2MicroRNAs in pathogenesis of breast cancer:Implications in diagnosis and treatment显示文摘MicroR NAs(miR NAs) are small non-coding RNAs generated by a two-step complex process and are post transcriptional negative regulators of their target m RNAs. Dysregulation of many of these miR NAs has been associated with tumorigenesis in various cancers including breast cancer. Aberrantly high expression of specific mi RNAs in breast cancer cells is demonstrated to be linked with inhibition of tumor suppressor genes and promote tumorigenesis. They are classified as oncogenic miR NAs. However, the tumor suppressor miR NAs are downregulated in breast cancer cells, since their major targets are oncogenic m RNAs. Understanding mechanism of action of specific miR NAs in breast cancer cells can be utilized to develop newer anti-cancer therapies. Recently, newer techniques are also developed to detect abundance of specific miR NA in the blood plasma samples and can be used in early diagnosis or prognosis in breast cancer. 3Key Recovery Against 3DES in CPU Smart Card Based on Improved Correlation Power Analysis显示文摘The security of CPU smart cards,which are widely used throughout China,is currently being threatened by side-channel analysis.Typical countermeasures to side-channel analysis involve adding noise and filtering the power consumption signal.In this paper,we integrate appropriate preprocessing methods with an improved attack strategy to generate a key recovery solution to the shortcomings of these countermeasures.Our proposed attack strategy improves the attack result by combining information leaked from two adjacent clock cycles.Using our laboratory-based power analysis system,we verified the proposed key recovery solution by performing a successful correlation power analysis on a Triple Data Encryption Standard(3DES)hardware module in a real-life 32-bit CPU smart card.All 112 key bits of the 3DES were recovered with about 80 000 power traces.Yanting Ren Liji Wu Hexin Li Xiangyu Li Xiangmin Zhang An Wang Hongyi Chen 2016Tsinghua Science and Technology2016,21,2:3

13Cancer resistance to immunotherapy: What is the role of cancer stem cells?显示文摘Immunotherapy is an emerging form of cancer therapy that is associated with promising outcomes.However,most cancer patients either do not respond to immunotherapy or develop resistance to treatment.The resistance to immunotherapy is poorly understood compared to chemotherapy and radiotherapy.Since immunotherapy targets cells within the tumor microenvironment,understanding the behavior and interactions of different cells within that environment is essential to adequately understand both therapy options and therapy resistance.This review focuses on reviewing and analyzing the special features of cancer stem cells(CSCs),which we believe may contribute to cancer resistance to immunotherapy.The mechanisms are classified into three main categories:mechanisms related to surface markers which are differentially expressed on CSCs and help CSCs escape from immune surveillance and immune cells killing;mechanisms related to CSC-released cytokines which can recruit immune cells and tame hostile immune responses;and mechanisms related to CSC metabolites which modulate the activities of infiltrated immune cells in the tumor microenvironment.This review also discusses progress made in targeting CSCs with immunotherapy and the prospect of developing novel cancer therapies.Gourab Gupta George Merhej Shakthika Saravanan Hexin Chen 2022Cancer Drug Resistance2022,5,4:0

15High-contrast and real-time visualization of membrane proteins in live cells with malachite green-based fluorogenic probes显示文摘Imaging dynamics of membrane proteins of live cells in a wash-free and real-time manner has been a challenging task. Herein, we report unprecedented applications of malachite green(MG), an organic dye widely used in pigment industry, as a switchable fluorophore to monitor membrane enzymes or noncatalytic proteins in live cells. Conformationally flexible MG is non-fluorescent in aqueous solution, yet covalent binding with endogenous proteins of cells significantly enhances its fluorescence at 670 nm by restricting flexibility of dye. Integrating a phosphate-caged quinone methide precursor with MG yielded a covalent labeling fluorogenic probe, allowing real-time imaging of membrane alkaline phosphatase(ALP,a model catalytic protein) activity in live cells with over 100-fold enhancement of fluorescence intensity.Moreover, MG is also applicable to image non-catalytic protein by conjugation with protein-specific ligand.
