| 1 | A structural view of the antibiotic degradation enzyme NDM-1 from a superbug显示文摘Gram-negative Enterobacteriaceae with resistance to carbapenem conferred by New Delhi metallo-β-lactamase 1(NDM-1)are a type of newly discovered antibioticresistant bacteria.The rapid pandemic spread of NDM-1 bacteria worldwide(spreading to India,Pakistan,Europe,America,and Chinese Taiwan)in less than 2 months characterizes these microbes as a potentially major global health problem.The drug resistance of NDM-1 bacteria is largely due to plasmids containing the blaNDM-1 gene shuttling through bacterial populations.The NDM-1 enzyme encoded by the blaNDM-1 gene hydrolyzes β-lactam antibiotics,allowing the bacteria to escape the action of antibiotics.Although the biological functions and structural features of NDM-1 have been proposed according to results from functional and structural investigation of its homologues,the precise molecular characteristics and mechanism of action of NDM-1 have not been clarified.Here,we report the threedimensional structure of NDM-1 with two catalytic zinc ions in its active site.Biological and mass spectroscopy results revealed that D-captopril can effectively inhibit the enzymatic activity of NDM-1 by binding to its active site with high binding affinity.The unique features concerning the primary sequence and structural conformation of the active site distinguish NDM-1 from other reported metallo-β-lactamases(MBLs)and implicate its role in wide spectrum drug resistance.We also discuss the molecular mechanism of NDM-1 action and its essential role in the pandemic of drug-resistant NDM-1 bacteria.Our results will provide helpful information for future drug discovery targeting drug resistance caused by NDM-1 and related metallo-β-lactamases. | Yu Guo Jing Wang Guojun Niu Wenqing Shui Yuna Sun Honggang Zhou Yaozhou Zhang Cheng Yang Zhiyong Lou Zihe Rao | 2011 | Protein & Cell2011,2,5: | 16 |
| 2 | Deep structure at northern margin of Tarim Basin显示文摘In this paper, a 2D velocity structure of the crust and the upper mantle of the northern margin of the Tarim Basin (TB) has been obtained by ray tracing and theoretical seismogram calculation under the condition of 2D lateral inhomogeneous medium using the data of seismic wide angle reflection/refraction profile from Baicheng to Da Qaidam crossing the Kuqa Depression (KD) and Tabei Uplift (TU). And along the Baicheng to Da Qaidam profile, 4 of the 10 shot points are located in the northern margin of the TB. The results show that the character of the crust is uniform on the whole between the KD and TU, but the depth of the layers, thickness of the crust and the velocity obviously vary along the profile. Thereinto, the variation of the crust thickness mainly occurs in the middle and lower crust. The Moho has an uplifting trend near the Baicheng shot point in KD and Luntai shot point in TU, and the thickness of the crust reduces to 42 km and 47 km in these two areas, respectively. The transition zone between the KD and TU has a thickest crust, up to 52 km. In this transition zone, there are high velocity anoma-lies in the upper crust, and low velocity anomalies in the lower crust, these velocity anomalies zone is near vertical, and the sediment above them is thicker than the other areas. According to the velocity distributions, the profile can be divided into three sections:KD, TU and transition zone between them. Each section has a special velocity structural feature, the form of the crystalline basement and the relationship between the deep structure and the shallow one. | 3 | Surface curvature-confined strategy to ultrasmall nickel-molybdenum sulfide nanoflakes for highly efficient deep hydrodesulfurization显示文摘Size-controlled synthesis of two-dimensional(2D)catalysts with low stacking numbers and small nanoflake lengths is crucial for promoting the catalytic performance in diverse heterogeneous catalysis.Herein,we report a facile and general“surface curvature-confined synthesis”strategy to modulate the slab lengths and stacking numbers of 2D transition metal sulfides by controlling the strain induced by different surface curvature of supports.An efficient NiMo sulfide with shorter slab length(average 3.71 nm),less stacking number(1–2 layers)and more edge active sites is synthesized onto ZSM-5 zeolites with the average size of 100 nm,which shows superior kHDS value of dibenzothiophene(14.05×10^−7 mol/(g·s)),enhanced stability up to 80 h,and high direct desulfurization selectivity(>95%).This design concept is also proved to be generally applicable to modulate the slab lengths and stacking numbers of other 2D catalysts such as MoS2 and WS2 nanoflakes,which shows great potentials for developing more ultrasmall 2D catalysts with controlled sizes and excellent catalytic activities. | Xin Kang Jiancong Liu Chungui Tian Dongxu Wang Yaorui Li Hongyan Zhang Xusheng Cheng Aiping Wu Honggang Fu | 2020 | Nano Research2020,13,3: | 5 |
| 4 | Stratigraphic Sequence and Sedimentary Systems in the Middle-Southern Continental Slope of the East China Sea from Seismic Reflection Data: Exploration Prospects of Gas Hydrate显示文摘Many evidences for gas hydrate bearing sediments had been found in the continental slope of the East China Sea,such as bottom simulating reflections(BSRs),undersea gas springs,pyrite associated with methane leakage,mud diapirs/mud volcanos,bottom-water methane anomalies and so on.In this study,six key stratigraphic interfaces including T_0(seafloor),T_1(LGM,23 kyr B.P.),T_2(2.58 Myr),T_3(5.33 Myr),T_4(11.02 Myr)and T_5(16.12 Myr)were identified,and then five third-order sequences of SQIII1 to SQIII5 were divided.However,T5 in southern continental slope is not found,which shows that the middle-northern Okinawa Trough had begun to rift in the early Miocene,earlier than the southern segment.Four system tracts including lowstand systems tract(LST),transgressive systems tract(TST),highstand systems tract(HST)and falling stage systems tract(FSST)are further divided.The marine erosion interface of 11.02 Myr and regressive unconformity interface of 23 kyr B.P.indicate two large-scale sea level drop events in the research area.Seven typical seismic facies identified in the continental slope are continental shelf-edge deltas,littoral fluvial-delta plains,incised channels or submarine canyons,slope fans,submarine fans or coastal sandbars,littoral-neritic finegrained sediments,mud volcanos and some other geological bodies respectively.The minimum water depth for hydrate occurrence in the Okinawa Trough is 630 m,and the thickness of gas hydrate stability zone in continental slope is between 0 and 590 m.The calculated bottom boundary of hydrate stability zone is slightly deeper than BSRs on the seismic sections.The re-depositional turbidite sand bodies,such as canyon channels,slope fans and submarine fans developed in Quaternary strata,are the predominant hydrate reservoirs.According to developing process,the dynamic accumulation of hydrate systems can be divided into three evolutionary stages including canyon erosion and hydrate stability zone migration stage,sediments destabilizing and methane leakage stage,and channel filling and hydrate re-occurrence stage. | LI Deyong CHEN Hongyan XU Shujuan XING Junhui CHENG Honggang WANG Jinkai | 2019 | Journal of Ocean University of China2019,18,6: | 1 |

| 9 | Hydrocarbon accumulation characteristics in basement reservoirs and exploration targets of deep basement reservoirs in onshore China显示文摘Based on the global basement reservoir database and the dissection of basement reservoirs in China,the characteristics of hydrocarbon accumulation in basement reservoirs are analyzed,and the favorable conditions for hydrocarbon accumulation in deep basement reservoirs are investigated to highlight the exploration targets.The discovered basement reservoirs worldwide are mainly buried in the Archean and Precambrian granitic and metamorphic formations with depths less than 4500 m,and the relatively large reservoirs have been found in rift,back-arc and foreland basins in tectonic active zones of the Meso-Cenozoic plates.The hydrocarbon accumulation in basement reservoirs exhibits the characteristics in three aspects.First,the porous-fractured reservoirs with low porosity and ultra-low permeability are dominant,where extensive hydrocarbon accumulation occurred during the weathering denudation and later tectonic reworking of the basin basement.High resistance to compaction allows the physical properties of these highly heterogeneous reservoirs to be independent of the buried depth.Second,the hydrocarbons were sourced from the formations outside the basement.The source-reservoir assemblages are divided into contacted source rock-basement and separated source rock-basement patterns.Third,the abnormal high pressure in the source rock and the normal–low pressure in the basement reservoirs cause a large pressure difference between the source rock and the reservoirs,which is conducive to the pumping effect of hydrocarbons in the deep basement.The deep basement prospects are mainly evaluated by the factors such as tectonic activity of basement,source-reservoir combination,development of large deep faults(especially strike-slip faults),and regional seals.The Precambrian crystalline basements at the margin of the intracontinental rifts in cratonic basins,as well as the Paleozoic folded basements and the Meso-Cenozoic fault-block basements adjacent to the hydrocarbon generation depressions,have favorable conditions for hydrocarbon accumulation,and thus they are considered as the main targets for future exploration of deep basement reservoirs. | WANG Zecheng JIANG Qingchun WANG Jufeng LONG Guohui CHENG Honggang SHI Yizuo SUN Qisen JIANG Hua ABULIMITI Yiming CAO Zhenglin XU Yang LU Jiamin HUANG Linjun | 2024 | Petroleum Exploration and Development2024,51,1: | 0 |
