1Adoptive cell transfer therapy for hepatocellular carcinoma显示文摘Hepatocellular carcinoma (HCC) is one of the most common cancers worldwide. This malignancy is associated with poor prognosis and high mortality. Novel approaches for prolonging the overall survival of patients with advanced HCC are urgently needed. The antitumor activities of adoptive cell transfer therapy (ACT), such as strategies based on tumor-infiltrating lymphocytes and cytokine-induced killer cells, are more effective than those of traditional strategies. Currently, chimeric antigen receptor T-cell (CAR-T) immunotherapy has achieved numerous breakthroughs in the treatment of hematological malignancies, including relapsed or refractory lymphoblastic leukemia and refractory large B-cell lymphoma? Nevertheless, this approach only provides a modest benefit in the treatment of solid tumors. The clinical results of CAR-T immunotherapy for HCC that could be obtained at present are limited. Some published studies have demonstrated that CAR-T could inhibit tumor growth and cause severe side effects. 2Chimeric antigen receptor T cell targeting EGFRvIII for metastatic lung cancer therapy显示文摘Lung cancer is the most common incident cancer and the leading cause of cancer death. In recent years, the development of tumor immunotherapy especially chimeric antigen receptor T (CAR-T) cell has shown a promising future. Epidermal growth factor receptor variant III (EGFRvlll) is a tumor-specific mutation expressed in various types of tumors and has been detected in non-small cell lung cancer with a mutation rate of 10%. Thus, EGFRvIII is a potential antigen for targeted lung cancer therapy. In this study, CAR vectors were constructed and transfected into virus-packaging cells. Then, activated T cells were infected with retrovirus harvested from stable virus-producing single clone cell lines. CAR expression on the surfaces of the T cells was detected by flow cytometry and Western blot. The function of CAR-T targeting EGFRvIII was then evaluated. The EGFRvIII-CAR vector was successfully constructed and confirmed by DNA sequencing. A stable virus-producing cell line was produced from a single clone by limited dilution. The culture conditions for the cell line, including cell density, temperature, and culture medium were optimized. After infection with retrovirus, CAR was expressed on more than 90% of the T cells. The proliferation of CAR-T cells were induced by cytokine and specific antigen in vitro. More importantly, EGFRvIII-CART specifically and efficiently recognized and killed A549-EGFRvIII cells with an effector/target ratio of 10:1 by expressing and releasing cytokines, including perforin, granzyme B, IFN-γ, and TNF-α. The in vivo study indicated that the metastasis of A549-EGFRvIII cells in mice were inhibited by EGFRvIII-CART cells, and the survival of the mice was significantly prolonged with no serious side effects. EGFRvIII-CART showed significantly efficient antitumor activity against lung cancer cells expressing EGFRvlll in vivo and in vitro. 4Scientific objectives of the Hot Universe Baryon Surveyor(HUBS) mission显示文摘The Hot Universe Baryon Surveyor(HUBS) is a proposed space-based X-ray telescope for detecting X-ray emissions from the hot gas content in our universe. With its unprecedented spatially-resolved high-resolution spectroscopy and large field of view,the HUBS mission will be uniquely qualified to measure the physical and chemical properties of the hot gas in the interstellar medium, the circumgalactic medium, the intergalactic medium, and the intracluster medium. These measurements will be valuable for two key scientific goals of HUBS, namely to unravel the AGN and stellar feedback physics that governs the formation and evolution of galaxies, and to probe the baryon budget and multi-phase states from galactic to cosmological scales. In addition to these two goals, the HUBS mission will also help us solve some problems in the fields of galaxy clusters, AGNs, difuse X-ray backgrounds, supernova remnants, and compact objects. 6Artificial Macrophage with Hierarchical Nanostructure for Biomimetic Reconstruction of Antitumor Immunity显示文摘Artificial cells are constructed from synthetic materials to imitate the biological functions of natural cells.By virtue of nanoengineering techniques,artificial cells with designed biomimetic functions provide alternatives to natural cells,showing vast potential for biomedical applications.Especially in cancer treatment,the deficiency of immunoactive macrophages results in tumor progression and immune resistance.To overcome the limitation,a BaSO_(4)@ZIF-8/transferrin(TRF)nanomacrophage(NMΦ)is herein constructed as an alternative to immunoactive macrophages.Alike to natural immunoactive macrophages,NMΦis stably retained in tumors through the specific affinity of TRF to tumor cells.Zn^(2+)as an“artificial cytokine”is then released from the ZIF-8 layer of NMΦunder tumor microenvironment.Similar as proinflammatory cytokines,Zn^(2+)can trigger cell anoikis to expose tumor antigens,which are selectively captured by the BaSO_(4)cavities.Therefore,the hierarchical nanostructure of NMΦs allows them to mediate immunogenic death of tumor cells and subsequent antigen capture for T cell activation to fabricate long-term antitumor immunity.As a proof-of-concept,the NMΦmimics the biological functions of macrophage,including tumor residence,cytokine release,antigen capture and immune activation,which is hopeful to provide a paradigm for the design and biomedical applications of artificial cells.Henan Zhao Renyu Liu Liqiang Wang Feiying Tang Wansong Chen You‑Nian Liu 2023Nano-Micro Letters2023,15,12:0

8DISCOVERY OF TRIKETONE-QUINOXALINE HYBRIDS AS POTENT HPPD INHIBITORS USING STRUCTUREBASED DRUG DESIGN显示文摘p-Hydroxyphenylpyruvate dioxygenase(EC 1.13.11.27,HPPD)belongs to the family of Fe(II)-dependent non-heme oxygenases that occur in the majority of aerobic organisms.HPPD has proved to be a promising target in herbicide research and development.A battery of novel triketone-quinoxaline compounds has been designed using a structure-based drug design strategy and then prepared.Enzyme inhibition assays show that these synthesized derivatives possess favorable inhibition capability against Arabidopsis thaliana HPPD with IC50 values ranging from 0.317 to 0.891 mmol$L–1.Subsequently,the molecular docking results indicate that two adjacent carbonyls of the triketone moiety of the representative compound 2-(2,3-dimethyl-8-(o-tolyl)quinoxaline-6-carbonyl)-3-hydroxycyclohex-2-en-1-one(7d)engage in chelation with the ferrous ion of A.thaliana HPPD in a bidentate pose,and its quinoxaline scaffold forms two sets of parallelπ-stacking interaction between two phenylalanine residues(Phe424 and Phe381).In addition,the extended phenyl group also interacts with Phe392 in aπ-πstacking way.This study indicates that triketone-quinoxaline is a promising scaffold for discovering HPPD inhibitors with substantially increased potency,providing insight into the molecular design of new herbicides.Baifeng ZHENG Yaochao YAN Can FU Guangyi HUANG Long ZHAO Qiong CHEN Renyu QU Guangfu YANG 2022Frontiers of Agricultural Science and Engineering2022,9,1:0
9Sex-specific facilitation and reproduction of the gynodioecious cushion plant Arenaria polytrichoides on the Himalaya-Hengduan mountains,SW China显示文摘When benefiting other beneficiaries,cushion plants may reciprocally receive feedback effects.The feedback effects on different sex morphs,however,remains unclear.In this study,taking the gynodioecious Arenaria polytrichiodes as a model species,we aimed to assess the sex-specific facilitation intensity of cushion plant by measuring the beneficiary cover ratio,and to assess the potential costs in cushion reproductive functions by measuring the flower and fruit cover ratios.The total beneficiary cover ratio was similar between females and hermaphrodites.Females produced much less flowers but more fruits than hermaphrodites.These results suggested that females and hermaphrodites possess similar facilitation intensity,and female cushion A.polytrichoides may allocate more resources saved from pollen production to seed production,while hermaphrodites possibly allocate more resources to pollen production hence reducing seed production.The surface areas covered by beneficiaries produced less flowers and fruits than areas without beneficiaries.In addition,strong negative correlations between beneficiary cover and flower cover were detected for both females and hermaphrodites,but the correlation strength were similar for these two sex morphs.However,the correlation between beneficiary cover and fruit cover was only significantly negative for females,suggesting that beneficiary plants negatively affect fruit reproduction of females while have neutral effects on hermaphrodites.All the results suggest that to facilitate other beneficiaries can induce reproductive costs on cushion A.polytrichoides,with females possibly suffering greater cost than hermaphrodites.Such differentiation in reproductive costs between sex morphs,in long-term perspective,may imply sex imbalance in population dynamics.Xufang Chen Yazhou Zhang Lishen Qian Renyu Zhou Hang Sun Jianguo Chen 2024Plant Diversity2024,46,2:0
