1Dynamic Expression of mi RNAs and Their Targets in the Response to Drought Stress of Grafted Cucumber Seedlings显示文摘Grafting of cucumber is widely used to improve growth and tolerance to biotic and abiotic stresses. Micro RNAs(miRNAs) regulate plant growth and development, and respond to various stresses through negative, post-transcriptional regulation of the expression of their target genes. We grafted cucumber(Cucumis sativus) scions onto pumpkin(Cucurbita moschata) rootstocks to study the molecular mechanisms of miRNA-mediated graftinginduced responses to drought stress. The relative expressions of 17 selected miRNAs and their predicted target m RNAs were detected by quantitative real-time PCR(q RT-PCR) in leaves and roots of hetero-grafted cucumber seedlings(cucumber as scion and pumpkin as rootstock) and autografted cucumber seedlings(cucumber as scion and rootstock) after 24 hours of 15% PEG6000 treatment. Compared with the expression in leaves of auto-grafted cucumber seedlings, the expressions of most miRNAs in the leaves of hetero-grafted seedlings changed dynamically: induced under normal conditions, and reduced after 3 h of drought stress, and then induced after 8 h and 24 h of drought stress. Similarly, compared with the expression in roots of auto-grafted cucumber seedlings, the expressions of most miRNAs in the roots of hetero-grafted cucumber seedlings changed dynamically: reduced under normal condition, and still reduced after 1 h, but induced after 3 h and 8 h, then reduced significant y after24 h of drought stress. 2Interference of CsGPA1,theα-submit of G protein,reduces drought tolerance in cucumber seedlings显示文摘The G proteinα-subunit,GPA1,is an integral component of several signaling pathways in plants,including response to abiotic stress.However,the molecular mechanism behind these processes remains largely unknown in the cucumber plant(Cucumis sativus L.).In order to further understand the role of CsGPA1 in cucumber under drought stress,changes in plant growth,physiological parameters,and gene expression of CsAQPs were all measured under water stress induced by polyethylene glycol(PEG)using wild type(WT)and CsGPA1-interference(RNAi)cucumber seedlings.Our results demonstrated that the RNAi plants had lower drought tolerance,displaying seriously withered leaves,lower relative growth rate,lower root-shoot ratio,and lower root activity under drought stress compared to WT plants.Physiological studies indicated that the suppression of CsGPA1 weakened drought stress tolerance due to higherwater loss rate in the leaves,higher levels of hydrogen peroxide(H2O2),increased malondialdehyde(MDA)content,lower free proline content,lower soluble sugar content,lower soluble protein content,and decreased antioxidant enzyme activities.qRT-PCR analysis demonstrated that the interference of CsGPA1 up-regulated the expression of most AQP genes(except for CsPIP2;3 in leaves)and down-regulated the expression of CsPIP1;2,CsPIP1;4,CsPIP2;1,and CsPIP2;4 in roots under drought stress when compared to WT plants.Our results demonstrated that CsGPA1 could function as a positive regulator in drought stress response by decreasing the accumulation of reactive oxygen species(ROS),improving permeable potentials,and reducing water loss in cucumber plants.Yumei Liu Qinghua Du Longqiang Bai Mintao Sun Yansu Li Chaoxing He Jun Wang Xianchang Yu Yan Yan 2021Horticultural Plant Journal2021,7,3:3

4Efficient Self-Assembled DNA Nanoparticles through Rolling Circle Amplification for siRNA Delivery in vitro显示文摘Effective and low toxicity delivery of siRNA is of great importance for clinical gene therapy. Herein, self-assembled DNA nanoparticles (NPs) based on rolling circle amplification (RCA) with a small interfering RNA (siRNA) payload were successfully developed as a facile and efficient siRNA delivery strategy. This intracellular gene silencing strategy exhibits various advantages including low toxicity, high efficiency, and good stability. The synthesized DNA NPs serve as siRNA carriers, protecting the siRNA against nuclease degradation. 19Multifaceted regulatory functions of CsBPC2 in cucumber under salt stress conditions显示文摘BASIC PENTACYSTEINE(BPC)transcription factors are essential regulators of plant growth and development.However,BPC functions and the related molecular mechanisms during cucumber(Cucumis sativus L.)responses to abiotic stresses,especially salt stress,remain unknown.We previously determined that salt stress induces CsBPC expression in cucumber.In this study,Csbpc2 transgene-free cucumber plants were created using a CRISPR/Cas9-mediated editing system to explore CsBPC functions associated with the salt stress response.The Csbpc2 mutants had a hypersensitive phenotype,with increased leaf chlorosis,decreased biomass,and increased malondialdehyde and electrolytic leakage levels under salt stress conditions.Additionally,a mutated CsBPC2 resulted in decreased proline and soluble sugar contents and antioxidant enzyme activities,which led to the accumulation of hydrogen peroxide and superoxide radicals.Furthermore,the mutation to CsBPC2 inhibited salinity-induced PM-H+-ATPase and V-H+-ATPase activities,resulting in decreased Na+efflux and increased K+efflux.These findings suggest that CsBPC2 may mediate plant salt stress resistance through its effects on osmoregulation,reactive oxygen species scavenging,and ion homeostasis-related regulatory pathways.However,CsBPC2 also affected ABA signaling.The mutation to CsBPC2 adversely affected salt-induced ABA biosynthesis and the expression of ABA signaling-related genes.Our results indicate that CsBPC2 may enhance the cucumber response to salt stress.It may also function as an important regulator of ABA biosynthesis and signal transduction.These findings will enrich our understanding of the biological functions of BPCs,especially their roles in abiotic stress responses,thereby providing the theoretical basis for improving crop salt tolerance.Shuzhen Li Mintao Sun Li Miao Qinghua Di Lijun Lv Xianchang Yu Yan Yan Chaoxing He Jun Wang Aokun Shi Yansu Li 2023Horticulture Research2023,10,5:0
