| 4 | Metabolic risk factors associated with sudden cardiac death(SCD)during acute myocardial ischemia显示文摘Sudden cardiac death(SCD)is the leading cause of death worldwide.Myocardial ischemia(MI)is the most common underlying causal disorder for SCD.Metabolic risks leading to SCD during acute MI are still not fully understood.Here,using tissue metabolomics,we aimed to investigate myocardial metabolic alterations relevant to SCD events in an acute MI rat model induced by coronary artery ligation(CAL).Thirty-four rats were successfully performed CAL,of which 13 developed lethal ventricular tachyarrhythmia(LVTA)-SCD and 7 developed severe atrioventricular block(AB)-SCD.Fourteen rats that survived within 70 min after the ligation were served as peer controls.The partial least squares-discriminant analysis plots demonstrated clear separations between the SCD rats and controls,indicating obvious differences in myocardial metabolome between these rats.The levels of isoleucine,lactate,glutamate choline,phosphorylcholine,taurine and asparagine in ischemic myocardia were positively associated with LVTA-SCD events;in contrast,the levels of alanine,urea,phenylalanine,linoleic acid,elaidic acid and stearic acid were inversely correlated with LVTA-SCD events.The levels of glutamate and urea were positively and negatively relevant to AB-SCD events,respectively.The dangerous metabolites indicated that lower levels of energy substrates,severe hypoxia,the inhibition of transamination and hyper sympathetic excitement and reactive oxygen species in myocardia were vulnerable to SCD during acute MI.The results suggest fatal metabolic alterations correlated with SCD events during acute MI,which could offer novel clues for the prevention or treatment of acute MI-related SCD. | Dian Wang Xingxing Wang Jiayan Wu Ruibing Su Jing Kong Xiaojun Yu | 2017 | Forensic Sciences Research2017,2,3: | 2 |

However, effective intracellular delivery of mRNA remains challenging, especially for its sensitivity to enzymatic degradation. Here, we propose a polyphenol-assisted handy delivery strategy for efficient in vivo delivery of IL-10 mRNA.IL-10 mRNA binds to polyphenol ellagic acid through supramolecular binding to yield a negatively charged core, followed by complexing with linear polyetherimide and coating with bilirubin-modified hyaluronic acid to obtain a layer-by-layer nanostructure. The nanostructure specifically up-regulated the level of IL-10, effectively inhibited the expression of inflammatory factors, promoted mucosal repair, protected colonic epithelial cells against apoptosis, and exerted potent therapeutic efficacy in dextran sulfate sodium salt-induced acute and chronic murine models of colitis. To measure the antiinterference performance of the electronic communication system in the complicated electromagnetic interference environment, a method of multi-dimensional and complicated electromagnetic interference hardware-in-the-loop simulation in an anechoic room is proposed. It takes into account the characteristics of interference signals and the positional relationship among interference, the receiver and the transmitter of the electronic communication system.It uses the grey relational method and the angular domain mapping error correction method to control the relevant parameters,the microwave switch and so on, thus achieving the approximately actual mapping of the outdoor multi-dimensional and complicated electromagnetic interference in the anechoic room. To verify the effectiveness of this method, the multi-dimensional and complicated electromagnetic interference of the UAV's data link is simulated as an example. | 15 | Reviving chloroquine for anti-SARS-CoV-2 treatment with cucurbit[7]uril-based supramolecular formulation显示文摘The wide-spreading SARS-CoV-2 virus has put the world into boiling water for more than a year,however pharmacological therapies to act effectively against coronavirus disease 2019(COVID-19)remain elusive.Chloroquine(CQ),an antimalarial drug,was found to exhibit promising antiviral activity in vitro and in vivo at a high dosage,thus CQ was approved by the FDA for the emergency use authorization(EUA)in the fight against COVID-19 in the US,but later was revoked the EUA status due to the severe clinical toxicity.Herein,we show that supramolecular formulation of CQ by a macrocyclic host,curcurbit[7]uril(CB[7]),reduced its non-specific toxicity and improved its antiviral activity against coronavirus,working in synergy with CB[7].CB[7]was found to form 1:1 host-guest complexes with CQ,with a binding constant of$104 L/mol.The CQ-CB[7]formulation decreased the cytotoxicity of CQ against Vero E6 and L-02 cell lines.In particular,the cytotoxicity of CQ(60 mmol/L)against both Vero E6 cell line and L-02 cell lines was completely inhibited in the presence of 300 mmol/L and 600 mmol/L CB[7],respectively.Furthermore,the CB[7]alone showed astonishing antiviral activity in SARS-CoV-2 infected Vero E6 cells and mouse hepatitis virus strain A59(MHV-A59)infected N2 A cells,and synergistically improved the antiviral activity of CQ-CB[7],suggesting that CB[7]-based CQ formulation has a great potential as a safe and effective antiviral agent against SARS-CoV-2 and other coronavirus. | Cheryl H.T.Kwong Jingfang Mu Shengke Li Yaohui Fang Qianyun Liu Xiangjun Zhang Hiotong Kam Simon M.Y.Lee Yu Chen Fei Deng Xi Zhou Ruibing Wang | 2021 | Chinese Chemical Letters2021,32,10: | 1 |
