1RAPD Analysis of M_1 Generation of Gladiolus hybridus Hort Treated by EMS显示文摘Gladiolus hybridus Hort is one of the world's famous cutting flowers. It is very popular because of the big size, bright color, various shape, and long period of bloom. New species should be cultivated in order to meet the consumers' need of asking for the new. Among the technologies of cultivating new species of flowers, mutagenic breeding is a shortcut. This study treated corm bud of G. hybridus Hort with EMS of different consistency. Then M1 after treated was analyzed by RAPD. The result showed that EMS was a very effective mutagenic agent for the corm bud of G. hybridus Hort. With the increase of consistency, the mutagenic range increased first, then decreased, among which 0.6% EMS treatment had the biggest influence. 2Cloning and Analysis of Full-Length cDNA of PumNPR1 Gene from Pyrus ussuriensis Maxim显示文摘The purpose of this study is to find a new gene resource for the researches of molecular breeding of Rosaceae plants disease-resistance.Pyrus ussuriensis Maxim is used as a starting material to clone the full-length cDNA of NPR1(nonexpressor of pathogenesis-related genes 1) which is a key regulator in SA(salicylic acid)-mediated systemic acquired resistance(SAR) by homologous cloning and RACE techniques.The length of the cDNA sequence was 1 767 bp,the ORF was 1 761 bp,it coded 586 amino acids,pI=5.58,the relative molecular weight was 65.009 ku,contained 19 kinds of amino acids,and had full BTB/POZ and ANK domains.Compared the homology of NPR1 gene in GenBank database,the homology with Pyrus pyrifolia,Arabidopsis thaliana,Nicotiana tabacum,Lycopersicon esculentum,Oryza sativa,Helianthus annuus were 98%,62%,68%,65%,57%,63%.The homology of functional area were 99%,78%,82%,79%,74%,77%.This NPR1 gene was considered as homologic gene of Pyrus ussuriensis Maxim and named PumNPR1.CHE Daidi FAN Jinping WANG Jingang XU Ping YANG Tao LIU Shenkui 2008Journal of Northeast Agricultural University(English Edition)2008,15,2:2
3Research on protoplast preparation and culture in Gladiolus hybridus Hort显示文摘Protoplasts prepared from the aseptic leaves of Rose Supreme in Gladiolus hybridus Hort. were cultured. The yield of protoplasts with vigour was the highest under the following conditions: 1.5% cellulase Onzuka RS, 0.5% pectinase Y-23, 0.6 mol· L^(-1) mannitol, pH 5.8, digestion time 2 h, and temperature 27℃. After isolation and purification, protoplasts were cultured in solid and liquid combined medium, which included KM8P, 0.6 mol· L^(-1) mannitol, 1.0 mg· L^(-1) 2, 4-D, 0.2 mg· L^(-1) NAA and 0.2 mg· L^(-1) KT. 4Observation on the modifying activity of the protein from Elytrzgia repens rhizome for ice crystal显示文摘In winter,spring and summer,the rhizome of wild Elytrzgia repens of Heilongjiang Province was selected to extract the soluble which whole protein and the apoplastic protein,and analyzed by SDS-PAGE.The result indicated that there were two specific polypeptides in two types protein from winter;their relative molecular weight were identified as 52 ku and 26 ku by analyzing software:the apoplastic protein from winter had the ability of modifing the growth of ice crystal which appeared hexagonal in shape observed with the phase-contrast photomicroscope.So the apoplastic protein from winter has the antifreeze characters and the 52 ku protein is more likely the antifreeze protein.YANG Tao GONG Shufang LI Yan CHE Daidi 2007Journal of Northeast Agricultural University(English Edition)2007,14,3:1
5Preliminary Study on Anther in vitro Culture of Lily“Prato”显示文摘The effects of the microspore developmental stage,hormones and culture condition on anther in vitro culture of lily(Lilium spp.) were discussed.The results showed that when the flower buds were about 23-26 mm long,the microspores were at the uninucleate stage which was suitable for culture and the culture under the darkness would promote the callus induction of anther.The induction frequency could reach 42.5% in the optimized medium which was MS+[6-BA(0.5)+KT(2.0)+2,4-D(1.0)] mg·L-1.The rate of callus differentiation could reach 31.57% in the optimized medium which was MS+ NAA(1.5,2.0) mg·L-1.FAN Jinping PING Zhongyuan XU Enjun PAN Bingbing CHE Daidi 2011Journal of Northeast Agricultural University(English Edition)2011,18,1:1
6Somatic Embryogenesis in Lily Bulb Scale Cultures显示文摘Somatic embryogenesis from lily bulb scales has not been studied in details,although tissue culture methods have been applied to the propagation for decades.The effects of different kinds and concentration of auxins for oriental lily somatic embryogenesis were investigated(Lilium hybrida var.Sorbonne).2,4-dichlorophenoxyacetic acid(2,4-D),thidiazuron(TDZ)and α-naphthaleneacetic acid(NAA)media with benzyladenine(6-BA)and lactalbumin hydrolysate(LH)were used for embryogenic callus in the darkness.The best response on embryogenic callus formation was obtained on MS media supplemented 2,4-D 2.0 mg?L-1, 6-BA 0.5 mg·L-1 and LH 300 mg·L-1.Transfer embryogenic callus to the media with TDZ,6-BA,kinetin(KT)supplemented 2,4-D. The highest number of somatic embryos has been produced on medium with 0.5 mg·L-1 2,4-D and 0.3 mg?L-1 KT.Germinated embryos with shoot axes were changed to MS media with 6-BA 0.5 mg?L-1.The results suggest that in vitro culture of somatic embryogenesis from lily bulb scales can be used for plant regeneration.WANG Shasha WANG Jingang FAN Jinping CHE Daidi 2008Journal of Northeast Agricultural University(English Edition)2008,15,4:1
7ISSR Analysis of 26 General Species in Gladiolus hybridus Hort显示文摘ISSR analysis is applied in this study to research the classification and genetic relationship of 26 cultivars of G.hybridus Hort.Total 19 arbitrary primers screened from 100 primers were used for further PCR and diversity analysis.A total of 110 ISSR sites were detected with a mean of 5.63 fragments amplified for each primer.A total of 103 polymorphic DNA fragments were detected among all the 110 amplified fragments,which accounted for a high level of 93.6%of all and could be used to identify different cultivars.The results revealed that the idioplasm resource of G.hybridus Hort cutting flower cultivar had a narrow genetic basis on molecular level,and certain genetic relationship existed in summer large-flowers of G.hybridus Hort varieties.WANG Jingang GUO Ying CHE Daidi LIU Shenkui YANG Chuanpin 2008Journal of Northeast Agricultural University(English Edition)2008,15,4:0
8Study on Cell Suspension Culture of Floribunda Rose显示文摘Friable callus was induced when immature seeds of floribunda rose were inoculated on MS medium supplemented with 2,4-D 3.0 mg·L-1. When transfered onto subculture media, friable callus developed into embryogenic callus, which was used to establish cell suspension lines. Cell suspensions had to be subcultured at a interval of 4-5 days at the first several culture cycles. The best subculturing cycle for the stable cell suspensions was 8-10 days. 9RAPD Analysis of Twelve General Species of Gladiolus hybridus Hort.显示文摘RAPD analysis was applied in studying the classification and genetic relationship of 12 cultivars of Gladiolus hybridus Hort. Twenty-six arbitrary primers screened from 80 primers were used for further PCR and diversity analysis. A total of 164 RAPD sites were detected with a mean of 6.31 fragments amplified for each primer. A total of 151 polymorphic DNA fragments were detected among all the 164 amplified fragments, which accounted for a high level of 92.8% of all and could be used for identification of different cultivars. 10Study on Transformation of Oriental Hybrid Lily by Agrobacterium-mediated显示文摘An efficient procedure was described for the transformation of the monocotyledonous oriental hybrid lily,Lilium cv. Siberia. by Agrobacterium-mediated genetic transformation via leaves regeneration. The leaves of lea· ets which derived from bulbs were sliced into 1.0 cm long and were co-cultivated with A. tumefaciens strain LBA4404/pB2AE12,which harbored a vector carrying the neomycin phosphotransferase,DREB2A genes in the T-DNA region. The suitable genetic transformation condition was determined as follows:the bacterial concentration reached 0.5-0.6(OD600) ,15 min infection time,20 mg · L-1 acetosyingone,and 10.6 mmol · L-1 NH4NO3 medium was used for co-cultivation 3 days,delayed 7 days for selecting by 30 mg · L-1 kanamycin containing regeneration medium. Effi cient shoot regeneration was observed on MS medium supplemented with 1.0 mg · L-1 naphthaleneacetic acid,0.5 mg · L-1 benzyladenine and 0.1 mg · L-1 Kinetin after about 6 weeks culture.
