2Icariside Ⅱ inhibits lipopolysaccharide-induced inflammation and amyloid production in rat astrocytes by regulating IKK/IκB/NF-κB/BACE1 signaling pathway显示文摘β-amyloid(Aβ)is one of the inducing factors of astrocytes activation and neuroinflammation,and it is also a crucial factor for the development of Alzheimer’s disease(AD).Icariside II(ICS II)is an active component isolated from a traditional Chinese herb Epimedium,which has shown to attnuate lipopolysaccharide(LPS)-induced neuroinflammation through regulation of NF-κB signaling pathway.In this study we investigated the effects of ICS II on LPS-induced astrocytes activation and Aβaccumulation.Primary rat astrocytes were pretreated with ICS II(5,10,and 20μM)or dexamethasone(DXMS,1μM)for 1 h,thereafter,treated with LPS for another 24 h.We found that ICS II pretreatment dose dependently mitigated the levels of tumor necrosis factor-alpha(TNF-α),interleukin-1 beta(IL-1β),inducible nitric oxide synthase(iNOS),cyclooxygenase-2(COX-2)in the astrocytes.Moreover,ICS II not only exerted the inhibitory effect on LPS-induced IκB-αdegradation and NF-κB activation,but also decreased the levels of Aβ1–40,Aβ1–42,amyloid precursor protein(APP)and beta secretase 1(BACE1)in the astrocytes.Interestingly,molecular docking revealed that ICS II might directly bind to BACE1.It is concluded that ICS II has potential value as a new therapeutic agent to treat neuroinflammation-related diseases,such as AD.Yong Zheng Yan Deng Jian-mei Gao Chun Lv Ling-hu Lang Jing-shan Shi Chang-yin Yu Qi-hai Gong 2020Acta Pharmacologica Sinica2020,41,2:8

5Towards context consistency by concurrent checking for Internetware applications显示文摘Internetware applications are emerging and being widely used.They can adapt their behavior based on environmental contexts and deliver smart services.These contexts can be subject to various noises,which cause them to be inaccurate,incomplete,or even to conflict with each other.This is known as context inconsistency problem.Context inconsistency can trigger unexpected behavior to applications,and therefore should be prevented.One promising approach is to check contexts against consistency constraints so as to detect the occurrences of context inconsistency at runtime.Existing techniques have attempted different ways to improve the checking efficiency or effectiveness with different trade-offs in space overhead or communication cost.However,none of them has exploited multi-core computing capability to systematically improve the checking efficiency.In this paper,we propose a novel concurrent checking technique Con-C to efficiently detect inconsistencies in huge volumes of dynamic contexts.Con-C derives checking subtasks for each consistency constraint based on its structure and semantics.It achieves this in a fully automated way,and at the same time can guarantee its derived checking subtasks to be persistently balanced.We evaluated Con-C by controlled experiments through a large-scale real-world application.It reported promising results that Con-C improved the checking efficiency by extra 57.0%,in addition to what had been gained by incremental checking.XU Chang LIU YePang CHEUNG S.C. 6Abrogation of Hn RNP L enhances anti-PD-1 therapy efficacy via diminishing PD-L1 and promoting CD8^(+) T cell-mediated ferroptosis in castration-resistant prostate cancer显示文摘Owing to incurable castration-resistant prostate cancer(CRPC)ultimately developing after treating with androgen deprivation therapy(ADT),it is vital to devise new therapeutic strategies to treat CRPC.Treatments that target programmed cell death protein 1(PD-1)and programmed death ligand-1(PD-L1)have been approved for human cancers with clinical benefit.However,many patients,especially prostate cancer,fail to respond to anti-PD-1/PD-L1 treatment,so it is an urgent need to seek a support strategy for improving the traditional PD-1/PD-L1 targeting immunotherapy.In the present study,analyzing the data from our prostate cancer tissue microarray,we found that PD-L1 expression was positively correlated with the expression of heterogeneous nuclear ribonucleoprotein L(Hn RNP L).Hence,we further investigated the potential role of Hn RNP L on the PD-L1 expression,the sensitivity of cancer cells to T-cell killing and the synergistic effect with anti-PD-1 therapy in CRPC.Indeed,Hn RNP L knockdown effectively decreased PD-L1 expression and recovered the sensitivity of cancer cells to T-cell killing in vitro and in vivo,on the contrary,Hn RNP L overexpression led to the opposite effect in CRPC cells.In addition,consistent with the previous study,we revealed that ferroptosis played a critical role in T-cell-induced cancer cell death,and Hn RNP L promoted the cancer immune escape partly through targeting YY1/PD-L1 axis and inhibiting ferroptosis in CRPC cells.Furthermore,Hn RNP L knockdown enhanced antitumor immunity by recruiting infiltrating CD8^(+)T cells and synergized with anti-PD-1 therapy in CRPC tumors.This study provided biological evidence that Hn RNP L knockdown might be a novel therapeutic agent in PD-L1/PD-1 blockade strategy that enhanced anti-tumor immune response in CRPC.Xumin Zhou Libin Zou Hangyu Liao Junqi Luo Taowei Yang Jun Wu Wenbin Chen Kaihui Wu Shengren Cen Daojun Lv Fangpeng Shu Yu Yang Chun Li Bingkun Li Xiangming Mao 2022Acta Pharmaceutica Sinica B2022,12,2:4

9O-glycosyltransferases from Homo sapiens contributes to the biosynthesis of Glycyrrhetic Acid 3-O-mono-β-D-glucuronide and Glycyrrhizin in Saccharomyces cerevisiae显示文摘Glycyrrhizin(GL)and Glycyrrhetic Acid 3-O-mono-β-D-glucuronide(GAMG)are the typical triterpenoid glycosides found in the root of licorice,a popular medicinal plant that exhibits diverse physiological effects and pharmacological manifestations.However,only few reports are available on the glycosylation enzymes involved in the biosynthesis of these valuable compounds with low conversion yield so far.In mammals,glycosyltransferases are involved in the phase II metabolism and may provide new solutions for us to engineer microbial strains to produce high valued compounds due to the substrate promiscuity of these glycosyltransferases.In this study,we mined the genomic databases of mammals and evaluated 22 candidate genes of O-glycosyltransferases by analyzing their catalytic potential for O-glycosylation of the native substrate,glycyrrhetinic acid(GA)for its glycodiversification.Out of 22 selected glycosyltransferases,only UGT1A1 exhibited high catalytic performance for biosynthesis of the key licorice compounds GL and GAMG.Molecular docking results proposed that the enzymatic activity of UGT1A1 was likely owing to the stable hydrogen bonding interactions and favorite conformations between the amino acid residues around substrate channels(P82~R85)and substrates.Furthermore,the complete biosynthesis pathway of GL was reconstructed in Saccharomyces cerevisiae for the first time,resulting in the production of 5.98±0.47 mg/L and 2.31±0.21 mg/L of GL and GAMG,respectively.Ke Xu Yu-jia Zhao Nadeem Ahmad Jing-nan Wang Bo Lv Ying Wang Jun Ge Chun Li 2021Synthetic and Systems Biotechnology2021,6,3:3
10Infection with severe fever with thrombocytopenia virus in healthy population:a cohort study in a high endemic region,China显示文摘Background: Severe fever with thrombocytopenia (SFTS) caused by SFTS virus (SFTSV) was a tick-borne hemorrhagic fever that posed significant threat to human health in Eastern Asia. The study was designed to measure the seroprevalence of SFTSV antibody in healthy population residing in a high endemic region.Methods: A cohort study was performed on healthy residents in Shangcheng County in Xinyang City from April to December in 2018, where the highest SFTS incidence in China was reported. Anti-SFTSV IgG was measured by indirect enzyme-linked immunosorbent assay and neutralizing antibody (NAb) was detected by using PRNT50. The logistic regression models were performed to analyze the variables that were associated with seropositive rates.Results: Totally 886 individuals were recruited. The baseline seroprevalence that was tested before the epidemic season was 11.9% (70/587) for IgG and 6.8% (40/587) for NAb, which was increased to 13.4% (47/350) and 7.7% (27/350) during the epidemic season, and further to 15.8% (80/508) and 9.8% (50/508) post epidemic. The IgG antibody-based seropositivity was significantly related to the patients aged ≥ 70 years old [adjusted odds ratio (OR) = 2.440, 95% confidence interval (CI): 1.334-4.461 compared to the group of < 50 years old,P = 0.004], recent contact with cats (adjustedOR = 2.195, 95%CI: 1.261-3.818,P = 0.005), and working in tea garden (adjustedOR = 1.698, 95%CI: 1.002-2.880,P = 0.049) by applying multivariate logistic regression model. The NAb based seropositivity was similarly related to the patients aged ≥ 70 years old (adjustedOR = 2.691, 95%CI: 1.271-5.695 compared to the group of < 50 years old,P = 0.010), and recent contact with cats (OR = 2.648, 95%CI: 1.419-4.941,P = 0.002). For a cohort of individuals continually sampled with 1-year apart, the anti-SFTSV IgG were maintained at a stable level, while the NAb level reduced.Conclusions: Subclinical infection might not provide adequate immunity to protect reinfection of SFTSV, thus highlighting the ongoing threats of SFTS in endemic regions, which called for an imperative need for vaccine development. 12Characteristics of the quasi-16-day wave in the mesosphere and lower thermosphere region as revealed by meteor radar,Aura satellite,and MERRA2 reanalysis data from 2008 to 2017显示文摘This study presents an analysis of the quasi-16-day wave(Q16DW)at three stations in the middle latitudes by using a meteor radar chain in conjunction with Aura Microwave Limb Sounder temperature data and MERRA2(Modern-Era Retrospective Analysis for Research and Applications,Version 2)reanalysis data from 2008 to 2017.The radar chain consists of three meteor radar stations located at Mohe(MH,53.5°N,122.3°E),Beijing(BJ,40.3°N,116.2°E),and Wuhan(WH,30.5°N,114.6°E).The Q16DW wave exhibits similar seasonal variation in the neutral wind and temperature,and the Q16DW amplitude is generally strong during winter and weak around summer.The Q16DW at BJ was found to have secondary enhancement around September in the zonal wind,which is rarely reported at similar latitudes.The latitudinal variations of the Q16DW in the neutral wind and temperature are quite different.The Q16DW at BJ is the most prominent in both neutral wind components among the three stations and the Q16DW amplitudes at MH and WH are comparable,whereas the wave amplitude in temperature decreases with decreasing latitude.The quasi-geostrophic refractive index squared at the three stations in the period from 2008 to 2017 was revealed.The results indicate that the Q16DW in the mesosphere and lower thermosphere(MLT)at MH has a limited contribution from the lower atmosphere.Around March and October,the Q16DW in the troposphere at BJ can propagate upward into the MLT region,whereas at WH,the contribution to the Q16DW in the MLT region is largely from the mesosphere.Yun Gong Zheng Ma Chun Li XieDong Lv ShaoDong Zhang QiHou Zhou ChunMing Huang KaiMing Huang You Yu GuoZhu Li 2020Earth and Planetary Physics2020,4,3:2

15Warm ischemia time and elevated serum uric acid are associated with metabolic syndrome after liver transplantation with donation after cardiac death显示文摘AIM To describe the prevalence of posttransplant metabolic syndrome(PTMS) after donation after cardiac death(DCD) liver transplantation(LT) and the pre-and postoperative risk factors.METHODS One hundred and forty-seven subjects who underwent DCD LT from January 2012 to February 2016 were enrolled in this study. The demographics and the clinical characteristics of pre-and post-transplantation were collected for both recipients and donors. PTMS was defined according to the 2004 Adult Treatment Panel-Ⅲ criteria. All subjects were followed monthly for the initial 6 mo after discharge, and then, every 3 mo for 2 years. The subjects were followed every 6 mo or as required after 2 years post-LT.RESULTS The prevalence of PTMS after DCD donor orthotopic LT was 20/147(13.6%). Recipient's body mass index(P = 0.024), warm ischemia time(WIT)(P = 0.045), and posttransplant hyperuricemia(P = 0.001) were significantly associated with PTMS. The change in serum uric acid levels in PTMS patients was significantly higher than that in non-PTMS patients(P < 0.001). After the 1 s t mo, the level of serum uric acid of PTMS patients rose continually over a period, while it was unaltered in non-PTMS patients. After transplantation, the level of serum uric acid in PTMS patients was not associated with renal function.CONCLUSION PTMS could occur at early stage after DCD LT with growing morbidity with the passage of time. WIT and post-LT hyperuricemia are associated with the prevalence of PTMS. 20Amisulpride augmentation therapy improves cognitive performance and psychopathology in clozapine‑resistant treatment‑refractory schizophrenia:a 12‑week randomized,double‑blind,placebo‑controlled trial显示文摘Background:Although clozapine is an effective option for treatment-resistant schizophrenia(TRS),there are still 1/3 to 1/2 of TRS patients who do not respond to clozapine.The main purpose of this randomized,double-blind,placebocontrolled trial was to explore the amisulpride augmentation efficacy on the psychopathological symptoms and cognitive function of clozapine-resistant treatment-refractory schizophrenia(CTRS)patients.Methods:A total of 80 patients were recruited and randomly assigned to receive initial clozapine plus amisulpride(amisulpride group)or clozapine plus placebo(placebo group).Positive and Negative Syndrome Scale(PANSS),Scale for the Assessment of Negative Symptoms(SANS),Clinical Global Impression(CGI)scale scores,Repeatable Battery for the Assessment of Neuropsychological Status(RBANS),Treatment Emergent Symptom Scale(TESS),laboratory measurements,and electrocardiograms(ECG)were performed at baseline,week 6,and week 12.Results:Compared with the placebo group,amisulpride group had a lower PANSS total score,positive subscore,and general psychopathology subscore at week 6 and week 12(PBonferroni<0.01).Furthermore,compared with the placebo group,the amisulpride group showed an improved RBANS language score at week 12(PBonferroni<0.001).Amisulpride group had a higher treatment response rate(P=0.04),lower scores of CGI severity and CGI efficacy at week 6 and week 12 than placebo group(PBonferroni<0.05).There were no differences between the groups in body mass index(BMI),corrected QT(QTc)intervals,and laboratory measurements.This study demonstrates that amisulpride augmentation therapy can safely improve the psychiatric symptoms and cognitive performance of CTRS patients.Ming‑Huan Zhu Zhen‑Jing Liu Qiong‑Yue Hu Jia‑Yu Yang Ying Jin Na Zhu Ying Huang Dian‑Hong Shi Min‑Jia Liu Hong‑Yang Tan Lei Zhao Qin‑Yu Lv Zheng‑Hui Yi Feng‑Chun Wu Ze‑Zhi Li 2023Military Medical Research2023,10,4:1
