| 2 | Neutrophil extracellular traps mediate the crosstalk between glioma progression and the tumor microenvironment via the HMGB1/RAGE/IL-8 axis显示文摘Objective:Neutrophil extracellular traps(NETs)produced by tumor-infiltrating neutrophils(TINs)are associated with poor prognosis in patients with several types of cancer.However,the mechanisms underlying the involvement of NETs in glioma progression remain largely unknown.This study aimed to elucidate the roles of NETs in biological processes that drive the crosstalk between glioma progression and the tumor microenvironment.Methods:Neutrophil infiltration and NETs formation were investigated in glioma tissue through immunohistochemistry,and their relationships with clinicopathological features and outcomes were statistically evaluated.The effects of NETs on glioma cell progression were studied in a co-culture system.In vivo and in vitro experiments validated the reactive oxygen species activity and cytokine production of TINs,as well as the ERK signaling pathway activation and the metastasis of gliomas.Results:Neutrophil infiltration and NETs formation were induced in high-grade glioma compared with low-grade glioma.NETs induced by TINs were determined to be an oncogenic marker of high-grade gliomas and to be involved in cell proliferation and invasion.NETs overproduction promoted glioma cell proliferation,migration,and invasion.Furthermore,HMGB1 was found to bind to RAGE and activate the NF-κB signaling pathway in vitro.In addition,NETs stimulated the NF-κB signaling pathway,thus promoting IL-8 secretion in glioblastoma.Subsequently,IL-8 recruited neutrophils which in turn mediated NETs formation via the PI3 K/AKT/ROS axis in TINs.Conclusions:Our results suggest that NETs produced by TINs mediate the crosstalk between glioma progression and the tumor microenvironment by regulating the HMGB1/RAGE/IL-8 axis.Targeting NETs formation or IL-8 secretion may be an effective approach to inhibit glioma progression. | Caijun Zha Xiangqi Meng Lulu Li Shan Mi Da Qian Ziwei Li Pengfei Wu Shaoshan Hu Shihong Zhao Jinquan Cai Yanhong Liu | 2020 | Cancer Biology & Medicine2020，17，1： | 19 |
| 3 | 二期尿道成型术更适合于需要横断尿道板并伴有严重弯曲畸形的近段型尿道下裂显示文摘对于伴有严重阴茎弯曲的尿道下裂,横断尿道板后是选择一期还是二期手术仍存在争议。这项回顾性研究比较评价了这两种方法的优劣。本研究共收集66例伴有严重阴茎弯曲的近端型尿道下裂的患者,根据手术方式分成两组,其中32例行一期手术(Duckett),34例行二期手术。一期和二期手术组的中位年龄分别为7.5岁和11.0岁,中位随访时间分别为28.5个月和35个月。所有患者术后尿道外口均位于龟头顶端。随访期间无患者出现阴茎弯曲复发,患者及其家属对阴茎的长度和外观均满意。术后两组各有8例患者出现并发症,两组间并发症率无统计学差异。一期手术尿道狭窄的远期并发症率高j(18.75%1,JO%)。在一期手术组中,青春期前患者的并发症率显著低于青春期后患者(10.52%w46.15%3。中期随访结果提示,横断尿道板可有效矫正近端型尿道下裂合并的重度阴茎弯曲。考虑到一期手术组中较高的尿道狭窄发生率,本文认为分期手术更适合伴有严重阴茎弯曲需要横断尿道板的近端型尿道下裂。 | Da-Chao Zheng Hai-Jun Yao Zhi-Kang Cai Jun Da Qi Chen Yan-Bo Chen Ke Zhang Ming-Xi Xu Mu-Jun Lu Zhong Wang | 2015 | Asian Journal of Andrology2015，17，1： | 13 |
| 4 | A feasible,economical,and accurate analytical method for simultaneous determination of six alkaloid markers in Aconiti Lateralis Radix Praeparata from different manufacturing sources and processing ways显示文摘Aconiti Lateralis Radix Praeparata(Fuzi) is a commonly used traditional Chinese medicine in clinic for its potency in restoring yang and rescuing from collapse. Aconiti alkaloids, mainly including monoester-diterpenoidaconitines(MDAs) and diester-diterpenoidaconitines(DDAs), are considered to act as both bioactive and toxic constituents. In the present study, a feasible, economical, and accurate HPLC method for simultaneous determination of six alkaloid markers using the Single Standard for Determination of Multi-Components(SSDMC) method was developed and fully validated. Benzoylmesaconine was used as the unique reference standard. This method was proven as accurate(recovery varying between 97.5%-101.8%, RSD < 3%), precise(RSD 0.63%-2.05%), and linear(R > 0.999 9) over the concentration ranges, and subsequently applied to quantitative evaluation of 62 batches of samples, among which 45 batches were from good manufacturing practice(GMP) facilities and 17 batches from the drug market. The contents were then analyzed by principal component analysis(PCA) and homogeneity test. The present study provided valuable information for improving the quality standard of Aconiti Lateralis Radix Praeparata. | 7 | Functional Diversity of CYCLOIDEA-like TCP Genes in the Control of Zygomorphic Flower Development in Lotus japonicus显示文摘CYCLOIDEA (CYC)-like TCP genes play key roles in dorsoventral differentiation of zygomorphic flowers in Papilionoideae legumes. In this study, we analyzed the kew mutants whose flowers lost lateral identity, and investigated the diverse functions of three LjCYC genes during zygomorphic flower development in the model legume Lotus japonicus. We showed that kew1 and kew3 are allelic mutants of LjCYC3, a CYC-like TCP gene. Through transgenic experiments, it was shown that LjCYC1 possesses dorsal activity similar to LjCYC2, and that LjCYC3 alone is sufficient to confer lateral activity, and an epistatic effect between dorsal and lateral activities was identified. Sequence analysis revealed a striking alteration at the 3 end of the LjCYC3 open reading frame (ORF) in comparison with those of LjCYC1 and LjCYC2 ORFs. Furthermore, it was found that LjCYC proteins could interact with each other and possess different activities by means of a transcriptional activity assay.
