1ALK gene expression status in pleural effusion predicts tumor responsiveness to crizotinib in Chinese patients with lung adenocarcinoma显示文摘Objective: The relationship between anaplastic lymphoma kinase(ALK) expression in malignant pleural effusion(MPE) samples detected only by Ventana immunohistochemistry(IHC) ALK(D5F3) and the efficacy of ALKtyrosine kinase inhibitor therapy is uncertain.Methods: Ventana anti-ALK(D5F3) rabbit monoclonal primary antibody testing was performed on 313 cell blocks of MPE samples from Chinese patients with advanced lung adenocarcinoma, and fluorescence in situ hybridization(FISH) was used to verify the ALK gene status in Ventana IHC ALK(D5F3)-positive samples. The follow-up clinical data on patients who received crizotinib treatment were recorded.Results: Of the 313 MPE samples, 27(8.6%) were confirmed as ALK expression-positive, and the Ventana IHC ALK(D5F3)-positive rate was 17.3%(27/156) in wild-type epidermal growth factor receptor(EGFR) MPE samples. Twenty-three of the 27 IHC ALK(D5F3)-positive samples were positive by FISH. Of the 11 Ventana IHC ALK(D5F3)-positive patients who received crizotinib therapy, 2 patients had complete response(CR), 5 had partial response(PR) and 3 had stable disease(SD).Conclusions: The ALK gene expression status detected by the Ventana IHC ALK(D5F3) platform in MPE samples may predict tumor responsiveness to crizotinib in Chinese patients with advanced lung adenocarcinoma.Zheng Wang Xiaonan Wu Xiaohong Han Gang Cheng Xinlin Mu Yuhui Zhang Di Cui Chang Liu Dongge Liu Yuankai Shi 2016Chinese Journal of Cancer Research2016,28,6:1

4Deformation-tolerant metal anodes for flexible sodium-air fiber batteries显示文摘Although flexible sodium-air(Na-air)batteries with high theoretical energy density offer promising opportunities for next-generation smart electronics,enhancing the safety and efficiency of flexible sodium metal anodes under dynamic and continuous deformation remains a challenge.Here,a flexible sodiated carbon nanotube layer to suppress dendrite growth under various deformations is demonstrated through a Fermi level-driven spontaneous synthetic process.The resulting sodiated carbon nanotube layer,which has a spontaneously formed solid-electrolyte interface and a robust interlocked structure,creates a uniformly distributed electric field and stable interface even under deformation,affording dendrite-free flexible Na metal anodes.With this deformation-tolerant Na metal anode,we have constructed a new family of highly flexible Na-air fiber batteries with excellent cycling performance for 400 cycles at a current density of 1000​mA·g^(−1) and a capacity limit of 500 mAh·g^(−1) under dynamic deformation.These Na-air fiber batteries can be further woven into self-powering systems to support flexible electronic devices.Lei Ye Xiangran Cheng Meng Liao Tiancheng Zhao Xinlin Huang Xinyue Kang Kun Zhang Xuemei Sun Bingjie Wang Huisheng Peng 2022eScience2022,2,6:0
5Inhibition of DNA restrictive endonucleases by aqueous nanoparticle suspension of methanophosphonate fullerene derivatives and its mechanisms显示文摘Aqueous nanoparticle suspension of fullerene and its derivatives are currently attracting much attention. To determine the effects of aqueous nanoparticle suspension of a mono-methanophosphonate fullerene and bis-methanophosphonate fullerene (denoted as n-MMPF and n-BMPF, respectively) on the activities of DNA restrictive endonucleases, plasmid pEGFP-N1 was cleaved at a single but differently restrictive site by EcoR I, BamH I, and isozymes Cfr9 I and Xma I, respectively. Both n-MMPF and n-BMPF inhibited the activity of EcoR I, while n-BMPF exhibited stronger inhibition than n-MMPF. Addition of n-BMPF into reaction mixtures inhibited the activities of all the four enzymes, and IC50 values for EcoR I, BamH I, Cfr9 I and Xma I were 4.3, >30, 11.7 and 8.3 μmol/L, respectively. When EcoR I was completely inhibited by n-BMPF, addition of excess amounts of pEGFP-N1 could not produce the product linear plasmid; however, increase of EcoR I amounts antagonized EcoR I inhibition of n-BMPF. Two scavengers of reactive oxygen species (ROS), mannitol and sodium azide at the concentrations of 2-10 mmol/L, did not reverse inhibition of n-BMPF, implying that this inhibition probably is not correlated to ROS.
