1Menthol-modified casein nanoparticles loading 10-hydroxycamptothecin for glioma targeting therapy显示文摘Chemotherapy outcomes for the treatment of glioma remains unsatisfactory due to the inefficient drug transport across the blood–brain barrier(BBB) and insufficient drug accumulation in the tumor region. Although many approaches, including various nanosystems, have been developed to promote the distribution of chemotherapeutics in the brain tumor, the delivery efficiency and the possible damage to the normal brain function still greatly restrict the clinical application of the nanocarriers.Therefore, it is urgent and necessary to discover more safe and effective BBB penetration and gliomatargeting strategies. In the present study, menthol, one of the strongest BBB penetration enhancers screened from traditional Chinese medicine, was conjugated to casein, a natural food protein with brain targeting capability. Then the conjugate self-assembled into the nanoparticles to load anti-cancer drugs.The nanoparticles were characterized to have appropriate size, spheroid shape and high loading drug capacity. Tumor spheroid penetration experiments demonstrated that penetration ability of mentholmodified casein nanoparticles(M-CA-NP) into the tumor were much deeper than that of unmodified nanoparticles. In vivo imaging further verified that M-CA-NPs exhibited higher brain tumor distribution than unmodified nanoparticles. The median survival time of glioma-bearing mice treated with HCPT-MCA-NPs was significantly prolonged than those treated with free HCPT or HCPT-CA-NPs. HE staining ofthe organs indicated the safety of the nanoparticles. 2Stratum energy of coal-bed gas reservoir and their control on the coal-bed gas reservoir formation显示文摘Stratum energy of coal-bed gas reservoir, including coal-radix flexibility energy, groundwater flexibility energy and gas flexibility energy (hereinafter 'three energy'), depends on the energy homeostasis system, the core process of which is the effective transfer of energy and the geological selective process. Combining with the mechanics experimentations of coal samples, different flexibility energy has been analyzed and researched quantificationally, and a profound discussion to their controls on the coal-bed gas reservoir formation has been made. It is shown that when gas reservoir is surrounded by edge water and bottom water, the deposited energy in the early phase of forming gas reservoir is mostly coal-radix and gas flexibility energy, but the effect of groundwater flexibility energy increases while water-body increases. The deposited energy in the middle and later phase of forming gas reser voir is mostly gas flexibility energy, which is greater than 80% of all deposited energy. In the whole process, larger groundwater body exerts greater influences on gas accumulation. The paper indicated that higher stratum energy is more propitious to forming coal-bed gas reservoir. And higher coal-radix flexibility energy and gas flexibility energy are more propitious to higher yield of gas reservoirs, while higher groundwater flexibility energy is more propitious to stable yield of gas reservoirs. 3Self-adjusted elastic action and its CBM pool-forming effect of the high rank coal reservoir显示文摘The coal reservoir is a kind of geologic body with considerable elasticity, and its response to stress is more sensitive than the routine ones. In the pool-forming process of the coalbed methane, (CBM) the action of many external geodynamic factors in the coal reservoir is represented by the ability of CBM’s diffusion and migration in the coal reservoir. Therefore, the occurrence of the natural fissures in the coal reservoir becomes a key that determines whether the CBM pool can be formed or not. Based on the principle, the authors have designed a comprehensive method of physical modeling to study the elasticity of the coal matrix block with different coal ranks in the light of the solid to fluid coupling, have established the model of the volume deformation (self-adjusted elastic effect) of the coal reservoir under the condition of the effective stress-adsorption/desorption, and have obtained a correct understanding of the law of the elastic deformation. 5Single Nucleotide Polymorphism of FSHβ Gene Associated with Reproductive Traits in Japanese Flounder (Paralichthys olivaceus)显示文摘Follicle stimulating hormone β (FSHβ) of Japanese flounder (Paralichthys olivaceus) plays a key role in the regulation of gonadal development.This study aimed to investigate molecular genetic characteristics of the FSHβ gene and elucidate the effects of single nucleotide polymorphisms (SNPs) of FSHβ on reproductive traits in Japanese flounder.We used polymerase chain reaction single-strand conformation polymorphism (PCR-SSCP) and sequencing of the FSHβ gene in 60 individuals.We identified only an SNP (T/C) in the coding region of exon3 of FSHβ.The SNP (T/C) did not lead to amino acid changes at the position 340 bp of FSHβ gene.Statistical analysis showed that the SNP was significantly associated with testosterone (T) level and gonadosomatic index (GSI) (P<0.05).Individuals with genotype TC of the SNP had significantly higher serum T levels and GSI (P<0.05) than that of genotype CC.Therefore, FSHβ gene could be a useful molecular marker in selection for prominent reproductive trait in Japanese Flounder.HE Feng WEN Haishen YU Dahui LI Jifang SHI Bao CHEN Caifang ZHANG Jiaren JIN Guoxiong CHEN Xiaoyan Shi Dan Yang Yanping 2010Journal of Ocean University of China2010,9,4:4
6Surface modification of PGP for a neutrophil–nanoparticle co-vehicle to enhance the anti-depressant effect of baicalein显示文摘Exploiting cells as vehicles combined with nanoparticles combined with therapy has attracted increasing attention in the world recently. Red blood cells, leukocytes and stem cells have been used for tumor immunotherapy, tissue regeneration and inflammatory disorders, and it is known that neutrophils can accumulate in brain lesions in many brain diseases including depression. N-Acetyl Pro–Gly–Pro(PGP) peptide shows high specific binding affinity to neutrophils through the CXCR2 receptor. In this study, PGP was used to modify baicalein-loaded solid lipid nanoparticles(PGP-SLNs) to facilitate binding to neutrophils in vivo. Brain-targeted delivery to the basolateral amygdala(BLA) was demonstrated by enhanced concentration of baicalein in the BLA. An enhanced anti-depressant effect was observed in vitro and in vivo. The mechanism involved inhibition of apoptosis and a decrease in lactate dehydrogenase release. Behavioral evaluation carried out with rats demonstrated that anti-depression outcomes were achieved. The results indicate that PGP-SLNs decrease immobility time, increase swimming time and climbing time and attenuate locomotion in olfactory-bulbectomized(OB) rats. 9Interlayer interference mechanism of multi-seam drainage in a CBM well:An example from Zhucang syncline显示文摘Based on the characteristics of the strong volatility of physical property in vertical direction, high gas content, high resource abundance and large exploitation potentiality of coal reservoir in Bide-Santang basin of Zhina coal field, we study the generation mechanism of interlayer interference, propagation rules of reservoir pressure drop and influencing factors of gas productivity in CBM multi-seam drainage in the paper. On the basis of the actual production data of X-2 well of Zhucang syncline in Bide-Santang basin,by simulating the gas production process of a CBM well under the condition of multiple seam with COMET3 numerical simulation software, we analyze the influencing factors of gas productivity during the process of multi-seam drainage, and illuminate the interlayer interference mechanism of multiseam drainage. The results show that permeability, reservoir pressure gradient, critical desorption pressure and fluid supply capacity of stratum have great influence on gas productivity of multi-seam drainage while coal thickness has little influence on it. Permeability, reservoir pressure gradient and fluid supply capacity of stratum affect the propagation velocity of reservoir pressure drop and thereby affect the final gas productivity. 10Uterine artery embolization in association with methotrexate infusion for the treatment of tubal ectopic pregnancy显示文摘Objective To investigate the safety, feasibility, and effectiveness of uterine artery embolization in association with methotrexate(MTX) infusion for the treatment of tubal ectopic pregnancy. Methods Fifty-one patients with tubal ectopic pregnancy were referred for interventional management. All patients received super-selective arteriography of the uterine artery, were infused with 50–100 mg methotrexate(MTX) through a catheter, and underwent embolization of the uterine artery with a gel-foam pledge. Clinical presentation, findings of physical examination, β-HCG values, and the size of the ectopic mass were documented for comparison. The concentration of MTX in blood was evaluated at 0.5, 6, 12, 24, 36, and 48 hours after the procedure. Results Forty-seven out of the 51 patients had clinical resolution of their tubal pregnancy(92.2%). The average time for the β-HCG value to decrease and come back to normal was 9.16 ± 2.54 days(mean +/-SD). MTX levels in peripheral blood could not be detected for patients who received 50 or 75 mg MTX at 36 hours after the procedure, while the MTX level was 0.01 μmol/L at 48 hours after the procedure for patients who received 100 mg. Out of the 4 cases whose ectopic mass size was ≥5 cm, 3 failed to respond to the treatment; however, those whose ectopic mass size was ≤5 cm responded positively to the treatment, regardless of the β-HCG concentration and abdominal bleeding, except for 1 patient who had to undergo laparoscopy for severe abdominal pain and who showed a reduction in her β-HCG level. Conclusion Uterine artery embolization in association with methotrexate infusion is safe and effective in the treatment of tubal ectopic pregnancy, especially for those women with mild to moderate bleeding, or for those at risk of a major hemorrhage. 11A polyphenol-assisted IL-10 mRNA delivery system for ulcerative colitis显示文摘With the development of synthesis technology, modified messenger RNA(mRNA) has emerged as a novel category of therapeutic agents for a broad of diseases. However, effective intracellular delivery of mRNA remains challenging, especially for its sensitivity to enzymatic degradation. Here, we propose a polyphenol-assisted handy delivery strategy for efficient in vivo delivery of IL-10 mRNA.IL-10 mRNA binds to polyphenol ellagic acid through supramolecular binding to yield a negatively charged core, followed by complexing with linear polyetherimide and coating with bilirubin-modified hyaluronic acid to obtain a layer-by-layer nanostructure. The nanostructure specifically up-regulated the level of IL-10, effectively inhibited the expression of inflammatory factors, promoted mucosal repair, protected colonic epithelial cells against apoptosis, and exerted potent therapeutic efficacy in dextran sulfate sodium salt-induced acute and chronic murine models of colitis. 12Biodegradable magnesium alloy with eddy thermal effect for effective and accurate magnetic hyperthermia ablation of tumors显示文摘Magnetic hyperthermia therapy(MHT)is able to ablate tumors using an alternating magnetic field(AMF)to heat up magnetocaloric agents(e.g.magnetic nanoparticles)administered into the tumors.For clinical applications,there is still a demand to find new magnetocaloric agents with strong AMF-induced heating performance and excellent biocompatibility.As a kind of biocompatible and biodegradable material,magnesium(Mg)and its alloys have been extensively used in the clinic as an implant metal.Herein,we discovered that the eddy thermal effect of the magnesium alloy(MgA)could be employed for MHT to effectively ablate tumors.Under low-field-intensity AMFs,Mg A rods could be rapidly heated,resulting in a temperature increase in nearby tissues.Such AMF-induced eddy thermal heating of MgA could not only be used to kill tumor cells in vitro,but also be employed for effective and accurate ablation of tumors in vivo.In addition to killing tumors in mice,we further demonstrated that VX2 tumors of much larger sizes growing in rabbits after implantation of Mg A rods could also be eliminated after exposure to an AMF,illustrating the ability of MgA-based MHT to kill large-sized tumors.Moreover,the implanted MgA rods showed excellent biocompatibility and~20%of their mass was degraded within three months.Our work thus discovered for the first time that non-magnetic biodegradable Mg A,an extensively used implant metal in clinic,could be used for effective magnetic thermal ablation of tumors under a low-field-intensity AMF.Such a strategy could be readily translated into clinical use.Nailin Yang Fei Gong Liang Cheng Huali Lei Wei Li Zongbin Sun Caifang Ni Zhanhui Wang Zhuang Liu 2021National Science Review2021,8,1:1
13Effective migration system of coalbed methane reservoirs in the southern Qinshui Basin显示文摘Effective migration system of coalbed methane(CBM)reservoir,which was controlled by development degree and opening-closing degree of fractures,determines the permeability of coal reservoir and can be characterized by the pore-fracture system in the extrinsic form.In this paper,based on coal matrix elastic self-regulating effect theory and coal reservoir combined elastic energy theory,the fracture opening-closing degree parameterΔand the fracture development degree parameterξare suggested for the quantitative study of the effective migration system of CBM reservoir in southern Qinshui Basin.Further,the control functions ofξandΔto CBM enrichment and high production are discussed.The results show that in present stage the area with highξvalue is located in Anze and Qinyuan,and then Zhengzhuang and Fangzhuang,where fracture development degree is high.The area with highΔvalue is located in Zhengzhuang and Fanzhuang,and then Anze and Qinyuan,indicating where coal matrix elastic self-regulating positive effect dominates and fractures tend to be open.Through the comprehensive analysis onξandΔ,it can be found that their best match area is located in Zhengzhuang and Fanzhuang,with high values for fracture development degree and opening-closing degree probably bringing about high fluid pressure and good permeability of reservoirs,which are advantageous to an abundant CBM production.WU CaiFang QIN Yong ZHOU LongGang 2014Science China Earth Sciences2014,57,12:1

17NMR studies of stock process water and reaction pathways in hydrothermal carbonization of furfural residue显示文摘Hydrothermal carbonization(HTC) is a valuable approach to convert furfural residue(FR) into carbon material. The prepared biochars are usually characterized comprehensively, while the stock process water still remains to be studied in detail. Herein, a NMR study of the main components in stock process water generated at different HTC reaction conditions was reported. Various qualitative and quantitative NMR techniques(~1H and ^(13)C NMR,~1H-~1H COSY and ~1H-^(13)C HSQC etc.) especially 1D selective gradient total correlation spectroscopy(TOCSY NMR) were strategically applied in the analysis of HTC stock process water. Without separation and purification, it was demonstrated that the main detectable compounds are 5-hydroxymethylfurfural, formic acid, methanol, acetic acid, levulinic acid, glycerol, hydroxyacetone and acetaldehyde in this complicate mixture. Furthermore, the relationship between the concentration of major products and the reaction conditions(180-240 ℃ at 8 h, and 1-24 h at 240 ℃) was established. Finally, reasonable reaction pathways for hydrothermal conversion of FR were proposed based on this result and our previously obtained characteristics of biochars.
