1An update: the role of Nephrin inside and outside the kidney显示文摘Nephrin is a key molecule in podocytes to maintain normal slit diaphragm structure. Nephin interacts with many other podocyte and slit diaphragm protein and also mediates important cell signaling pathways in podocytes. Loss of nephrin during the development leads to the congenital nephrotic syndrome in children. Reduction of nephrin expression is often observed in adult kidney diseases including diabetic nephropathy and HIV-associated nephropathy. The critical role of nephrin has been confirmed by different animal models with nephrin knockout and knockdown. Recent studies demonstrate that knockdown of nephrin expression in adult mice aggravates the progression of unilateral nephrectomy and Adriamycin-induced kidney disease. In addition to its critical role in maintaining normal glomerular filtration unit in the kidney, nephrin is also expressed in other organs. However, the exact role of nephrin in kidney and extra-renal organs has not been well characterized. 2AGE与RAGE相互作用通过活性氧引起足细胞凋亡显示文摘目的:探讨晚期糖基化终末产物(AGE)对足细胞凋亡的影响,及氧化应激在其中的作用。方法:小鼠足细胞株由美国纽约西奈山医学院Peter Mundel教授馈赠。用钙磷脂结合蛋白Ⅴ-荧光异硫氰酸盐(FITC)和碘化物(PI)标记细胞,采用荧光激活细胞分类(FACS)法来计数凋亡和坏死的足细胞。Dharmacon On TargetPlus SMARTpool si RNA试剂和Amaxa RNAi nucleofection试剂盒成功转染si RNA到足细胞。绿荧光蛋白载体证明转染的有效性,分别采用Western Blot和实时定量PCR(RT-PCR)方法来检测si RNA转染足细胞后AGE受体蛋白(RAGE)靶基因蛋白质和mRNA的表达。用LS50B型荧光分光光度计测活性氧,根据波长485nm在530nm发射的荧光来判断活性氧(ROS)的产生。观察活性氧的清除剂N-乙酰基-半胱氨酸(NAC)能否减少AGE-BSA诱导的足细胞凋亡。结果:AGE引起足细胞的凋亡呈剂量依赖性,随AGE浓度的增大,凋亡的发生率逐渐升高;RAGE siRNA能减少60%~70%RAGE mRNA和蛋白质的表达;ROS的清除剂NAC可明显减少AGE-BSA引起的ROS的产生和足细胞凋亡。结论:AGE与RAGE作用后活性氧产生增加,活性氧的增加可能是AGE引起足细胞凋亡的途径之一,可通过抗氧化减少ROS的产生延缓糖尿病肾病的进展。刘智辉 于青 袁伟杰 Jone Cijiang He 2008中国中西医结合肾病杂志2008,9,10:5

4Correlates of the School-Based Guidance Program to Freshman Students in Guangdong Province,China:Basis for Enhanced Guidance Program显示文摘Guidance programs have significant roles in preparing students for the future.The study recognizes that guidance and counseling programs are a transformative process of helping students strengthen their career planning,education,and preparation for employment.The guidance programs of first-year college students from Guangdong Peizheng College(GPC)in Guangdong Province,China was investigated,with 357 freshman students currently enrolled in the participating school involved in this study.Moreover,12 guidance counselors were interviewed through focused-group discussions,including the school director,for the qualitative component of the study.Utilizing the convergent parallel design mixed method,the study was able to elucidate the quantitative and qualitative relevance of the data gathered from the selected participants.The emerging results found that Chinese freshman students were satisfied with all of the parameters of the guidance programs of the school.The school counselors are able to provide students with a rich reservoir of academic cand guidance support,career mentoring,and career guidance.There were contributing factors that need to be addressed if improvement and innovation of the guidance program is the primary goal.Support and resources should be prioritized in order to enrich the development not only of the students but also of the guidance counselors.The study serves as empirical evidence for proposing an improved school-based guidance program as practical outputs of the study further to innovate the effectiveness of the implementation of guidance services.Cijiang Xiong 2022Journal of Contemporary Educational Research2022,6,9:0
