1Cryo-EM structures of inactive and active GABAB receptor显示文摘Metabotropic GABAB G protein-coupled receptor functions as a mandatory heterodimer of GB1 and GB2 subunits and mediates inhibitory neurotransmission in the central nervous system.Each subunit is composed of the extracellular Venus flytrap(VFT)domain and transmembrane(TM)domain.Here we present cryo-EM structures of full-length human heterodimeric GABAB receptor in the antagonist-bound inactive state and in the active state complexed with an agonist and a positive allosteric modulator in the presence of Gi1 protein at a resolution range of 2.8–3.0Å.Our structures reveal that agonist binding stabilizes the closure of GB1 VFT,which in turn triggers a rearrangement of TM interfaces between the two subunits from TM3-TM5/TM3-TM5 in the inactive state to TM6/TM6 in the active state and finally induces the opening of intracellular loop 3 and synergistic shifting of TM3,4 and 5 helices in GB2 TM domain to accommodate theα5-helix of Gi1.We also observed that the positive allosteric modulator anchors at the dimeric interface of TM domains.These results provide a structural framework for understanding class C GPCR activation and a rational template for allosteric modulator design targeting the dimeric interface of GABAB receptor.Chunyou Mao Cangsong Shen Chuntao Li Dan-Dan Shen Chanjuan Xu Shenglan Zhang Rui Zhou Qingya Shen Li-Nan Chen Jianfeng Liu Yan Zhang 2020Cell Research2020,30,7:3

7Relationship between Ponkan Mandarin Fruit Quality and Nutrient Concentrations显示文摘Five age groups of trees were carried out in order to study relationships between the deterioration of Ponkan mandarin fruit quality and changes in tissues nutrient concentrations with tree age.Fruit quality and nutrient concentrations in fruits and leaves were measured.Comprehensive fruit quality decreased in the order:5-year-old > 13-year-old > 38-year-old > 8-year-old >18-year-old.In 18-year-old trees,calcium(Ca) concentrations of fruit and leaf were lower,while potassium(K) and boron(B) concentrations of fruit and magnesium(Mg) and manganese(Mn) concentrations of leaf were higher.Negative relationships were observed between fruit total soluble solids and leaf B,while positive relationships were observed between fruit total soluble solids and leaf and fruit Ca concentrations,and between titratable acidity and leaf Mn.Fruit quality decreasing with tree age most likely resulted from an imbalance of high K,Mg and B but lower Ca concentrations in leaves and fruits.Hong LENG Qiling TAN Chengxiao HU Cangsong ZHENG Shaokang SU 2016Agricultural Science & Technology2016,17,7:0
8Effects of soil potassium levels on dry matter and nutrient accumulation and distribution in cotton显示文摘Background Potassium(K)is an essential nutrient for plant growth and development.However,plant fertilization ignoring the soil K level is very likely to cause excessive fertilizer use,and further arouse a series of side effects.This study investigated the response of cotton growth to different soil K levels and the uptake of major nutrients,aiming to evaluate the appropriate K supply level for cotton growth.Using a random block design with 6 soil K levels,we conducted 18 micro-zones field experiments over two continuous years.The soil available K concentration of each treatment was K1(99.77-100.90 mg·kg^(-1)),K2(110.90-111.26 mg·kg^(-1)),K3(123.48-128.88 mg·kg^(-1)),K4(140.13-145.10 mg·kg^(-1)),K5(154.43-155.38 mg·kg^(-1)),and K6(165.77-168.75 mg·kg^(-1)).Cotton nutrient contents,soil nutrient contents,accumulation and distribution of dry matter in cotton were determined,and the relationships between K content in soil and plants and dry matter accumulation were analyzed.Results The soil K content had a significantly positive relationship with dry matter and K accumulation in cotton plants.There were significant differences in dry matter accumulation,single-plant seed cotton yield,mineral nutrient uptake and the proportion of K accumulation in reproductive organs among different soil K levels.The results showed that there was significant difference between K4 and lower K level treatments(K1 and K2),but no significant difference between K4 and higher K level treatments(K5 and K6)in dry matter,single-plant seed cotton yield,or accumulation,distribution and seed cotton production efficiency of N,P and K.Conclusion The soil K level of K4 was able to provide sufficient K for cotton growth in our experiment.Therefore,when the soil K level reached 140.13 mg·kg^(-1),further increasing the soil K concentration no longer had a significant positive effect on cotton growth.Jingjing SHAO Helin DONG Yinan JIN Pengcheng LI Miao SUN Weina FENG Cangsong ZHENG 2023Journal of Cotton Research2023,6,2:0
9The Breeding Ecology of a Critically Endangered Salamander,Hynobius amjiensis(Caudata:Hynobiidae),Endemic to Eastern China显示文摘Hynobius amjiensis is a critically endangered salamander species(IUCN Red List) endemic to eastern China. It currently has three known populations: one in Longwangshan, Zhejiang Province(type locality), and two in Qingliangfeng between Anhui and Zhejiang Provinces. We examined the relatively unstudied breeding ecology of this species in the field and at laboratory from March 2007 to May 2014. Adult males and females were year-round terrestrial, except for the February–April breeding season. During this period, we captured only a total of 16 breeding adults(11 males and 5 females). As few as 100 breeding females were estimated based on the number of egg sacs observed since 2007. This number was significantly reduced from the estimated number between 1992 and 1998. Males(mean total length = 16.21 cm, mean body mass = 18.8 g) were slightly smaller than females(16.51 cm, 19.2 g). Size of breeding pools ranged from 0.2 m^2 to 1.2 m^2(0.1–1.2 m depths). Each female deposits a pair of egg sacs by attaching the adhesive tips of the sacs to aquatic plants or dead twigs. Fifteen pairs of egg sacs had an average length of 28.6 cm and a diameter of 3.3 cm. On average, each egg sac contained 75 eggs with a diameter of 0.3 cm. Our field survey revealed that H. amjiensis used oviposition sites in small, cool, and weakly acidic pools at high elevations(1 300–1 600 m) where peat moss was abundant. Reduction in wetland size and disappearance of suitable breeding pools suggest that this salamander species is under threat of extinction, particularly at Longwangshan, where 5 of the 9 breeding pools have either dried up or disappeared. Combined size of the remaining 4 pools is less than 2 m^2. 10Minimally invasive direct coronary artery bypass plus coronary stent for acute coronary syndrome:a case report显示文摘A 69-year old female patient was admitted because of 3 days of worsened chest pain.Coronary angiography showed 60% stenosis of distal left main stem,chronic total occlusion of left anterior descending (LAD),70% stenosis at the ostium of a small left circumflex,70-90% stenosis at the paroxysmal and middle part of a dominant right coronary artery (RCA),and a normal left internal mammary artery (LIMA) with normal origination and orientation.Percutaneous intervention was attempted but failed on the occluded lesion of LAD.The patient received minimally invasive direct coronary artery bypass (MIDCAB) with left LIMA isolation by Davinci robot.Eleven days later,the RCA lesion was treated by Sirolimus Rapamicin eluting stents implantation percutaneously.Then the patient was discharged uneventfully after 3 days hospitalization.Our experience suggests that two stop shops of hybrid technique be feasible and safe in the treatment of elderly patient with multiple coronary diseases.Caiyi Lu~1 Cangqing Gao~2 Shiwen Wang~1 Yuxiao Zhang~1 Ming Yang~2 Qiao Xue~1 Cangsong Xiao Wei Gao~1 Yang Wu~2 Gang Wang~3 Qi Zhou~3 Jinwen Tian~1 Lei Gao~1 Shenhua Zhou~1 Jinyue Zhai~1 Rui Chen~1 Zhongren Zhao~1 1.
