| 1 | Impact of antiviral therapy on post-hepatectomy outcome for hepatitis B-related hepatocellular carcinoma显示文摘The outcome after curative resection for hepatocellular carcinoma(HCC)remains unsatisfactory due to the high recurrence rate after surgery.In patients with hepatitis B virus(HBV)-related HCC,which is the majority of patients with HCC in Asia,a high viral load is a strong risk factor for HCC recurrence.It is logical to believe that antiviral therapy may improve the postoperative outcome by promoting viral clearance and hepatocyte regeneration,as well as improving residual liver volume in HCC patients with hepatitis B.However,the effect of antiviral therapy on clinical outcomes after liver resection in patients with HBV-related HCC remains to be established.There are two main groups of antiviral treatment for HBV-oral nucleos(t)ide analogues and interferon.Interferon treatment reduces the overall incidence of HBV-related HCC in sustained re-sponders.However,side effects may limit its long-term clinical application.Nucleos(t)ide analogues carry fewer side effects and are potent in terms of viral suppression when compared to interferon and are typically implemented for patients with more advanced liver diseases.They may also improve the outcome after curative resection for HBV-related HCC.There are increasing evidence to suggest that antiviral therapy could suppress HBV,decrease the perioperative reactivation of viral replication,reduce liver injury,preserve the liver function before and after operation,and may lower the risk of HCC recurrence.After all,antiviral therapy may improve the survival after liver resection by reducing recurrence and delaying the liver damage by the virus,resulting in a higher chance of receiving aggressive salvage therapy during HCC recurrence. | Charing Ching Ning Chong Grace Lai Hung Wong Paul Bo San Lai | 2014 | World Journal of Gastroenterology2014,20,20: | 15 |

| 3 | Cryogenic 3D printing of dual-delivery scaffolds for improved bone regeneration with enhanced vascularization显示文摘Three-dimensional(3D)printing has been increasingly employed to produce advanced bone tissue engineering scaffolds with biomimetic structures and matched mechanical strengths,in order to induce improved bone regeneration in defects with a critical size.Given that the successful bone regeneration requires both excellent osteogenesis and vascularization,endowing scaffolds with both strong bone forming ability and favorable angiogenic potential would be highly desirable to induce improved bone regeneration with required vascularization.In this investigation,customized bone tissue engineering scaffolds with balanced osteoconductivity/osteoinductivity were produced via cryogenic 3D printing ofβ-tricalcium phosphate and osteogenic peptide(OP)containing water/poly(lactic-co-glycolic acid)/dichloromethane emulsion inks.The fabricated scaffolds had a hierarchically porous structure and were mechanically comparable to human cancellous bone.Angiogenic peptide(AP)containing collagen I hydrogel was then coated on scaffold surface to further provide scaffolds with angiogenic capability.A sequential release with a quick AP release and a slow but sustained OP release was obtained for the scaffolds.Both rat endothelial cells(ECs)and rat bone marrow derived mesenchymal stem cells(MSCs)showed high viability on scaffolds.Improved in vitro migration and angiogenesis of ECs were obtained for scaffolds delivered with AP while enhanced osteogenic differentiation was observed in scaffolds containing OP.The in vivo results showed that,toward scaffolds containing both AP and OP,the quick release of AP induced obvious angiogenesis in vivo,while the sustained OP release significantly improved the new bone formation.This study provides a facile method to produce dual-delivery scaffolds to achieve multiple functions. | Chong Wang Jiahui Lai Kai Li Shaokui Zhu Bingheng Lu Jia Liu Yujin Tang Yen Wei | 2021 | Bioactive Materials2021,6,1: | 7 |
| 4 | 腹腔镜肝切除术、经皮射频消融术及传统开放手术对小肝癌的治疗比较(英文)显示文摘目的:评估比较目前三种主流技术对小肝癌的治疗作用及对患者预后的影响。创新点:首次同时比较两种手术技术和一种微创射频技术对小肝癌患者的治疗效果,并且将小肝癌患者通过年龄分层,获得最优的治疗选择,以期指导临床上更个性化及精准化的治疗方案。方法:选取2005年至2010年间94例符合单个肿瘤直径小于5 cm或不超过3个肿瘤且每个肿瘤直径小于3 cm的小肝癌患者,分至3个治疗组,分别为腹腔镜肝切除组(28例)、经皮射频消融组(33例)和开放肝切除组(33例)。经过3年的跟踪随访,统计比较3组患者的疾病预后及相关指标。结论:本研究中统计结果显示,经皮射频消融组的无瘤生存率和总生存率明显比两个手术组低(图2和3)。腹腔镜手术组和开放手术组的生存及预后情况无明显差异,而腹腔镜手术组的创伤和术后并发症率要低于开放手术组(表2)。本研究根据中心经验将全部患者以60岁进行年龄划分(图5和6)。对于60岁以下的小肝癌患者来说,手术的效果明显优于射频消融术,故应优先选择微创的腹腔镜肝切除术;60岁以上的小肝癌患者经射频治疗获得的生存结果与手术无显著差异,同时射频治疗具有创伤轻微、可重复、并发症少等优势,故可以酌情选择手术或射频治疗。 | Chong LAI Ren-an JIN Xiao LIANG Xiu-jun CAI | 2016 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2016,17,3: | 5 |

| 10 | Programmed death 1, ligand 1 and 2 correlated genes and their association with mutation, immune infiltration and clinical outcomes of hepatocellular carcinoma显示文摘BACKGROUND The exact regulation network of programmed death 1(PD-1), programmed death ligand 1(PD-L1), and programmed death ligand 2(PD-L2) signaling in immune escape is largely unknown. We aimed to describe the gene expression profiles related to PD-1 as well as its ligands PD-L1 and PD-L2, thus deciphering their possible biological processes in hepatocellular carcinoma(HCC).AIM To find the possible mechanism of function of PD-1, PD-L1, and PD-L2 in HCC.METHODS Based on the expression data of HCC from The Cancer Genome Atlas, the PD-1/PD-L1/PD-L2 related genes were screened by weighted correlation network analysis method and the biological processes of certain genes were enriched. Relation of PD1/PD-L1/PD-L2 with immune infiltration and checkpoints was investigated by co-expression analysis. The roles of PD-1/PD-L1/PD-L2 in determination of clinical outcome were also analyzed.RESULTS Mutations of calcium voltage-gated channel subunit alpha1 E, catenin beta 1, ryanodine receptor 2, tumor suppressor protein p53, and Titin altered PD-1/PDL1/PD-L2 expression profiles in HCC. PD-1, PD-L1, and PD-L2 related genes were mainly enriched in biological procedures of T cell activation, cell adhesion, and other important lymphocyte effects. In addition, PD-1/PD-L1/PD-L2 was related with immune infiltration of CD8 T cells, cytotoxic lymphocytes,fibroblasts, and myeloid dendritic cells. Immune checkpoints of CTLA4, CD27, CD80, CD86, and CD28 were significantly related to the PD-1/PD-L1/PD-L2 axis. Clinically, PD-1 and PD-L2 expression was correlated with recurrence(P = 0.005 for both), but there was no significant correlation between their expression and HCC patient survival.CONCLUSION Mutations of key genes influence PD-1, PD-L1, and PD-L2 expression. PD-1, PDL1, and PD-L2 related genes participate in T cell activation, cell adhesion, and other important lymphocyte effects. | 11 | E3 ligase FBXW7 aggravates TMPD-induced systemic lupus erythematosus by promoting cell apoptosis显示文摘Systemic lupus erythematosus(SLE)is a systemic autoimmune disease,and the pathogenesis of SLE has not been fully elucidated.The E3 ubiquitin ligase FBXW7 has been well characterized in cancer as a tumor suppressor that can promote the ubiquitination and subsequent degradation of various oncoproteins;however,the potential role of FBXW7 in autoimmune diseases is unclear.In the present study,we identified that FBXW7 is a crucial exacerbating factor for SLE development and progression in a mouse model induced by 2,6,10,14-tetramethylpentadecane(TMPD).Myeloid cell-specific FBXW7-deficient(Lysm+FBXW7f/f)C57BL/6 mice showed decreased immune complex accumulation,glomerulonephritis,glomerular mesangial cell proliferation,and basemembrane thickness in the kidney.Lysm+FBXW7f/f mice produced fewer anti-Sm/RNP and anti-ANA autoantibodies and showed a decreased MHC II expression in B cells.In Lysm+FBXW7f/f mice,we observed that cell apoptosis was reduced and that fewer CD11b+Ly6Chi inflammatory monocytes were recruited to the peritoneal cavity.Consistently,diffuse pulmonary hemorrhage(DPH)was also decreased in Lysm+FBXW7f/f mice.Mechanistically,we clarified that FBXW7 promoted TMPD-induced cell apoptosis by catalyzing MCL1 degradation through K48-linked ubiquitination.Our work revealed that FBXW7 expression in myeloid cells played a crucial role in TMPD-induced SLE progression in mice,which may provide novel ideas and theoretical support for understanding the pathogenesis of SLE. | Zhenlu Chong Chunjing Bao Jia He Tianxiao Chen Lijia Zhong Gaopeng Li Huanle Li Lutong Fang Yinjing Song Guoxiang Fu Xuyan Yang Lihua Lai Yang Liu Qingqing Wang | 2018 | Cellular & Molecular Immunology2018,15,12: | 2 |
