| 1 | Repurposing vitamin D for treatment of human malignancies via targeting tumor microenvironment显示文摘Tumor cells along with a small proportion of cancer stem cells exist in a stromal microenvironment consisting of vasculature, cancer-associated fibroblasts, immune cells and extracellular components.Recent epidemiological and clinical studies strongly support that vitamin D supplementation is associated with reduced cancer risk and favorable prognosis. Experimental results suggest that vitamin D not only suppresses cancer cells, but also regulates tumor microenvironment to facilitate tumor repression. In this review, we have outlined the current knowledge on epidemiological studies and clinical trials of vitamin D. Notably, wesummarized and discussed the anticancer action of vitamin D in cancer cells, cancer stem cells and stroma cells in tumor microenvironment, providing a better understanding of the role of vitamin D in cancer. | 2 | Metals in water and surface sediments from Henan reaches of the Yellow River,China显示文摘The concentrations of Cd,Cr,Cu,Ni,Pb and Zn were determined in the water and surface sediments from the Henan reaches of the Yellow River.Twenty-three sampling sites along the Yellow River and its tributaries were selected.Generally,metal concentrations were found to decrease in sequences of Zn>Cu>Pb>Cr>Ni>Cd in water and Zn>Cr>Pb>Ni>Cu>Cd in sediments.High levels of metal concentration were determined at a few stations of the river and its tributaries,such as Yiluo River,Si River and Qin River.The pollution of the Yellow River by Cd,Cr,Cu,Ni,Pb and Zn can be regarded as much higher compared to the background values,US EPA criteria(1999) and China water quality criteria(2002) .For sediments,metal lev-els except Pb did not significantly exceed the average shale levels and backgrounds in several countries including China.Data analysis manifests that positive correlations were found between Cu,Ni and Zn in water,and Pb,Ni,Zn and Cr in sediments. The Pearson correlation coefficient analysis and Cluster analysis were provided to assess the possible contamination sources. | 3 | Numerical study of hydrofoil surface jet flow on cavitation suppression显示文摘Cavitation caused vibration and noise of hydraulic machinery. To some extent,cavitation made fatigue damage in advance. Many scholars found that the re-entrant jets were the reasons of the shedding of cavities. To suppress cavitation,based on the idea of blocking the re-entrant jets,a special surface flow structure of 2D hydrofoil was proposed. The through-hole was made in the proper position of the hydrofoil. The incoming flow can outflow from this jet-hole automatically depending on the pressure difference between pressure side and suction side. Re-entrant jet growth can be weakened by optimizing the jet-hole geometry. Based on the standard k-ε turbulence model and Schnerr & Sauer cavitation model,under different cavitation numbers( σ) and jet-angles( β) for NACA0066( 2D) hydrofoil with 8° angles of attack,cavitation field numerical analysis was carried out. The results show that 2D hydrofoil cavitation flow had a strong unsteadiness. Making a jet-hole at the junction between the re-entrant jet and cavity can effectively minimize cloud cavitation. For a certain cavitation condition,optimal jet-angles( β) can be obtained to control cavitation growth. For the same β,the effects of cavitation suppression were changed with different cavitation numbers( σ). | 4 | Joints of continuous carbon fiber reinforced lithium aluminosilicate glass ceramics matrix composites to Ti60 alloy brazed using Ti-Zr-Ni-Cu active alloy显示文摘Continuous carbon fiber reinforced lithium aluminosilicate glass ceramics matrix composites(C_f/LAS composites) are joined to Ti60 alloy vacuum brazed using Ti-Zr-Ni-Cu brazing alloy. The effects of the brazing temperature on the interfacial microstructure and mechanical properties of brazed joints are investigated in details. The interfacial microstructure varies apparently with an increase of the brazing temperature. The thicknesses of the banded Ti solid solution(Ti(s, s)) and the reactive layer between Cf/LAS composites and the interlayer grow gradually. The mechanical properties of brazed joints increase firstly and then decrease with an increasing temperature. In addition, a joint that is brazed at 980 °C for 10 min shows the highest shear strength of$38.13 MPa. At the same time, the fracture paths of brazed joints also change as the temperature increases. When the brazing temperature is 950 °C, the fracture position is in the TiC + ZrC +Ti_2O + ZrSi_2+ Ti_5Si_3 layer on the composite side. When the brazing temperature is 980 °C, the fracture position is on the side of the braze seam(Ti, Zr)2(Ni, Cu), Ti_2O + ZrSi_2+ Ti_5Si_3 layer,and carbon fiber in the composite material. | 6 | Rabdosia serra alleviates dextran sulfate sodium salt-induced colitis in mice through anti-inflammation,regulating Th17/Treg balance,maintaining intestinal barrier integrity,and modulating gut microbiota显示文摘Rabdosia serra(R.serra),an important component of Chinese herbal tea,has traditionally been used to treat hepatitis,jaundice,cholecystitis,and colitis.However,the chemical composition of R.serra and its effect against colitis remain unclear.In this study,the chemical composition of the water extract of R.serra was analyzed using ultra performance liquid chromatography coupled with a hybrid linear ion trap quadrupole-orbitrap mass spectrometer(UPLC-LTQ-Orbitrap-MS).A total of 46 compounds,comprising ent-kaurane diterpenoids,flavonoids,phenolic acids,and steroids,were identified in the water extract of R.serra,and the extract could significantly alleviate dextran sulfate sodium salt-induced colitis by improving colon length,upregulating anti-inflammatory factors,downregulating proinflammatory factors,and restoring the balance of T helper 17/T regulatory cells.R.serra also preserved intestinal barrier function by increasing the level of tight junction proteins(zonula occludens 1 and occludin)in mouse colonic tissue.In addition,R.serra modulated the gut microbiota composition by increasing bacterial richness and diversity,increasing the abundance of beneficial bacteria(Muribaculaceae,Bacteroides,Lactobacillus,and Prevotellaceae_UCG-001),and decreasing the abundance of pathogenic bacteria(Turicibacter,Eubacterium_fissicatena_group,and Eubacterium_xylanophilum_group).Gut microbiota depletion by antibiotics further confirmed that R.serra alleviated colitis in a microbiota-dependent manner.Overall,our findings provide chemical and biological evidence for the potential application of R.serra in the management of colitis. | Hongyi Li Yi Wang Shumin Shao Hui Yu Deqin Wang Chuyuan Li Qin Yuan Wen Liu Jiliang Cao Xiaojuan Wang Haibiao Guo Xu Wu Shengpeng Wang | 2022 | Journal of Pharmaceutical Analysis2022,12,6: | 4 |
| 7 | Solar-Driven Producing of Value-Added Chemicals with Organic Semiconductor-Bacteria Biohybrid System显示文摘Photosynthetic biohybrid systems exhibit promising performance in biosynthesis;however,these systems can only produce a single metabolite and cannot further transform carbon sources into highly valuable chemical production.Herein,a photosynthetic biohybrid system integrating biological and chemical cascade synthesis was developed for solar-driven conversion of glucose to value-added chemicals.A new ternary cooperative biohybrid system,namely bacterial factory,was constructed by self-assembling of enzyme-modified light-harvesting donor-acceptor conjugated polymer nanoparticles(D-A CPNs)and genetically engineered Escherichia coli(E.coli).The D-A CPNs coating on E.coli could effectively generate electrons under light irradiation,which were transferred into E.coli to promote the 37%increment of threonine production by increasing the ratio of nicotinamide adenine dinucleotide phosphate(NADPH).Subsequently,the metabolized threonine was catalyzed by threonine deaminase covalently linking with D-A CPNs to obtain 2-oxobutyrate,which is an important precursor of drugs and chemicals.The 2-oxobutyrate yield under light irradiation is increased by 58%in comparison to that in dark.This work provides a new organic semiconductor-microorganism photosynthetic biohybrid system for biological and chemical cascade synthesis of highly valuable chemicals by taking advantage of renewable carbon sources and solar energy. | Wen Yu Haotian Bai Yue Zeng Hao Zhao Shengpeng Xia Yiming Huang Fengting Lv Shu Wang | 2022 | Research2022,,3: | 2 |
| 8 | A polyphenol-assisted IL-10 mRNA delivery system for ulcerative colitis显示文摘With the development of synthesis technology, modified messenger RNA(mRNA) has emerged as a novel category of therapeutic agents for a broad of diseases. However, effective intracellular delivery of mRNA remains challenging, especially for its sensitivity to enzymatic degradation. Here, we propose a polyphenol-assisted handy delivery strategy for efficient in vivo delivery of IL-10 mRNA.IL-10 mRNA binds to polyphenol ellagic acid through supramolecular binding to yield a negatively charged core, followed by complexing with linear polyetherimide and coating with bilirubin-modified hyaluronic acid to obtain a layer-by-layer nanostructure. The nanostructure specifically up-regulated the level of IL-10, effectively inhibited the expression of inflammatory factors, promoted mucosal repair, protected colonic epithelial cells against apoptosis, and exerted potent therapeutic efficacy in dextran sulfate sodium salt-induced acute and chronic murine models of colitis. | 18 | Rational Design of Thermosensitive Hydrogel to Deliver Nanocrystals with Intranasal Administration for Brain Targeting in Parkinson’s Disease显示文摘Mitochondrial dysfunction is commonly detected in individuals suffering from Parkinson’s disease(PD),presenting within the form of excessive reactive oxygen species(ROS)generation as well as energy metabolism.Overcoming this dysfunction within brain tissues is an effective approach to treat PD,while unluckily,the blood-brain barrier(BBB)substantially impedes intracerebral drug delivery.In an effort to improve the delivery of efficacious therapeutic drugs to the brain,a drug delivery platform hydrogel(MAG-NCs@Gel)was designed by complexing magnolol(MAG)-nanocrystals(MAG-NCs)into the noninvasive thermosensitive poly(Nisopropylacrylamide)(PNIPAM)with self-gelation.The as-prepared MAG-NCs@Gel exhibited obvious improvements in drug solubility,the duration of residence with the nasal cavity,and the efficiency of brain targeting,respectively.Above all,continuous intranasal MAG-NCs@Gel delivery enabled MAG to cross the BBB and enter dopaminergic neurons,thereby effectively alleviating the symptoms of MPTP-induced PD.Taking advantage of the lower critical solution temperature(LCST)behavior of this delivery platform increases its viscoelasticity in nasal cavity,thus improving the efficiency of MAG-NCs transit across the BBB.As such,MAG-NCs@Gel represented an effective delivery platform capable of normalizing ROS and adenosine triphosphate(ATP)in the mitochondria of dopaminergic neurons,consequently reversing the mitochondrial dysfunction and enhancing the behavioral skills of PD mice without adversely affecting normal tissues. | Yun Tan Yao Liu Yujing Liu Rui Ma Jingshan Luo Huijie Hong Xiaojia Chen Shengpeng Wang Chuntai Liu Yi Zhang Tongkai Chen | 2021 | Research2021,,1: | 1 |
