1Free fatty acid-induced miR-22 inhibits gluconeogenesis via SIRT-1-mediated PGC-1α expression in nonalcoholic fatty liver disease显示文摘Background and aims:Free fatty acids(FFAs)are one of the important regulators of the progression of nonalcoholic fatty liver disease.The FFAs are shown to modulate the metabolic status of the liver by modulating several cellular pathways in hepatocytes.Here,we elucidated the role of miR-22 in modulating FFAs-mediated gluconeogenesis.Methods:Huh7 and WRL68 cells were transfected with nonspecific miRNA,miR-22 premiRs or anti-miR-22 oligos followed by incubation with palmitic acid,oleic acid,and linoleic acid(300μM each)for 48 and 72 h after transfection.The expression of miR-22 was performed using real-time polymerase chain reaction and Western blots were performed for SIRT-1,PGC-1α,PEPCK,and glucose-6-phosphatase.Three groups of C57BL/6 mice(6 mice per group)were fed with standard diet,choline sufficient L-amino acid defined diet or choline-deficient L-amino acid defined(CDAA)diet for 6,18,32,or 54 weeks.Triglycerides content was measured in the serum.Expression of miR-22 and the protein expression of gluconeogenic enzymes were analyzed in the tissue samples.Results:Incubation of miR-22-transfected cells with FFAs inhibited the expression of SIRT-1,PGC-1α,PEPCK,and glucose-6-phosphatase,while miR-22 expression was increased.These changes were reversed when the cells were transfected with anti-miR-22 oligos.CDAA-fed mice showed the significant increase in triglycerides content and miR-22 expression,while there was an inhibition of SIRT-1,PGC-1α,PEPCK,and glucose-6-phosphatase expression in CDAA-fed mice.Ajay K.Yadav Teja Naveen Sata Daksh Verma Amrendra K.Sah Amit K.Mishra Mrinalini Md.Musa Hossain Kishor Pant Senthil K.Venugopal 2023iLIVER2023,2,1:1

6Self-aligned on-chip coupled photonic devices using individual cadmium sulfide nanobelts显示文摘Nanowires(NWs)and nanobelts(NBs)have been widely studied and fabricated into a variety of nanoscale devices such as light emitting diodes(LEDs),lasers and biosensors.These unique materials have attracted sustained attention due to their novel properties,ease of growth,and the ability to fabricate highly engineered devices.However,their widespread application remains hindered due to the difficulty in integrating multiple NWs or NBs together for more complex devices.Integration of multiple NWs and NBs together on the same chip can enable the coupling of different devices to help realize complex on-chip architectures such as photonic integrated circuits or nanoscale diagnostic tools,which currently require outcoupling using larger components.In this letter we report the coupling of on-chip NB LEDs and photodetectors using a single,precisely self-aligned,cadmium sulfide(CdS)NB fabricated on a silicon-on-insulator(SOI)substrate.Electroluminescence generated by the CdS NB is waveguided and measured across the self-aligned device and demonstrates an on/off ratio of 10^2-10^3.This work describes a new method for fabricating and integrating more complex nanoscale devices that can enable advances in areas such as on-chip optical computational components and nanoscale optical biodiagnostics.Jacob S.Berger Ho-Seok Ee Mingliang Ren Daksh Agarwal Wenjing Liu Ritesh Agarwal 2020Nano Research2020,13,5:0
7Impact of frailty on endoscopic retrograde cholangiopancreatography outcomes in nonagenarians:A United States national experience显示文摘BACKGROUND Endoscopic retrograde cholangiopancreatography(ERCP)is an essential therapeutic tool for biliary and pancreatic diseases.Frail and elderly patients,especially those aged≥90 years are generally considered a higher-risk population for ERCP-related complications.AIM To investigate outcomes of ERCP in the Non-agenarian population(≥90 years)concerning Frailty.METHODS This is a cohort study using the 2018-2020 National Readmission Database.Patients aged≥90 were identified who underwent ERCP,using the international classification of diseases-10 code with clinical modification.Johns Hopkins’s adjusted clinical groups frailty indicator was used to classify patients as frail and non-frail.The primary outcome was mortality,and the secondary outcomes were morbidity and the 30 d readmission rate related to ERCP.We used univariate and multivariate regression models for analysis.RESULTS A total of 9448 patients were admitted for any indications of ERCP.Frail and non-frail patients were 3445(36.46%)and 6003(63.53%)respectively.Indications for ERCP were Choledocholithiasis(74.84%),Biliary pancreatitis(9.19%),Pancreatico-biliary cancer(7.6%),Biliary stricture(4.84%),and Cholangitis(1.51%).Mortality rates were higher in frail group[adjusted odds ratio(aOR)=1.68,P=0.02].The Intra-procedural complications were insigni-ficant between the two groups which included bleeding(aOR=0.72,P=0.67),accidental punctures/lacerations(aOR=0.77,P=0.5),and mechanical ventilation rates(aOR=1.19,P=0.6).Post-ERCP complication rate was similar for bleeding(aOR=0.72,P=0.41)and post-ERCP pancreatitis(aOR=1.4,P=0.44).Frail patients had a longer length of stay(6.7 d vs 5.5 d)and higher mean total charges of hospitalization($78807 vs$71392)compared to controls(P<0.001).The 30 d all-cause readmission rates between frail and non-frail patients were similar(P=0.96).CONCLUSION There was a significantly higher mortality risk and healthcare burden amongst nonagenarian frail patients undergoing ERCP compared to non-frail.Larger studies are warranted to investigate and mitigate modifiable risk factors.Sanket Dhirubhai Basida Dushyant Singh Dahiya Muhammad Nadeem Yousaf Brinda Basida Bhanu Siva Mohan Pinnam Manesh Kumar Gangwani Hassam Ali Sahib Singh Yash R Shah Daksh Ahluwalia Mihir Prakash Shah Saurabh Chandan Neil R Sharma Shyam Thakkar 2024World Journal of Gastrointestinal Endoscopy2024,16,3:0
