| 1 | Sini powder ameliorates the inflammatory response in rats with stress-induced non-alcoholic fatty liver disease by inhibiting the nuclear factor kappa-B/pyrin domain-containing protein 3 pathway显示文摘OBJECTIVE: To evaluate the effectiveness of Sini powder for the treatment of non-alcoholic fatty liver disease(NAFLD) in rats and the molecular mechanisms involved.METHODS: A rat model of stress-induced NAFLD was established by a combination of long-term tethering and feeding of a high-fat, high-calorie diet. These rats were then intragastrically administered with either simvastatin, Sini powder, or vehicle for 1 week. The body mass and field test scores for each group were recorded weekly. Serum aspartate aminotransferase and alanine aminotransferase activities, and triglyceride, total cholesterol,and free fatty acid concentrations were measured.Liver tissue histopathology was examined on hematoxylin and eosin-stained paraffin sections and oil red O-stained frozen sections. The hepatic m RNA expression of nuclear factor kappa-B(NF-κB),NOD-, LRR-, and pyrin domain-containing protein 3(NLRP3), apoptosis-associated speck-like protein containing CARD(ASC), and caspase-1 were measured by reverse transcription-polymerase chain reaction(RT-PCR). The hepatic protein concentrations of NF-κB and NLRP3, ASC, caspase-1, interleukin-1β(IL-1β), interleukin-6(IL-6), and the serum concentrations of IL-1β and IL-6 were measured by enzyme-linked immunosorbent assay.RESULTS: Compared with the Blank group, rats in the Compound model group showed significant pathologic manifestations of NAFLD, and the expression of NF-κB, NLRP3, ASC, caspase-1, IL-1βand IL-6 were significantly higher(all P < 0.01). Both simvastatin and Sini powder significantly ameliorated the NAFLD pathology and the abnormal expression of NF-κB, NLRP3, ASC, caspase-1, IL-1β, and IL-6(all P < 0.01).CONCLUSION: Sini powder inhibits the inflamma-tory response in rats with NAFLD, which is mediated by NF-κB/NLRP3, IL-1β, and IL-6, reduces the effects of psychological stress, and improves lipid metabolism. | 2 | Molecular mechanism of non-alcoholic fatty liver disease induced and aggravated by chronic stress through HSL/ATGL-FFA which promotes fat mobilization显示文摘Objective:To study the effects of social stress(CS)and social stress combined with high-fat diet on fat mobilization as a candidate mechanism for the induction or aggravation of non-alcoholic fatty liver disease(NAFLD).Methods:Thirty-two male Wistar rats(250±10 g)were randomly allocated to a blank control group(BC),a high-fat diet group(HFD),a CS group,and a combined CS and high-fat diet group(CS t HFD).Rats were sacrificed and tissues were collected after 8 weeks.Liver and body mass were measured and used to calculate the liver index.Serum aspartate aminotransferase(AST),alanine aminotransferase(ALT),and free fatty acids(FFAs)were measured.Liver sections were examined microscopically after oil red O and hematoxylin and eosin staining.The relative mRNA expression of acetyl-CoA carboxylase(ACCase),hydroxymethylglutaryl coenzyme A(HMG-CoA)reductase in liver,and hormone-sensitive lipase(HSL),and adipose triglyceride lipase(ATGL)in subcutaneous adipose tissue,were measured by real-time PCR.The liver concentrations of triglyceride,reactive oxygen species,and ACCase were measured by ELISA and HSL activity was determined using turbidimetry.Results:NAFLD developed in the CS,HFD,and CS t HFD groups,with the most severe NAFLD being in the CS t HFD group.Serum AST,ALT,and FFA,liver index,and hepatic triglyceride,FFA,and tumor necrosis factor-a were significantly higher in both the CS and CS t HFD groups.However,food intake and ACCase mRNA expression were lower.The mRNA expression of HSL and ATGL in adipose tissue was much higher,and HSL activity was higher in the CS group than in the BC group,and in the CS t HFD group than in the HFD group.Conclusion:We have successfully established two models of stress-induced NAFLD,suggesting stress can induce and aggravate NAFLD by promoting fat mobilization through upregulation of HSL and ATGL. | Jie Mu Xueqian Wang Qingguo Wang Fafeng Cheng Wenxiang Zhu Changxiang Li Chongyang Ma Changming Zhai Yajun Lian Xin Du | 2019 | Journal of Traditional Chinese Medical Sciences2019,6,4: | 2 |
| 3 | Dynamic evolution in mechanical characteristics of complex supporting structures during large section tunnel construction显示文摘The shallow tunnelling method(STM)often uses temporary supports to divide large section tunnels into several closed or semiclosed sections so as to share the upper load.The complex support system composed of primary and temporary supports can ensure safety during tunnel construction.Based on the large section tunnel of Beijing Subway Line 12,the mechanical characteristics of support system by the double-side-drift method(DSDM)during excavation and demolition were analyzed through numerical simulation and monitoring.The study showed that the middle cave excavation was the most critical stage of the DSDM,during which the load on the supporting structure increased significantly.The temporary vertical support bore most of the new load during middle cave excavation.During the demolition stage,the load was redistributed,which caused arch settlement and section convergence.The removal of the temporary vertical support exerted the greatest impact in this process.The lateral temporary inverted arch changed from axial compression to axial tension after the middle and lower caves were excavated.Based on the mechanical characteristics of the support system,some engineering suggestions were proposed for large section tunnel construction.These research results can provide reference for the design and construction of similar large section tunnels. | Hua Jiang Jianhua Mu Jinxun Zhang Yusheng Jiang Chongyang Liu Xiaoyan Zhang | 2022 | Deep Underground Science and Engineering2022,1,2: | 0 |
| 4 | Single-cell transcriptome atlas reveals spatiotemporal developmental trajectories in the basal roots of moso bamboo (Phyllostachys edulis)显示文摘Roots are essential for plant growth and development.Bamboo is a large Poaceae perennial with 1642 species worldwide.However,little is known about the transcriptional atlas that underpins root cell-type differentiation.Here,we set up a modified protocol for protoplast preparation and report single-cell transcriptomes of 14279 filtered single cells derived from the basal root tips of moso bamboo.We identified four cell types and defined new cell-type-specific marker genes for the basal root.We reconstructed the developmental trajectories of the root cap,epidermis,and ground tissues and elucidated critical factors regulating cell fate determination.According to in situ hybridization and pseudotime trajectory analysis,the root cap and epidermis originated from a common initial cell lineage,revealing the particularity of bamboo basal root development.We further identified key regulatory factors for the differentiation of these cells and indicated divergent root developmental pathways between moso bamboo and rice.Additionally,PheWOX13a and PheWOX13b ectopically expressed in Arabidopsis inhibited primary root and lateral root growth and regulated the growth and development of the root cap,which was different from WOX13 orthologs in Arabidopsis.Taken together,our results offer an important resource for investigating the mechanism of root cell differentiation and root system architecture in perennial woody species of Bambusoideae. | Zhanchao Cheng Changhong Mu Xiangyu Li Wenlong Cheng Miaomiao Cai Chongyang Wu Jutang Jiang Hui Fang Yucong Bai Huifang Zheng Ruiman Geng Junlei Xu Yali Xie Yuping Dou Juan Li Shaohua Mu Jian Gao | 2023 | Horticulture Research2023,10,8: | 0 |
| 5 | Distance-basedα-amylase biosensor fabricated with amylopectin-coated mesoporous membrane显示文摘Paper-based biosensors are widely employed in point-of-care testing(POCT)due to their convenience,portability,low cost,and ease of use.This study reports an integrated distance-based paper biosensor fabricated with a mesoporous membrane coated with stimuli-responsive polymer.The detection ofα-amylase(AMY)using amylopectin-coated mesoporous membrane is demonstrated as an example.After introducing the AMY solution,it is observed that the aqueous solution flows along the paper strip due to AMY-catalyzed hydrolysis of amylopectin.The flow distance is proportional to the concentration of AMY with a detection limit as low as 4 mU/mL.In addition,the detection of AMY is demonstrated in human serum.Furthermore,the inhibitory effect of acarbose on AMY is evaluated.This reagent-free and disposable biosensor allows single-step rapid detection of the analyte.This approach is very promising for the development of user-friendly,equipment-free,and cost-effective biosensors with remarkable sensitivity and excellent selectivity for disease diagnosis and hypoglycemic drug screening. | Binglu Zhao Mashooq Khan Yulin Liu Wenjun Tai Chongyang Mu Wenli Wu Mei Zhao Yaohong Ma Li Yu Jin-Ming Lin Qiongzheng Hu | 2024 | Chinese Chemical Letters2024,35,1: | 0 |
