1Dual Functions of Microglia in Ischemic Stroke显示文摘Ischemic stroke is a leading cause of morbidity and mortality worldwide. Resident microglia are the principal immune cells of the brain, and the first to respond to the pathophysiological changes induced by ischemic stroke. Traditionally, it has been thought that microglial activation is deleterious in ischemic stroke, and therapies to suppress it have been intensively explored. However,increasing evidence suggests that microglial activation is also critical for neurogenesis, angiogenesis, and synaptic remodeling, thereby promoting functional recovery after cerebral ischemia. Here, we comprehensively review the dual role of microglia during the different phases of ischemic stroke, and the possible mechanisms controlling the post-ischemic activity of microglia. 3肝炎 E : 传染病学和预防显示文摘 Hepatitis E is caused by the hepatitis E virus (HEV), the major etiologic agent of enterically transmitted non-A hepatitis worldwide. HEV is responsible for major outbreaks of acute hepatitis in developing countries, especially in many parts of Africa and Asia. The HEV is a spherical, non-enveloped, single-stranded, positive sense RNA virus that is approximately 32 nm to 34 nm in diameter and is the only member in the family Hepeviridae and genus Hepevirus. There are four distinct genotypes of HEV (genotypes 1-4). While genotype 1 is predominantly associated with large epidemics in developing countries, genotype 3 has recently emerged as a significant pathogen in developed countries. The clinical manifestations and the laboratory abnormalities of hepatitis E are not distinguishable from that caused by other hepatitis viruses. However, high mortality among pregnant women particularly during the third trimester distinguishes HEV from other causes of acute viral hepatitis. Specific etiologic diagnosis among infected cases can be made by serological testing or detection of viral nucleic acid by reverse transcription polymerase chain reaction. Although there are vaccine candidates that had been shown to be safe and efficacious in clini-cal trials, none are approved currently for use. 13N-acetyldopamine dimer inhibits neuroinflammation through the TLR4/NF-κB and NLRP3/Caspase-1 pathways显示文摘Neuroinflammation mediated by microglia is an important pathophysiological mechanism in neurodegenerative diseases.However,there is a lack of effective drugs to treat neuroinflammation.N-acetyldopamine dimer(NADD)is a natural compound from the traditional Chinese medicine Isaria cicada.In our previous study,we found that NADD can attenuate DSS-induced ulcerative colitis by suppressing the NF-κB and MAPK pathways.Does NADD inhibit neuroinflammation,and what is the target of NADD?To answer this question,lipopolysaccharide(LPS)-stimulated BV-2 microglia was used as a cell model to investigate the effect of NADD on neuroinflammation.Nitric oxide(NO)detection,reactive oxygen species(ROS)detection and enzyme-linked immunosorbent assay(ELISA)results show that NADD attenuates inflammatory signals and proinflammatory cytokines in LPS-stimulated BV-2 microglia,including NO,ROS,tumor necrosis factor(TNF)-α,interleukin(IL)-1βand interleukin-6(IL-6).Western blot analysis show that NADD inhibits the protein levels of Toll-like receptor 4(TLR4),nuclear factor kappa-B(NF-κB),NOD-like receptor thermal protein domain associated protein 3(NLRP3),ASC and cysteinyl aspartate specific proteinase(Caspase)-1,indicating that NADD may inhibit neuroinflammation through the TLR4/NF-κB and NLRP3/Caspase-1 signaling pathways.In addition,surface plasmon resonance assays and molecular docking demonstrate that NADD binds with TLR4 directly.Our study reveals a new role of NADD in inhibiting the TLR4/NF-κB and NLRP3/Caspase-1 pathways,and shows that TLR4-MD2 is the direct target of NADD,which may provide a potential therapeutic candidate for the treatment of neuroinflammation.Lijun Huang Leiqiang Gong Xueyan Huo Lirong Lei Qi Zhang Yunjie Hu Qixuan Kuang Yu Gui Yifei Dai Yucheng Gu Yun Deng Dong Wang Dale Guo 2023Acta Biochimica et Biophysica Sinica2023,55,1:0
14Correlation between the distribution pattern of virulence genes and virulence of Aeromonas hydrophila strains显示文摘Nine strains of Aeromonas hydrophila isolated from diseased fish or soft-shelled tortoise were tested for the presence of three virulence genes including the genes encoding aerolysin,hemolysin,and extracellular serine protease(i.e.,aerA,hlyA,and ahpA,respectively).These genes were investigated using polymerase chain reaction(PCR)with specific primers for each gene.And the pathogenicities to Carrassius auratus ibebio of these strains were also assayed.PCR results demonstrated that the distribution patterns of aerA,hlyA,and ahpA were different in these strains.6/9 of A.hydrophila strains were aerA positive,8/9 of strains hlyA positive,7/9 of strains ahpA positive,respective-ly.However,the assay for pathogenesis showed that two strains(A.hydrophila XS91-4-1 and C2)were strong virulent,two strains(A.hydrophila ST78-3-3 and 58-20-9)avirulent and the rest middle virulent was to the fish.In conclusion,there are significant correlation between the distribution pattern of the three virulence genes and the patho-genicity to Carrassius auratus ibebio.All strong virulent A.hydrophila strains were aerA+hlyA+ahpA+genotype,and all aerA+hlyA+ahpA+strains were virulent.Strains with the genotype of aerA−hlyA−ahpA+have middle pathogenicity.In the present study,we found for the first time that all A.hydrophila isolated from the ahpA positive were virulent to Carrassius auratus ibebio.Additionally,there was a positive correlation between the virulence of A.hydrophila and the presence of aerA and ahpA.ZHU Daling LI Aihua WANG Jianguo LI Ming CAI Taozhen HU Jing 2007Frontiers in Biology2007,2,2:0
