| 1 | Synthesis and characteristics of a novel artificial hapten using the copper mercaptide of penicillenic acid from penicillin G for immunoassay of heavy metal ions显示文摘In this paper, we describe the synthesis of a novel copper ion hapten using the copper mercaptide of penicillenic acid (CMPA) derived from penicillin. Results from tests with immune rabbits indicate that: (i) A new antigen synthesized with CMPA has good stability and is safe for immunizing animals with no toxic phenomena being found in animal experiments; (ii) the immunogenic antigen (CMPA-BSA) can stimulate the immune system to produce specific antibodies with high titrations, up to 150000; and (iii) antibodies in antisera showed higher affinity to OVA-GSH-CuCl than OVA-GSH, which indicates that the antibodies have specific affinity towards copper ions. | 4 | Experimental investigations on validity of moving chemical reaction boundary equation for the strong acid——weak base system with an excess of potassium chloride显示文摘A moving chemical reaction boundary (MCRB) is formed by the weak alkali NH3*H2O and strong acid HCl with an excess of potassium chloride as background electrolyte. The experimental results are compared with the predictions in accordance with the moving chemical reaction boundary equation (MCRBE). If the MCRB is cathodic, the experimental results are in agreement with the predictions by the MCRBE. However, if the MCRB is anodic, the experimental results significantly deviate from the predictions of the MCRBE. A corrected MCRB equation is proposed for such conditions. The findings are evidently different from those in our early reports(J. Chromatogr. A, 891 (2000): 337~347; 907 (2001): 347~352; 922 (2001) 283~393). | 5 | In-Depth Metaproteomics Analysis of Oral Microbiome for Lung Cancer显示文摘The human oral microbiome correlates with numerous diseases,including lung cancer.Identifying the functional changes by metaproteomics helps understand the disease-related dysbiosis,yet characterizing low-abundant bacteria is challenging.Here,we developed a free-flow isoelectric focusing electrophoresis-mass spectrometry-(FFIEF-MS-)based metaproteomics strategy to reduce host interferences and enrich low-abundant bacteria for in-depth interpretation of the oral microbiome.With our method,the number of interfering peptides decreased by 52.87%,whereas the bacterial peptides and species increased by 94.97%and 44.90%,respectively,compared to the conventional metaproteomics approach.We identified 3647 bacterial proteins,which is the most comprehensive oral metaproteomics study to date.Lung cancer-associated bacteria were validated among an independent cohort.The imbalanced Fusobacterium nucleatum and Prevotella histicola and their dysregulated functions in inhibiting immune response and maintaining cell redox homeostasis were revealed.The FFIEF-MS may serve as a valuable strategy to study the mechanisms between human diseases and microbiomes with broader applications. | Xiaoteng Jiang Yan Zhang Huiyu Wang Zeyuan Wang Shen Hu Chengxi Cao Hua Xiao | 2023 | Research2023,,1: | 0 |
