1Equalization of Ti 6Al 4V alloy welded joint by scanning electron beam welding显示文摘The equalization of Ti 6Al 4V alloy welded joint with base metal on corrosion resistance, strength and ductility was studied. The solidification microstructure is transformed from 650 μm columnar grains to 100 μm equiaxed grains by scanning electron beam welding. The anodic polarization curve of 150 μm equiaxed grains coincides with that of base metal. Equal corrosion resistance between weld metal and base metal was obtained. Uniform microstructure and solute distribution are the basis of equalization. Corrosion rate of weld with 150 μm equiaxed grains is the lowest, 2.45 times lower than that of 650 μm columnar grains. 2Efficient transfer and expression of human clotting factor IX cDNA in neonatal hemophilia B mice mediated by VSV-G pseudotyped retrovi-rus显示文摘The feasibility of in vivo gene therapy for hemophilia B by VSV-G pseudotyped retroviral vector was introduced. The novel packaging cell line 293GPG was used to produce VSV-G/G1NaBAIX pseudotyped virus with the highest liters up to 8.5×108cfu .mL-1. In contrast to the conventional retrovirus, VSV-G pseudotyped virus was more resistant to inactivation by serum complements (P<0.001). Our results also demonstrated that VSV-G pseudotyped virus was more stable in neonatal mice serum than in adult mice serum (P<0.01). After intraperitoneal injection of different doses of virus, hFIX antigen was detected and lasted for more than 120 d, the highest level reached (72.5+6.1) ng- mL11. Moreover, the functional activity was improved to some extent in all hFIX-treated mice, the most remarkable improvement was observed in the mice treated with higher dose of virus whose clotting activity increased to (3.4±1.5)% and APTT (activated partial thromboplastin time) reduced to (43.2±7.2) s. 12Morphological structures of poly(vinylidene fluoride)/montmorillonite nanocomposites显示文摘Poly(vinylidene fluoride) (PVDF)/montmorillonite (MMT) nanocomposites were prepared by melt blen- ding a kind of organically modified montmorillonite with PVDF. The morphological structures of the nanocomposites were studied using X-ray diffraction (XRD), transmission electron microscopy (TEM), Fourier transform infrared (FTIR) spectroscopy, and differential scanning calorimetry (DSC). The re- sults indicate that organically modified montmorillonites are in the form of intercalation, exfoliation, and fragments in the PVDF matrix. For the composites, the (001) peak position of MMT was found to shift to a lower angle in XRD patterns, and some MMT fragments could be observed under TEM. MMT loading was favorable to producing the piezoelectric β phase in the PVDF matrix and caused internal stress in α crystals. At the same time, the crystallinity and spherulite size of PVDF decreased with the MMT content. MMT induced β phase is stable even at high temperatures (160℃). 13Prediction rule for estimating advanced colorectal neoplasm risk in average-risk populations in southern Jiangsu Province显示文摘Objective:The aim of this study was to establish the risk scoring system towards the advanced colorectal neoplasm(CN) risk in the average-risk populations in the southern Jiangsu Province,and to evaluate the screening efficacy.Methods:Totally 905 cases of the average-risk populations who received the colonoscopy were selected as the objective.The multivariate logistic regression analysis method was used to establish the scoring system towards the occurrence risk of the advanced tumor,and its screening efficacy was evaluated through the prediction consistency,distinguishing ability and screening accuracy.Results:The scoring system consisted of five variables,namely age,gender,coronary heart disease,egg intake and stool frequency.The results revealed that it had good prediction consistency(P=0.205) and distinguishing ability [the area under the receiver operating characteristic(ROC) curve was 0.75,with 95% confidence interval(95% CI) of 0.69-0.82].Thus,2.5 points was set as the screening cutoff value,and its sensitivity,specificity,accuracy,positive predictive value,negative predictive value,positive likelihood ratio and negative likelihood ratio were 93.8 %,47.6%,50.1%,9.1%,99.3%,1.79 and 0.13,respectively.Conclusions:The established scoring system had good screening efficacy,and can be used as the screening tool applying to the CN screening within the average-risk populations in the southern Jiangsu Province.Guochang Chen Boneng Mao Qi Pan Qian Liu Xinfang Xu Yueji Ning 2014Chinese Journal of Cancer Research2014,26,1:1

18Effects of lipid shell microbubble on ultrasound mediated EGFP gene delivery to transplanted tumors:initial experience显示文摘Objective: To investigate the feasibility of ultrasound (US) mediated enhanced green fluorescent protein (EGFP) gene delivery in subcutaneous transplanted tumors of human cervical carcinoma (Hela) and the contribution of lipid shell microbubble (LSMB) on gene transfection. Methods: LSMB and plasmid were injected into nude mice by tail vein followed local US irradiation (P + LSMB + US group). US exposure parameter was set at 2.0 W/cm2, 2 min, duty cycle 20%. EGFP expression was evaluated by imaging for 7 days. Nude mice undergoing plasmid injection alone (P group), plasmid injection and US exposure (P + US group), plasmid and LSMB injection (P + LSMB group) were used as controls. Frozen section and histological examinations were conducted. Expression of EGFP was scored. Kinetics of protein expression post transfection and localization in vivo were evaluated. Results: Plasmid injection with LSMB plus US exposure strongly increased gene transfer efficiency. Strong EGFP expression was mainly seen in LSMB + P + US group. It was significantly higher than any of the following groups, P group, US + P group, or LSMB + P group (P < 0.01). In vivo expression level of post-US 3 days was significantly higher than any other time points (P < 0.01). There was not significant expression level of EGFP in other organs or tissues regardless of US exposure. No tissue damage was seen histologically. Conclusion: The combination of LSMB and US exposure could effectively transfer plasmid DNA to transplanted tumors without causing any apparently adverse effect. LSMB could be effective as a non-viral vector system in in vivo gene delivery.
