2Genome analysis of Taraxacum kok-saghyz Rodin provides new insights into rubber biosynthesis显示文摘The Russian dandelion Taraxacum kok-saghyz Rodin(TKS), a member of the Composite family and a potential alternative source of natural rubber(NR) and inulin, is an ideal model system for studying rubber biosynthesis. Here we present the draft genome of TKS, the first assembled NR-producing weed plant. The draft TKS genome assembly has a length of 1.29 Gb, containing 46 731 predicted protein-coding genes and68.56% repeats, in which the LTR-RT elements predominantly contribute to the genome enlargement. We analyzed the heterozygous regions/genes, suggesting its possible involvement in inbreeding depression.Through comparative studies between rubber-producing and non-rubber-producing plants, we found that enzymes of the mevalonate(MVA) pathway and rubber elongation might be critical for rubber biosynthesis, and several key isoforms have been isolated and shown to be predominantly expressed in the latex, indicating their crucial functions in rubber biosynthesis. Moreover, for two important families in rubber elongation, the CPT/CPTL and REF/SRPP families, diverse evolutionary tracks have been revealed. 3Geology,Geochemistry and Zircon U-Pb Geochronology of Porphyries in the Dabate Mo-Cu Deposit,Western Tianshan,China:Petrogenesis and Tectonic Implications显示文摘The Dabate Mo-Cu deposit is a medium-sized porphyry-type deposit in the Sailimu Lake region, western Tianshan, China. We present the geology, geochemistry and zircon U-Pb geochronology of granite porphyries from the Dabate district with the intent to constrain their tectonic setting and petrogenesis. Porphyries in the Dabate district include granite porphyry I(gray white color with large phenocrysts), granite porphyry II(pink color with small phenocrysts) and quartz porphyry. Granite porphyry II is the Cu and Mo ore-bearing granitoid in the Dabate deposit. LA-ICPMS zircon U-Pb analyses indicate that granite porphyry II was emplaced at 284.2±1.8 Ma. Granite porphyry I and II have similar geochemical features and are both highly fractionated granites:(1) They have high SiO_2 content(70.93–80.18 wt% and 72.14–72.64 wt%, respectively), total alkali(7.58–8.95 wt% and 9.35–9.68 wt%, respectively), mafic index(0.95–0.98 and 0.93–0.94, respectively) and felsic index(0.79–0.94 and 0.89–0.91, respectively);(2) They are characterized by pronounced negative Eu anomaly, 'seagullstyle' chondrite-normalized REE patterns and 'tetrad effect' of REE;(3) They are rich in Rb, K, Th, Ta, Zr, Hf, Y and REE, but depleted in Sr, P, Ti and Nb. 4Image matching algorithm based on SIFT using color and exposure information显示文摘Image matching based on scale invariant feature transform(SIFT) is one of the most popular image matching algorithms, which exhibits high robustness and accuracy. Grayscale images rather than color images are generally used to get SIFT descriptors in order to reduce the complexity. The regions which have a similar grayscale level but different hues tend to produce wrong matching results in this case. Therefore, the loss of color information may result in decreasing of matching ratio. An image matching algorithm based on SIFT is proposed, which adds a color offset and an exposure offset when converting color images to grayscale images in order to enhance the matching ratio. Experimental results show that the proposed algorithm can effectively differentiate the regions with different colors but the similar grayscale level, and increase the matching ratio of image matching based on SIFT. 5CMTM4 inhibits cell proliferation and migration via AKT, ERK1/2,and STAT3 pathway in colorectal cancer显示文摘CMTM4(CKLF-like MARVEL transmembrane domain containing 4),a potential tumor suppressor gene,is involved in several types of malignancies.It has been reported to be downregulated and exhibit anti-tumorigenic activities by regulating cell growth and cell cycle in clear cell renal cell carcinoma.It has also been identified as a tumor suppressor in hepatocellular carcinoma(HCC),and its negative expression is a risk factor for poor prognosis of HCC patients.In the present study,an integrated bioinformatics analysis based on The Cancer Genome Atlas(TCGA)database showed that CMTM4 was frequently reduced in colorectal cancer(CRC)and high expression of CMTM4 was associated with increased overall survival rates.Based on these findings,we adopted gain-of-function and lost-of-function strategies using SW480 and HT29 CRC cell lines which have relatively low and high endogenous CMTM4 levels,respectively.We observed impeded cell proliferation and migration upon overexpression of CMTM4 in SW480 cells,and the opposite effects were observed upon knockdown of CMTM4 in HT-29 cells.Cell signaling pathways essential for CRC progression were then examined,and the phosphorylation levels of AKT,ERK1/2,and STAT3 were found to be decreased by CMTM4 overexpression in SW480 cells and elevated by CMTM4 silencing in HT29 cells.Their inhibitors were used to validate that the three signaling pathways contributed to the inhibitory effects of CMTM4 on CRC cells.Taken together,our results suggest that CMTM4 plays a tumor suppressive role in CRC.Hui Xue Ting Li Pingzhang Wang Xiaoning Mo Hejun Zhang Shigang Ding Dalong Ma Wenping Lv Jing Zhang Wenling Han 2019Acta Biochimica et Biophysica Sinica2019,51,9:5
6Protection of Li metal anode by surface-coating of PVDF thin film to enhance the cycling performance of Li batteries显示文摘Lithium metal, the ideal anode material for next-generation high-energy batteries, suffers from the severe safety problem of Li dendrites. Herein, we report a simple approach to effectively maintain the morphology of Li-metal anode and enhance the cycling performance of Li batteries by surface coating of a porous polyvinylidene fluoride (PVDF) thin film. In symmetrical cells testing, the cells with the Li@PVDF electrode display stable cycling performance more than 1300 h (650 cycles) at the current density of 0.5 mA/cm^2 with a stripping/plating capacity of 0.5 mAh/cm^2. 7Effect of Oxidation-Induced Material Parameter Variation on the High Temperature Oxidation Behavior of Nickel显示文摘In high temperature oxidation environment, the oxidation reaction will induce variations in material parameters, such as Young's modulus, thermal expansion coefficient(CTE),coefficient of oxygen diffusion(COD), etc. The oxidation-induced material parameter variations should be considered in high temperature mechanical analysis. In this paper, high temperature oxidation behavior of an oxide film/metal substrate system was investigated through a modified phase field approach. The oxidative stress and oxidation weight gain induced by high temperature oxidation were studied. Effects of Young's modulus, COD and CTE on oxidative stress in the oxide film were studied particularly. The simulation results showed that a better agreement with the experimental results could be obtained when considering the oxidation-induced material parameter variations in the high temperature mechanical analysis of oxide film/metal substrate system. 8Roles of insulin-like growth factor Ⅱ in regulating female reproductive physiology显示文摘The number of growth factors involved in female fertility has been extensively studied, but reluctance to add essential growth factors in culture media has limited progress in optimizing embryonic growth and implantation outcomes, a situation that has ultimately led to reduced pregnancy outcomes. Insulin-like growth factor Ⅱ(IGF-Ⅱ) is the most intricately regulated of all known reproduction-related growth factors characterized to date, and is perhaps the predominant growth factor in human ovarian follicles. This review aims to concisely summarize what is known about the role of IGF-Ⅱ in follicular development, oocyte maturation, embryonic development, implantation success, placentation, fetal growth, and in reducing placental cell apoptosis, as well as present strategies that use growth factors in culture systems to improve the developmental potential of oocytes and embryos in different species. 9The Effect of Body Pitching on Leg-Spring Behavior in Quadruped Running显示文摘In this paper we investigated how the running speed would affect the dynamics of body pitching, and whether body inertiais important for animals. Passive trotting of spring-mass model and passive bounding of spring-beam model were studied atdifferent speeds for different sets of body parameters respectively. Furthermore, different body inertias were used in bounding.We found that running speed exerts effect on leg performance by means of centrifugal force. The centrifugal force can be understoodas an enhancement to the natural frequency of the spring-mass system. The disadvantage of body pitching may beoffset by the great increase in centrifugal force at high speed. 10BDNF improves axon transportation and rescues visual function in a rodent model of acute elevation of intraocular pressure显示文摘Optic neuropathies lead to blindness;the common pathology is the degeneration of axons of the retinal ganglion cells. In this study, we used a rat model of retinal ischemia-reperfusion and a one-time intravitreal brain-derived neurotrophic factor(BDNF)injection;then we examined axon transportation function, continuity, physical presence of axons in different part of the optic nerve, and the expression level of proteins involved in axon transportation. We found that in the disease model, axon transportation was the most severely affected, followed by axon continuity, then the number of axons in the distal and proximal optic nerve. BDNF treatment relieved all reductions and significantly restored function. The molecular changes were more minor,probably due to massive gliosis of the optic nerve, so interpretation of protein expression data should be done with some caution.The process in this acute model resembles a fast-forward of changes in the chronic model of glaucoma. Therefore, impairment in axon transportation appears to be a common early process underlying different optic neuropathies. 13BmCH25H,a vertebrate interferon-stimulated gene(ISG)homolog,inhibits BmNPV infection dependent on its hydroxylase activity in Bombyx mori显示文摘Cholesterol-25-hydroxylase(CH25H)has been identified as an interferon-stimulated gene(ISG)in mammals that exerts its antiviral effects by catalyzing the conversion of cholesterol to 25-hydroxycholesterol(25HC).However,invertebrates lack an antiviral system homologous to vertebrate interferons(IFNs)because the genomes of invertebrates do not encode IFN-like cytokines.Nevertheless,CH25H is present in insect genomes and it therefore deserves further study of whether and by which mechanism it could exert an antiviral effect in invertebrates.In this study,the Bombyx mori CH25H(BmCH25H)gene,of which the encoded protein has high homology with other lepidopteran species,was identified and located on chromosome 9.Interestingly,we found that the expression of BmCH25H was significantly upregulated in B.mori nucleopolyhedrovirus(BmNPV)-infected BmN cells and silkworm(B.mori)larvae at the early infection stage.The inhibitory effect of BmCH25H on BmNPV replication was further demonstrated to depend on its catalytic residues to convert cholesterol to 25HC.More importantly,we demonstrated that during BmNPV infection,BmCH25H expression was increased through the Janus kinase–signal transducer and activator of transcription(JAK–STAT)pathway,similar to the induction of ISGs following virus infection in vertebrates.This is the first report that CH25H has antiviral effects in insects;the study also elucidates the regulation of its expression and its mechanism of action.Hongyun Wu Junming Xia Shigang Fei Yeyuan Wang Mengmeng Zhang Yiyao Guo Xian Li Luc Swevers Jingchen Sun Min Feng 2023Insect Science2023,30,2:2
14Long-lasting impairments in rodent oxygen-induced retinopathy measured by retinal vessel density and visual function显示文摘In mild or moderate retinopathy of prematurity(ROP), retinal vessels undergo obliteration, proliferation, and regression. Despite complete regression of vessel abnormalities, a variety of visual impairments have been reported. Rodent oxygen-induced retinopathy(OIR) is widely used as a model to study ROP. However, the long-term changes of OIR model remain unclear. The aim of this study is to examine long term changes of retinal vessel and visual function in a rodent OIR model resembling human mild or moderate ROP. In this study, after subjecting the animals to 80% oxygen(O_2) for 5–7 d, the retinal vessel density at postnatal day 12(P12) was approximately 30% lower than that in the age-matched control, but this difference was not significant between the groups. Vessel abnormalities, such as vessel tortuosity, neovascular tufts, and the number of vessels protruding into the vitreous, peaked between P17 and P20. Despite the regression of many abnormalities, vessel density in the OIR group was 36% and 32% lower than that in the control animals at 6 weeks and 4 months, respectively. The visual acuity and contrast sensitivity were impaired in the OIR group when measured at 2, 3 and 4 months. Therefore, the rodent OIR model exhibited long-lasting reduction in retinal vessel density and visual impairments, similar to those observed in mild or moderate human ROP. 15Association between essential hypertension and polymorphisms of beta 1 adrenergic receptor gene G1165C (Gly389Arg) in Chinese Mongolian population显示文摘BACKGROUND: The prevalences of hypertension, cerebrovascular diseases, etc. are higher in Mongolian population because of the influence of various factors including genetics, geography, diet, etc. Therefore, it is helpful to develop researches on the genetics of various diseases including hypertension in Mongolian population. OBJECTIVE: To analyze the association between the polymorphism of beta1 adrenergic receptor (β1-AR) gene G1165C (Arg389Gly), an important candidate gene for various diseases of cardiovascular system, and essential hypertension in Mongolian population. DESIGN: A cross-sectional study. SETTINGS: Department of Neurology, the First Affiliated Hospital of Inner Mongolia Medical College; Wulate Houqi Red Cross Society. PARTICIPANTS: The survey was carried out from February 2003 to March 2005. Totally 239 Mongolian residents, whose blood relations of 3 generations were all Mongolians, were selected from Wulate Houqi, Inner Mongolia, and they were all informed with the survey and detected items. Based on the diagnostic standard of hypertension set by WHO in 1999, the subjects were divided into two groups according to the level blood pressure: ① Normal blood pressure group (n=117): systolic blood pressure (SBP) < 140 mm Hg (1 mm Hg =0.133 kPa), diastolic blood pressure (DBP) < 90 mm Hg, and those having histories of cerebrovascular disease, heart disease, diseases of liver, kidney and tiroides, and diabetes mellitus were excluded. ② Essential hypertension group (n=122): including 51 patients with simple high SBP. All the enrolled subjects had no blood relationship with each other, and had no history of miscegenation. METHODS: The body height, body mass, waist circumference and blood lipids were measured routinely, and their habits of smoking and drinking were also investigated. Peripheral venous blood (5 mL) was drawn, the genome DNA was extracted, and the polymorphisms of the β1-AR G1165C (Gly389Arg) genotype were detected with the Sequenom system. Polymerase chain reaction (PCR) experiment and SNP detection were performed in Huada Gene Laboratory of Bejing, then the univariate analysis of variance was applied in the sample comparison among groups, and the chi-square test was used to compare the genotypes and allele frequencies. The odd ratio (OR) and 95% confidence interval (CI) were calculated. MAIN OUTCOME MEASURES: The distributions of β1-AR G1165C (Gly389Arg) genotypes and alleles were observed. RESULTS: All the 239 subjects were involved in the analysis of results, and no one missed. ①Comparison of β1-AR G1165C (Gly389Arg) genotypes and allele distributions: In Mongolian population, the frequencies of CC and GG+GC genotypes at β1-AR G1165C (Gly389Arg) site in the essential hypertension group (72%, 28%) were not significantly different from those in the normal blood pressure group (67%, 33%) (χ2=0.841, P=0.359; OR: 0.773, 95%CI: 0.445-1.342); The frequencies of C and G alleles also had no significant differences between the essential hypertension group (85%, 15%) and the normal blood pressure group (82%, 18%) (χ2=1.136, P=0.287; OR: 0.769, 95%CI: 0.747-1.248). ②The frequencies of CC and GG+GC genotypes at β1-AR G1165C (Gly389Arg) site had no significant differences between the patients with simple high SBP (71%, 29%) and the normal blood pressure group (χ2=0.250, P=0.617; OR: 0.833, 95%CI: 0.408-1.703); The frequencies of C and G alleles were not significantly different between the patients with simple high SBP (86%, 14%) and the normal blood pressure group (χ2=0.670, P=0.413; OR: 0.766, 95%CI: 0.404-1.453). 16C1orf106, an innate immunity activator, is amplified in breast cancer and is required for basal-like/luminal progenitor fate decision显示文摘Basal-like breast cancer with a luminal progenitor gene expression profile is an aggressive subtype of breast cancer with a poorer prognosis compared with other subtypes.However,genes that specifically promote basal-like breast cancer development remain largely unknown.Here,we report that a novel gene C1orf106 plays an important role in maintaining the feature of basal-like/luminal progenitors.C1orf106 is frequently amplified and overexpressed in basal-like breast cancer and is associated with a poor outcome in patients.In human TCGA database,C1orf106 expression was correlated with upregulation of ELF5 and downregulation of GATA3,two transcription factors that regulate mammary gland stem cell fate.Enhanced expression of C1orf106 promotes tumor progression and expression of basal-like/luminal progenitor marker ELF5;depletion of C1orf106 suppresses tumorigenesis and expression of basal-like/luminal progenitor marker GATA3.These findings suggest that C1orf106 maintains the basal-like/luminal progenitor character through balancing the expression of ELF5 and GATA3.Taken together,we demonstrated that C1orf106 is an important regulator for basal-like/luminal progenitors and targeting C1orf106 is of therapeutic value for breast cancer.Ji Ma Cheng Liu Decao Yang Jiagui Song Jing Zhang Tianzhuo Wang Mengyuan Wang Weizhi Xu Xueying Li Shigang Ding Jun Zhan Hongquan Zhang 2019Science China(Life Sciences)2019,62,9:2

18A phase-field study on the oxidation behavior of Ni considering heat conduction显示文摘Phase-field modeling approach has been used to study the oxidation behavior of pure Ni when considering heat conduction. In this calculation, the dependence of the coefficient of the Cahn–Hilliard equation Lc on the temperature T was considered. To this end, high-temperature oxidation experiments and phase-field modeling for pure Ni were performed in air under atmospheric pressure at 600,700, and 800?C. The oxidation rate was measured by thermogravimetry and Lc at these temperatures was determined via interactive algorithm. With the Lc-T relationship constructed, oxidation behavior of Ni when considering heat conduction was investigated. The influence of temperature boundaries on the oxidation degree, oxide film thickness, and specific weight gain were discussed. 19Antisense Oligonucleotide Targeting TGF-β1 Abrogates Tumorigenicity of Rhabdomyosarcoma in vivo显示文摘OBJECTIVE Over-expression of transforming growth factorβ1(TGF-β1)has been observed in many advanced cancers. The present study was aimed at developing potential antisense oligonucleotides(ASONs)to repress TGF-β1 expression in rhabdomyosarcoma(RMS)RD cells,and to examine their effect on tumorigenicity of RD cells in vivo. METHODS ASONs targeting the region surrounding the start codon of TGF-β1 were synthesized and transferred into cells in the form of complexes with Lipofectamine 2000.The TGF-β1 protein was determined by immunofluorescence and ELISA. The cell viability and cell cycle were examined by MTT and flow cytometry.The RD cells,with or without TGF-β1ASON,in 50μ1 of serum-free EMDM medium were injected subcutaneously into the right flank of nude mice.The tumors were then measured and weighed. RESULTS The ASON sequence targeting the first start site at bases 841-855 of the human TGF-β1 gene had the greatest effect on attenuating the expression of TGF-β1(P<0.05).The ASONs induced a decrease in OD values after 6 d(P<0.05).Analvsis of the cell cycle revealed that the ASON induced a significant decrease in cells in the S phase and an increase in cells in the G1 phase(P<0.05).In the nude mice model,the mean tumor volume,after 2 weeks of treatment with Lipofectamine or ASON, decreased to 88.5% or 55% respectively,compared to the control tumor size,resulting in a significant difference(P<0.01). 20Decreasing intraocular pressure significantly improves retinal vessel density,cytoarchitecture and visual function in rodent oxygen induced retinopathy显示文摘We attempted to explore a noninvasive,easily applicable and economically affordable therapy for retinopathy of prematurity(ROP).Rat pups were raised in 80%oxygen from postnatal day 7 to P12,and returned to room air.Travoprost eye drops were administered twice a day for 7 days,to reduce intraocular pressure(IOP)by about 20%.Immunohistochemical staining was performed to visualize vessel endothelial cells,to analyze retinal neurons and cytoarchitecture.Behavioral experiments were carried out to test visual acuity and contrast sensitivity.At the end of the 7-day treatment,the number of vessels extending to the vitreous body was significantly reduced and retinal vessel density increased.This improvement was maintained to the end of the12 th week.In the central retina of the model group,the horizontal cells were completely wiped out,the outer plexiform layer was undetectable,and the rod bipolar cell dendrites sprouted into the outer nuclear layer.The treatment partially reverted these architectural changes.Most importantly,behavioral experiments revealed significantly improved visual acuity and contrast sensitivity in the treated group.Therefore,reducing IOP could potentially serve as a safe and economical measure to treat ROP.Rui Du Xu Wang Ke Shen Shigang He 2020Science China(Life Sciences)2020,63,2:2
