| 1 | Genome and transcriptome analysis of the grapevine(Vitis vinifera L.) WRKY gene family显示文摘The plant WRKY gene family represents an ancient and complex class of zinc-finger transcription factors(TFs)that are involved in the regulation of various physiological processes,such as development and senescence,and in plant response to many biotic and abiotic stresses.Despite the growing number of studies on the genomic organisation of WRKY gene family in different species,little information is available about this family in grapevine(Vitis vinifera L.).In the present study,a total number of 59 putative grapevine WRKY transcription factors(VvWRKYs)were identified based on the analysis of various genomic and proteomic grapevine databases.According to their structural and phylogentic features,the identified grapevine WRKY transcription factors were classified into three main groups.In order to shed light into their regulatory roles in growth and development as well as in response to biotic and abiotic stress in grapevine,the VvWRKYs expression profiles were examined in publicly available microarray data.Bioinformatics analysis of these data revealed distinct temporal and spatial expression patterns of VvWRKYs in various tissues,organs and developmental stages,as well as in response to biotic and abiotic stresses.To also extend our analysis to situations not covered by the arrays and to validate our results,the expression profiles of selected VvWRKYs in response to drought stress,Erysiphe necator(powdery mildew)infection,and hormone treatments(salicilic acid and ethylene),were investigated by quantitative real-time reverse transcription PCR(qRT-PCR).The present study provides a foundation for further comparative genomics and functional studies of this important class of transcriptional regulators in grapevine. | Min Wang Alessandro Vannozzi Gang Wang Ying-Hai Liang Giovanni Battista Tornielli Sara Zenoni Erika Cavallini Mario Pezzotti Zong-Ming(Max)Cheng | 2014 | Horticulture Research2014,1,1: | 19 |

| 9 | Physical energies to the rescue of damaged tissues显示文摘Rhythmic oscillatory patterns sustain cellular dynamics, driving the concerted action of regulatory molecules, microtubules, and molecular motors. We describe cellular microtubules as oscillators capable of synchronization and swarming, generating mechanical and electric patterns that impact biomolecular recognition. We consider the biological relevance of seeing the inside of cells populated by a network of molecules that behave as bioelectronic circuits and chromophores. We discuss the novel perspectives disclosed by mechanobiology, bioelectromagnetism, and photobiomodulation, both in term of fundamental basic science and in light of the biomedical implication of using physical energies to govern (stem) cell fate. We focus on the feasibility of exploiting atomic force microscopy and hyperspectral imaging to detect signatures of nanomotions and electromagnetic radiation (light), respectively, generated by the stem cells across the specification of their multilineage repertoire. The chance is reported of using these signatures and the diffusive features of physical waves to direct specifically the differentiation program of stem cells in situ, where they already are resident in all the tissues of the human body. We discuss how this strategy may pave the way to a regenerative and precision medicine without the needs for (stem) cell or tissue transplantation. | 10 | Bimanual microincision cataract surgery with implantation of the new Incise~? MJ14 intraocular lens through a 1.4 mm incision显示文摘AIM: To analyze the visual outcomes and the posterior capsule opacification(PCO) with the new Incise? MJ14 intraocular lens(IOL) implanted through a 1.4 mm clear corneal incision(CCI) in patients who underwent bimanual microincision cataract surgery(B-MICS). METHODS: Eighty eyes which underwent cataract surgery using B-MICS technique performed by the same experienced surgeon were included in the study: 40 eyes were implanted with an Incise? MJ14 IOL through a 1.4 mm CCI(group A)without enlargement of the main CCI, while 40 eyes were implanted with an Akreos? MI60 IOL with enlargement of the main CCI to 1.8 mm(group B). Best corrected visual acuity(BCVA), astigmatism and endothelial cell loss were evaluated before and after surgery at 7, 30 d and 6 mo.Anterior segment-optical coherence tomography(AS-OCT) of CCI was performed at 1, 3, 7, 30 d, 6 and 18 mo. PCO incidence was evaluated at 18 mo using EPCO 2000 Software. RESULTS: Mean BCVA improvement and endothelial cell loss were statistically significant at 18 mo in both groups with no difference between the two groups; no statistically significant difference in surgically induced astigmatism(SIA) was noticed in the two groups. At AS-OCT the only significant alterations in the CCI were endothelial gaping and local detachment of Descemet's membrane at 1 and 7 d after surgery; no statistically significant alterations were found at 1, 6 and 18 mo. PCO score at 18 mo was 0.03±0.07 for group A and 0.08±0.18 for group B(P=0.11) with no sign of central optic plate invasion in both groups.CONCLUSION: The implant of the new Incise? MJ14 IOL through a 1.4 mm CCI and B-MICS technique appeared to be a safe and effective procedure with rapid visual recovery.
