| 1 | Biological imaging without autofluorescence in the second near-infrared region显示文摘Fluorescence imaging is capable of acquiring anatomical and functional infor- mation with high spatial and temporal resolution. This imaging technique has been indispensable in biological research and disease detection/diagnosis. Imaging in the visible and to a lesser degree, in the near-infrared (NIR) regions below 900 nm, suffers from autofluorescence arising from endogenous fluorescent molecules in biological tissues. This autofluorescence interferes with fluorescent molecules of interest, causing a high background and low detection sensitivity. Here, we report that fluorescence imaging in the 1,500-1,700-nm region (termed "NIR-IIb") under 808-nm excitation results in nearly zero tissue autofluorescence, allowing for background-free imaging of fluorescent species in otherwise notoriously autofluorescent biological tissues, including liver. Imaging of the intrinsic fluorescence of individual fluorophores, such as a single carbon nanotube, can be readily achieved with high sensitivity and without autofluorescence background in mouse liver within the 1,500-1,700-nm wavelength region. | Shuo Diao Guosong Hong Alexander L. | 2 | Performance Evaluation of Urban Comprehensive Carrying Capacity of Harbin, Heilongjiang Province in China显示文摘Urban comprehensive carrying capacity is an important guarantee and external representation of regional sustainable development. Based on urban comprehensive carrying capacity, this paper constructed a performance evaluation index system of urban comprehensive carrying capacity, and used entropy method, urban comprehensive carrying capacity measurement model and urban sustainable development model to measure spatial and temporal comprehensive carrying capacity of Harbin City in Heilongjiang Province from 2012 to 2017. The results show that: 1) index weight analysis suggested the regional development mode in Harbin still followed an epitaxial development mode, which pursued the expansion of scale and the growth of total amount of regional development, neglecting the effective utilization of resources and the improvement of structural benefits. 2) In the pressure system, the index of resource support has dropped sharply. The index of environmental capacity and social progress has risen circuitously, while the degree of agglomeration and the value of transportation facilities have risen steadily;in the pressure system, the index of population development and economic growth tended to fluctuate, while energy consumption and environmental pollution showed a more synchronous change in characteristics, and the livable demand remained at a high level. 3) The carrying capacity index of resources and environment in Harbin has been declining, and the acquired carrying capacity index fluctuated. The long-term regional development model has severely impaired the carrying capacity of resources and environment. 4) The comprehensive carrying capacity of Harbin has clear spatial differentiation characteristics. | 3 | Identification of blast-resistance loci through genome-wide association analysis in foxtail millet(Setaria italica (L.)Beauv.)显示文摘Blast disease caused by the fungus Magnaporthe grisea results in significant yield losses of cereal crops across the world.To date,very few regulatory genes contributing to blast resistance in grass species have been identified and the genetic basis of blast resistance in cereals remains elusive.Here,a core collection of foxtail millet(Setaria italica)containing 888 accessions was evaluated through inoculation with the blast strain HN-1 and a genome-wide association study(GWAS)was performed to detect regulators responsible for blast disease resistance in foxtail millet.The phenotypic variation of foxtail millet accessions inoculated with the blast strain HN-1 indicated that less than 1.60% of the samples were highly resistant,35.25% were moderately resistant,57.09% were moderately susceptible,and 6.08% were highly susceptible.The geographical pattern of blast-resistant samples revealed that a high proportion of resistant accessions were located in lower latitude regions where the foxtail millet growing season has higher rain precipitation.Using 720000 SNP markers covering the Setaria genome,GWAS showed that two genomic loci from chromosomes 2 and 9 were significantly associated with blast disease resistance in foxtail millet.Finally,eight putative genes were identified using rice blast-related transcriptomic data.The results of this work lay a foundation for the foxtail millet blast resistance biology and provide guidance for breeding practices in this promising crop species and other cereals. | LI Zhi-jiang JIA Guan-qing LI Xiang-yu LI Yi-chu ZHI Hui TANG Sha MA Jin-feng ZHANG Shuo LI Yan-dong SHANG Zhong-lin DIAO Xian-min | 2021 | Journal of Integrative Agriculture2021,20,8: | 2 |
| 4 | Effects of dietary amylose to amylopectin ratio on growth performance,carcass quality characteristics and meat fatty acids in Chinese Qinchuan cattle显示文摘This study evaluated the effects of the dietary starch amylose/amylopectin ratio on growth performance,rumen fermentation and blood parameters,carcass characteristics and marbling score,and meat fatty acid profiles and transcriptional changes in the genes involved in the gluconeogenesis pathway in Chinese Qinchuan cattle.Forty-five cattle were randomly divided into three groups.The bulls were fed a control diet(middle amylose/amylopectin ratio=0.47)or diets with either waxy corn starch(WS)inclusion(low amylose/amylopectin ratio=0.23)or pea starch(PS)inclusion(high amylose/amylopectin ratio=0.60)for 90 days.The bulls were individually allowed to receive the diets at the daily amount of at least 2.1%of their individual BW twice daily.The bulls were weighed at 45-day intervals,and blood and rumen fluid samples were also collected at 45-day intervals.The bulls were slaughtered and longissimus thoracis(LT)samples were collected for meat quality measurements.Compared with PS,the average daily gain(ADG)and feed efficiency in control and WS were increased(P<0.05)during d 0 to 45.However,the feed efficiency in PS was increased(P<0.05)compared with the control and WS during d 46 to 90.During the whole feeding trial,the ADG in control and WS showed trends that were higher(0.05 0.05)among the three groups.Compared with PS,the serum insulin concentration in WS was increased(P<0.05)at d 45,but not at d 90.Compared with control and WS,the serum SOD and T-AOC concentrations in PS were increased(P<0.05)at d 45,but not at d 90.Compared with control,the rumen microbial crude protein contents in WS and PS were increased(P<0.05)at d 0,while those of control and WS were higher(P<0.05)than that of PS at d 45,but not at d 90.Compared with PS,the backfat thickness in control and WS were increased(P<0.05),however,the marbling score and expression of genes related to the gluconeogenesis pathway in liver of the three groups did not differ(P>0.05).Chemical and physio-chemical compositions of LT did not differ(P>0.05)among the three groups.In conclusion,diets with low or middle amylose/amylopectin ratios increased the backfat thickness,and tended to improve ADG,but had no effect on other carcass characteristics,fat content in the LT,or the expression of hepatic genes related to the gluconeogenesis pathway in Chinese Qinchuan cattle. | PIAO Min-yu HU Feng-ming KONG Fan-lin LIU Yun-long WANG Shuo CUI Kai SUN Tao DIAO Qi-yu TU Yan | 2021 | Journal of Integrative Agriculture2021,20,12: | 1 |

| 7 | Sulfation predominates the pharmacokinetics, metabolism, and excretion of forsythin in humans: major enzymes and transporters identified显示文摘Forsythin extracted from Forsythiae Fructus is widely used to treat fever caused by the common cold or influenza in China,Japan and Korea.The present study aimed to analyze the pharmacokinetics,metabolism and excretion routes of forsythin in humans and determine the major enzymes and transporters involved in these processes.After a single oral administration,forsythin underwent extensive metabolism via hydrolysis and further sulfation.In total,3 of the 13 metabolites were confirmed by comparison to reference substances,i.e.,aglycone M1,M1 sulfate(M2),and M1 glucuronide(M7).Hydrolysis was the initial and main metabolic pathway of the parent compound,followed by extensive sulfation to form M2 and a reduced level of glucuronidation to form M7.In addition,the plasma exposure of M2 and M7 were 86-and 4.2-fold higher than that of forsythin.Within 48 h,~75.1%of the administered dose was found in urine,with M2 accounting for 71.6%.Further phenotyping experiments revealed that sulfotransferase 1A1 and UDP-glucuronosyltransferase 1A8 were the most active hepatic enzymes involved in the formation of M2 and M7,respectively.The in vitro kinetic study provided direct evidence that M1 showed a preference for sulfation.Sulfated conjugate M2 was identified as a specific substrate of organic anion transporter 3,which could facilitate the renal excretion of M2.Altogether,our study demonstrated that sulfation dominated the metabolism and pharmacokinetics of forsythin,while the sulfate conjugate was excreted mainly in the urine. | Lu-lu Pan Yong Yang Min Hui Shuo Wang Cui-yun Li Hong Zhang Yan-hua Ding Li Fu Xing-xing Diao Da-fang Zhong | 2021 | Acta Pharmacologica Sinica2021,42,2: | 0 |
