomiting (1 case)and general malaise(3 cases).Side effects were treated with conventional therapy resulting in their amelioration.No patients developed shock,blood capillary leakage syndrome,or abnormalities in routine blood,urine,liver and renal function tests. CONCLUSION Adoptive immunotherapy with autologous CIK cells may decrease the clinical signs and symptoms of elderly patients who suffer from advanced gastric cancer.Adverse reactions of patients can be alleviated by conventional therapy. 3Investigation into the flotation response of refractory molybdenum ore to depressant mixtures:A case study显示文摘In this paper, bulk flotation followed by separation was investigated to concentrate purified molybdenite product from Jinduicheng molybdenum ores(Shanxi province, China). The bench scale tests mainly focussed on separation of molybdenite from other sulfide minerals using the new type of depressants.The effect of each single depressant, including organic depressant-modified dextrin(MD), P-Nokes reagent(PN) and sodium trithiocarbonate(ST), and their mixtures on galena, chalcopyrite and other sulfide ores, was examined in turn by changing the concentrations used in cleaner flotation tests. Closed circuit experiments were carried out under the optimal condition and satisfying recovery and grade of molybdenite concentrate could be achieved(86.294% and 53.157%, respectively). 4Entinostat,a classⅠselective histone deacetylase inhibitor,plus exemestane for Chinese patients with hormone receptor-positive advanced breast cancer:A multicenter,randomized,double-blind,placebo-controlled,phase 3 trial显示文摘Entinostat plus exemestane in hormone receptor-positive(HR+)advanced breast cancer(ABC)previously showed encouraging outcomes.This multicenter phase 3 trial evaluated the efficacy and safety of entinostat plus exemestane in Chinese patients with HR+ABC that relapsed/progressed after≥1 endocrine therapy.Patients were randomized(2:1)to oral exemestane 25 mg/day plus entinostat(n=235)or placebo(n=119)5 mg/week in 28-day cycles.The primary endpoint was the independent radiographic committee(IRC)-assessed progression-free survival(PFS).The median age was 52(range,28—75)years and 222(62.7%)patients were postmenopausal.CDK4/6 inhibitors and fulvestrant were previously used in 23(6.5%)and 92(26.0%)patients,respectively.The baseline characteristics were comparable between the entinostat and placebo groups.The median PFS was 6.32(95%CI,5.30—9.11)and 3.72(95%CI,1.91—5.49)months in the entinostat and placebo groups(HR,0.76;95%CI,0.58—0.98;P=0.046),respectively.Grade≥3 adverse events(AEs)occurred in 154(65.5%)patients in the entinostat group versus 23(19.3%)in the placebo group,and the most common grade≥3 treatment-related AEs were neutropenia[103(43.8%)],thrombocytopenia[20(8.5%)],and leucopenia[15(6.4%)].Entinostat plus exemestane significantly improved PFS compared with exemestane,with generally manageable toxicities in HR+ABC(ClinicalTrials.gov#NCT03538171).Binghe Xu Qingyuan Zhang Xichun Hu Qing Li Tao Sun Wei Li Quchang Ouyang Jingfen Wang Zhongsheng Tong Min Yan Huiping Li Xiaohua Zeng Changping Shan Xian Wang Xi Yan Jian Zhang Yue Zhang Jiani Wang Liang Zhang Ying Lin Jifeng Feng Qianjun Chen Jian Huang Lu Zhang Lisong Yang Ying Tian Hongyan Shang 2023Acta Pharmaceutica Sinica B2023,13,5:3
5N-acetyltransferase 10 promotes colon cancer progression by inhibiting ferroptosis through N4-acetylation and stabilization of ferroptosis suppressor protein 1 (FSP1) mRNA显示文摘Background:N-acetyltransferase 10(NAT10)is the only enzyme known tomediate the N4-acetylcytidine(ac4C)modification of mRNA and is crucial formRNA stability and translation efficiency.However,its role in cancer development and prognosis has not yet been explored.This study aimed to examine the possible role of NAT10 in colon cancer.Methods:The expression levels ofNAT10were evaluated by immunohistochemical analyses with a colon cancer tissue microarray,and its prognostic value in patients was further analyzed.Quantitative real-time polymerase chain reaction(qRT-PCR)and Western blotting were performed to analyze NAT10 expression in harvested colon cancer tissues and cell lines.Stable NAT10-knockdown and NAT10-overexpressing colon cancer cell lines were constructed using lentivirus.The biological functions of NAT10 in colon cancer cell lines were analyzed in vitro by Cell Counting Kit-8(CCK-8),wound healing,Transwell,cell cycle,and ferroptosis assays.Xenograft models were used to analyze the effect of NAT10 on the tumorigenesis and metastasis of colon cancer cells in vivo.Dot blotting,acetylated RNA immunoprecipitation-qPCR,and RNA stability analyses were performed to explore the mechanism by which NAT10 functions in colon cancer progression.Results:NAT10 was upregulated in colon cancer tissues and various colon cancer cell lines.This increased NAT10 expression was associated with shorter patient survival.Knockdown of NAT10 in two colon cancer cell lines(HT-29 and LoVo)impaired the proliferation,migration,invasion,tumor formation and metastasis of these cells,whereas overexpression of NAT10 promoted these abilities.Further analysis revealed that NAT10 exerted a strong effect on the mRNA stability and expression of ferroptosis suppressor protein 1(FSP1)in HT-29 and LoVo cells.In these cells,FSP1 mRNA was found to be modified by ac4C acetylation,and this epigenetic modification was associated with the inhibition of ferroptosis.Conclusions:Our study revealed that NAT10 plays a critical role in colon cancer development by affecting FSP1 mRNA stability and ferroptosis,suggesting that NAT10 could be a novel prognostic and therapeutic target in colon cancer.Xiao Zheng Qi Wang You Zhou Dachuan Zhang Yiting Geng Wenwei Hu Changping Wu Yufang Shi Jingting Jiang 2022Cancer Communications2022,42,12:3
6Wavelength-selective 2 × 2 optical switch based on a Ge2Sb2Te5-assisted microring显示文摘A novel wavelength-selective 2×2 optical switch based on a Ge2Sb2Te5(GST)-assisted microring-resonator(MRR)is proposed.The present GST-assisted MRR consists of two access optical waveguides and an MRR coupled with a bent GST-loaded silicon photonic waveguide.The 2×2 optical switch is switched ON or OFF by modifying the GST state to be crystalline or amorphous.In particular,the microring waveguide and the bent GST-loaded waveguide are designed to satisfy the phase-matching condition when the GST is crystalline.As a result,the MRR becomes highly lossy and the resonance peak is depressed significantly.On the other hand,when it is off,there is little coupling due to the significant phase mismatching.Consequently,one has a low-loss transmission at the drop port for the resonance wavelength.In this paper,the simulation using the three-dimensional finite-difference method shows that the extinction ratio of the designed photonic switch is^20 d B at the resonance wavelength,while the excess losses at the through port and drop port are 0.9 d B and 2 d B.In particular,the resonance wavelength changes little between the ON and OFF states,which makes it suitable for multichannel wavelength-division-multiplexing systems.Changping Zhang Ming Zhang Yiwei Xie Yaocheng Shi Rajesh Kumar Roberto R.Panepucci Daoxin Dai 2020Photonics Research2020,8,7:3
7Tectonic Evolution of the Junggar Foreland Basin in the Late Carboniferous-Permian显示文摘A comprehensive study has been carried out to subdivide andcorrelate the Upper Carboniferous and Permian sedimentary successions in the Junggar basin based on outcrops and drilling and geophysical data. The study results, combined with geological analyses of the basin's periphery and the basement, as well as studies of the sedimentary rocks within the basin, the unconformities, tectonic geometry, kinematics and geodynamics, lead to the conclusion that the Junggar basin was characterized by the development of foreland basin systems during the Late Carboniferous and Per mian, During that period, three foreland basin systems were developed: (1) the northwest foreland basin system, which trended nearly north-south from Mahu to the Chepaizi Palaeo-mountain during its early stage of development and thus it was also referred to as the west foreland basin system; (2) the Karamaili foreland basin system in the east and (3) the Northern Tianshan foreland basin system in the south. These systems are different in size, development stage and time of formation. The first two are developed earlier than the third, but they are smaller in size. All the structures in the Junggar basin have resulted from the integration and superposition of structural elements in the above three systems. In general, the development of the Junggar basin can be divided into four stages. Stage I was marked by the creation and evolution of the marginal western foreland and the peripheral Karamaili foreland basin systems during the Late Carboniferous-early Early Permian (C3-Pja). Stage Ⅱ was characterized by the development of complicated foreland basin systems during the mid dle-late Early Permian (Pjb-Pf) when the three foreland basin systems took their shapes. Stage Ⅲ was the integration stage of peripheral foreland basin systems during the Middle Permian (P2) in which steady and slow tectonic subsidence prevailed. 9Horizontal and vertical gene transfer drive sediment antibiotic resistome in an urban lagoon system显示文摘Rapid urbanization has resulted in pervasive occurrence of antibiotic resistance genes(ARGs)in urban aquatic ecosystems.However,limited information is available concerning the ARG profiles and the forces responsible for their assembly in urban landscape lagoon systems.Here,we employed high-throughput quantitative PCR(HT-q PCR)to characterize the spatial variations of ARGs in surface and core sediments of Yundang Lagoon,China.The results indicated that the average richness and absolute abundance of ARGs were 11 and 53 times higher in the lagoon sediments as compared to pristine reference Tibetan lake sediments,highlighting the role of anthropogenic activities in ARG pollution.Co-occurrence network analysis indicated that various anaerobic prokaryotic genera belonging to Alpha-,Deltaproteobacteria,Bacteroidetes,Euryarchaeota,Firmicutes and Synergistetes were the potential hosts of ARGs.The partial least squares-path modeling(PLS-PM)analysis revealed positive and negative indirect effects of physicochemical factors and heavy metals on the lagoon ARG profiles,via biotic factors,respectively.The horizontal(mediated by mobile genetic elements)and vertical(mediated by prokaryotic communities)gene transfer may directly contribute the most to drive the abundance and composition of ARGs,respectively.Furthermore,the neutral community model demonstrated that the assembly of sediment ARG communities was jointly governed by deterministic and stochastic processes.Overall,this study provides novel insights into the diversity and distribution of ARGs in the benthic habitat of urban lagoon systems and underlying mechanisms for the spread and proliferation of ARGs.Hongjie Wang Liyuan Hou Yongqin Liu Keshao Liu Lanping Zhang Fuyi Huang Lin Wang Azhar Rashid Anyi Hu Changping Yu 2021Journal of Environmental Sciences2021,33,4:2
10Polyploidy events shaped the expansion of transcription factors in Cucurbitaceae and exploitation of genes for tendril development显示文摘Cucurbitaceae is one of the most important plant families distributed worldwide.Transcription factors(TFs)regulate plant growth at the transcription level.Here,we performed a systematic analysis of 42641 TFs from 63 families in 14 Cucurbitaceae and 10 non-cucurbit species.Whole-genome duplication(WGD)was the dominant event type in almost all Cucurbitaceae plants.The TF families were divided into 1210 orthogroups(OGs),of which,112 were unique to Cucurbitaceae.Although the loss of several gene families was detected in Cucurbitaceae,the gene families expanded in five species that experienced a WGD event comparing with grape.Our findings revealed that the recent WGD events that had occurred in Cucurbitaceae played important roles in the expansion of most TF families.The functional enrichment analysis of the genes that significantly expanded or contracted uncovered five gene families,AUX/IAA,NAC,NBS,HB,and NF-YB.Finally,we conducted a comprehensive analysis of the TCP gene family and identified 16 tendril-related(TEN)genes in 11 Cucurbitaceae species.Interestingly,the characteristic sequence changed from CNNFYFP to CNNFYLP in the TEN gene(Bhi06M000087)of Benincasa hispida.Furthermore,we identified a new characteristic sequence,YNN,which could be used for TEN gene exploitation in Cucurbitaceae.In conclusion,this study will serve as a reference for studying the relationship between gene family evolution and genome duplication.Moreover,it will provide rich genetic resources for functional Cucurbitaceae studies in the future.Yu Zhang Yingchao Zhang Bing Li Xiao Tan Changping Zhu Tong Wu Shuyan Feng Qihang Yang Shaoqin Shen Tong Yu Zhuo Liu Xiaoming Song 2022Horticultural Plant Journal2022,8,5:2
11An empirical approach for predicting burden velocities in rock blasting显示文摘An analytical relation between burden velocity and ratio of burden to blasthole diameter is developed in this paper.This relation is found to be consistent with the measured burden velocities of all 37 full-scale blasts found from published articles.These blasts include single-hole blasts,multi-hole blasts,and simultaneously-initiated blasts with various borehole diameters such as 64 mm,76 mm,92 mm,115 mm,142 mm and 310 mm.All boreholes were fully charged.The agreement between measured and calculated burden velocities demonstrates that this relation can be used to predict the burden velocity of a wide range of full-scale blast with fully-coupled explosive charge and help to determine a correct delay time between adjacent holes or rows in various full-scale blasts involved in tunnelling(or drifting),surface and underground mining production blasts and underground opening slot blasts.In addition,this theoretical relation is found to agree with the measured burden velocities of 9 laboratory small-scale blasts to a certain extent.To predict the burden velocity of a small-scale blast,a further study or modification to the relation is necessary by using more small-scale blasts in the future.Zong-Xian Zhang Li-Yuan Chi Changping Yi 2021Journal of Rock Mechanics and Geotechnical Engineering2021,13,4:2
