| 1 | Sini powder ameliorates the inflammatory response in rats with stress-induced non-alcoholic fatty liver disease by inhibiting the nuclear factor kappa-B/pyrin domain-containing protein 3 pathway显示文摘OBJECTIVE: To evaluate the effectiveness of Sini powder for the treatment of non-alcoholic fatty liver disease(NAFLD) in rats and the molecular mechanisms involved.METHODS: A rat model of stress-induced NAFLD was established by a combination of long-term tethering and feeding of a high-fat, high-calorie diet. These rats were then intragastrically administered with either simvastatin, Sini powder, or vehicle for 1 week. The body mass and field test scores for each group were recorded weekly. Serum aspartate aminotransferase and alanine aminotransferase activities, and triglyceride, total cholesterol,and free fatty acid concentrations were measured.Liver tissue histopathology was examined on hematoxylin and eosin-stained paraffin sections and oil red O-stained frozen sections. The hepatic m RNA expression of nuclear factor kappa-B(NF-κB),NOD-, LRR-, and pyrin domain-containing protein 3(NLRP3), apoptosis-associated speck-like protein containing CARD(ASC), and caspase-1 were measured by reverse transcription-polymerase chain reaction(RT-PCR). The hepatic protein concentrations of NF-κB and NLRP3, ASC, caspase-1, interleukin-1β(IL-1β), interleukin-6(IL-6), and the serum concentrations of IL-1β and IL-6 were measured by enzyme-linked immunosorbent assay.RESULTS: Compared with the Blank group, rats in the Compound model group showed significant pathologic manifestations of NAFLD, and the expression of NF-κB, NLRP3, ASC, caspase-1, IL-1βand IL-6 were significantly higher(all P < 0.01). Both simvastatin and Sini powder significantly ameliorated the NAFLD pathology and the abnormal expression of NF-κB, NLRP3, ASC, caspase-1, IL-1β, and IL-6(all P < 0.01).CONCLUSION: Sini powder inhibits the inflamma-tory response in rats with NAFLD, which is mediated by NF-κB/NLRP3, IL-1β, and IL-6, reduces the effects of psychological stress, and improves lipid metabolism. | 2 | Design Bistatic Interferometric DEM Generation Algorithm and Its Theoretical Accuracy Analysis for LuTan-1 Satellites显示文摘LuTan-1(LT-1)is a constellation with two full-polarization L-band radar satellites designed by China,and the first satellite was scheduled to be launched in December 2021 and the second one in January 2022.The LT-1 will be operated for deformation monitoring in repeat-pass mode,and for DEM generation in bistatic mode,improving self-sufficiency of SAR data for the field of geology,earthquake,disaster reduction,geomatics,forestry and so on.In this paper,we focused on designing an algorithm for interferometric DEM generation using LT-1 bistatic satellites.The basic principle,main error sources and errors control of the DEM generation algorithm of LT-1 were systematically analyzed.The experiment results demonstrated that:①The implemented algorithm had rigorous resolution with a theoretic accuracy better than 0.03 m for DEM generation.②The errors in satellite velocity and Doppler centroid had no obvious effect on DEM accuracy and they could be neglected.While the errors in position,baseline,slant range and interferometric phase had a significant effect on DEM accuracy.And the DEM error caused by baseline error was dominated,followed by the slant range error,interferometric phase error and satellite position error.③To obtain an expected DEM accuracy of 2 m,the baseline error must be strictly controlled and its accuracy shall be 1.0 mm or better for Cross-Track and Normal-Direction component,respectively.And the slant range error and interferometric phase error shall be reasonably controlled.The research results were of great significance for accurately grasping the accuracy of LT-1 data products and their errors control,and could provide a scientific auxiliary basis for LT-1 in promoting global SAR technology progress and the generation of high-precision basic geographic data. | Bing XU Liqun LIU Zhiwei LI Yan ZHU Jingxin HOU Wenxiang MAO | 2022 | Journal of Geodesy and Geoinformation Science2022,5,1: | 6 |

| 4 | Characterization and validation of somatic mutation spectrum to reveal heterogeneity in gastric cancer by single cell sequencing显示文摘Gastric cancer(GC) is a highly heterogeneous disease with multiple cellular types and poor prognosis.However, the cellular evolution and molecular basis of GC at the individual intra-tumor level has not been well demonstrated. We performed single-cell whole exome sequencing to detect somatic singlenucleotide variants(SNVs) and significantly mutated genes(SMGs) among 34 tumor cells and 9 normal cells from a patient with GC. The Complete Prediction for Protein Conformation(CPPC) approach directly predicting the folding conformation of the protein 3D structure with Protein Folding Shape Code, combined with functional experiments were used to confirm the characterization of mutated SMGs in GC cells. We identified 201 somatic SNVs, including 117 non-synonymous mutations in GC cells. Further analysis identified 24 significant mutated genes(SMGs) in single cells, for which a single amino acid change might affect protein conformation. Among them, two genes(CDC27 and FLG) that were mutated only in single cells but not in the corresponding tumor tissue, were recurrently present in another GC tissue cohort, and may play a potential role to promote carcinogenesis, as confirmed by functional characterization. | 6 | Molecular mechanism of non-alcoholic fatty liver disease induced and aggravated by chronic stress through HSL/ATGL-FFA which promotes fat mobilization显示文摘Objective:To study the effects of social stress(CS)and social stress combined with high-fat diet on fat mobilization as a candidate mechanism for the induction or aggravation of non-alcoholic fatty liver disease(NAFLD).Methods:Thirty-two male Wistar rats(250±10 g)were randomly allocated to a blank control group(BC),a high-fat diet group(HFD),a CS group,and a combined CS and high-fat diet group(CS t HFD).Rats were sacrificed and tissues were collected after 8 weeks.Liver and body mass were measured and used to calculate the liver index.Serum aspartate aminotransferase(AST),alanine aminotransferase(ALT),and free fatty acids(FFAs)were measured.Liver sections were examined microscopically after oil red O and hematoxylin and eosin staining.The relative mRNA expression of acetyl-CoA carboxylase(ACCase),hydroxymethylglutaryl coenzyme A(HMG-CoA)reductase in liver,and hormone-sensitive lipase(HSL),and adipose triglyceride lipase(ATGL)in subcutaneous adipose tissue,were measured by real-time PCR.The liver concentrations of triglyceride,reactive oxygen species,and ACCase were measured by ELISA and HSL activity was determined using turbidimetry.Results:NAFLD developed in the CS,HFD,and CS t HFD groups,with the most severe NAFLD being in the CS t HFD group.Serum AST,ALT,and FFA,liver index,and hepatic triglyceride,FFA,and tumor necrosis factor-a were significantly higher in both the CS and CS t HFD groups.However,food intake and ACCase mRNA expression were lower.The mRNA expression of HSL and ATGL in adipose tissue was much higher,and HSL activity was higher in the CS group than in the BC group,and in the CS t HFD group than in the HFD group.Conclusion:We have successfully established two models of stress-induced NAFLD,suggesting stress can induce and aggravate NAFLD by promoting fat mobilization through upregulation of HSL and ATGL. | Jie Mu Xueqian Wang Qingguo Wang Fafeng Cheng Wenxiang Zhu Changxiang Li Chongyang Ma Changming Zhai Yajun Lian Xin Du | 2019 | Journal of Traditional Chinese Medical Sciences2019,6,4: | 2 |

| 13 | Energy-efficient cell-association bias adjustment algorithm for ultra-dense networks显示文摘In recent years, energy efficiency has become an important topic, especially in the field of ultradense networks(UDNs). In this area, cell-association bias adjustment and small cell on/off are proposed to enhance the performance of energy efficiency in UDNs. This is done by changing the cell association relationship and turning off the extra small cells that have no users. However, the variety of cell association relationships and the switching on/off of the small cells may deteriorate some users' data rates, leading to nonconformance to the users' data rate requirement. Considering the discreteness and non-convexity of the energy efficiency optimization problem and the coupled relationship between cell association and scheduling during the optimization process, it is difficult to achieve an optimal cell-association bias. In this study, we optimize the network energy efficiency by adjusting the cell-association bias of small cells while satisfying the users' data rate requirement. We propose an energy-efficient centralized Gibbs sampling based cell-association bias adjustment(CGSCA) algorithm. In CGSCA, global information such as channel state information, cell association information, and network load information need to be collected. Then, considering the overhead of the messages that are exchanged and the implementation complexity of CGSCA to obtain the global information in UDNs, we propose an energy-efficient distributed Gibbs sampling based cell-association bias adjustment(DGSCA) algorithm with a lower message-exchange overhead and implementation complexity.Using DGSCA, we derive the updated formulas for calculating the number of users in a cell and the users' SINR. We analyze the implementation complexities(e.g., computation complexity and communication complexity) of the proposed two algorithms and other existing algorithms. We perform simulations, and the results show that CGSCA and DGSCA have faster convergence speed, as well as a higher performance gain of the energy efficiency and throughput compared to other existing algorithms. In addition, we analyze the importance of the users' data rate constraint in optimizing the energy efficiency, and we compare the energy efficiency performance of different algorithms with different number of small cells.
