| 1 | Th17 plasticity and its changes associated with inflammatory bowel disease显示文摘CD4 T helper(Th) cell differentiation into distinct T cell subsets is critical to the normal function of the immune system. Until recently,the paradigm held that na?ve T cells differentiated into distinct subsets under the guidance of environmental cues(e.g.,cytokines) and that once polarized,these cells were committed to a particular functional state. However,the existence of transdifferentiated T cell populations,which express signature transcription factors and cytokines associated with more than one Th subset,challenges the immutability of T helper subsets and suggests that plasticity is a feature of multifaceted immune responses. How this process impacts immune dysregulation in diseases such as inflammatory bowel diseases(IBD) and the machinery that underlies this process is far from fully understood. Interleukin(IL)-17 secreting helper T(Th17) cells have been heavily implicated in tissue-specific immune pathology including murine models of IBD,human Crohn's disease and ulcerative colitis. Plasticity within this subset is suggested by the existence of IL-17 secreting cells,which,can also secrete interferon-γ,the signature cytokine for Th1 cells or,can co-express the anti-inflammatory transcription factor forkhead box p3,a signature transcription factor of regulatory T cells. | 3 | Follow up of serial urea breath test results in patients after consumption of antibiotics for non-gastric infections显示文摘AIM: The widespread use of antibacterial therapy hasbeen suggested to be the cause for the decline in theprevalence of Helicobacter pyloriinfection. This studyexamine the serial changes of urea breath test resultsin a group of hospitalized patients who were givenantibacterial therapy for non-gastric infections.METHODS: Thirty-five hospitalized patients who weregiven antibacterial therapy for clinical infections,predominantly chest and urinary infections, werestudied. Most (91%) patients were given singleantibiotic of either a penicillin or cephalosporin group.Serial 13C-urea breath tests were performed within 24hours of initiation of antibiotics, at one-week and atsix-week post-therapy. H. pylori infection wasdiagnosed when one or more urea breath tests waspositive.RESULTS: All 35 patients completed three serial ureabreath tests and 26 (74 %) were H. pylori-positive. Ten(38 %) H. pylori-infected patients had at least onenegative breath test results during the study period.The medium delta 13C values were significantly lowerat baseline (8.8) than at one-week (20.3) and six-week(24.5) post-treatment in H. pylori-positive individuals(P=0.022). Clearance of H. pyloriat six-week was onlyseen in one patient who had received anti-helicobactertherapy from another source.CONCLUSION: Our results suggested that one-third ofH. pylori-infected individuals had transient false-negative urea breath test results during treatment withantibacterial agent. | 5 | Ecological and behavioural risk factors of scrub typhus in central Vietnam:a case-control study显示文摘Background The risk factors for scrub typhus in Vietnam remain unknown.Scrub typhus caused by Orientia tsutsugamushi often presents as an undifferentiated febrile illness and remains under appreciated due to the limited availability of diagnostic tests.This tropical rickettsial illness is increasingly recognized as an important cause of non-malaria acute undifferentiated fever in Asia.This study aimed to investigate behavioural and ecological related risk factors of scrub typhus to prevent this potentially life-threatening disease in Vietnam.Methods We conducted a clinical hospital-based active surveillance study,and a retrospective residence-enrolment date-age-matched case–control study in Khanh Hoa province,Vietnam,from August 2018 to March 2020.Clinical examinations,polymerase chain reaction and enzyme-linked immunosorbent assay IgM tests were applied to define cases and controls.All enrolled participants filled out a questionnaire including demographic socio-economic status,personal behaviors/protective equipment,habitat connections,land use,and possible exposure to the vector.Multivariable conditional logistic regression was used to define the scrub typhus associated risk factors.Results We identified 44 confirmed cases and matched them with 152 controls.Among cases and controls,the largest age group was the 41–50 years old and males accounted for 61.4%and 42.8%,respectively.There were similarities in demographic characteristics between the two groups,with the exception of occupation.Several factors were significantly associated with acquisition of scrub typhus,including sitting/laying directly on household floor[adjusted OR(aOR)=4.9,95%CI:1.6–15.1,P=0.006],household with poor sanitation/conditions(aOR=7.9,95%CI:1.9–32.9,P=0.005),workplace environment with risk(aOR=3.0,95%CI:1.2–7.6,P=0.020),always observing mice around home(aOR=3.7,95%CI:1.4–9.9,P=0.008),and use of personal protective equipment in the field(aOR=0.4,95%CI:0.1–1.1,P=0.076).Conclusions Ecological and household hygiene-related factors were more associated with scrub typhus infection,than individual-level exposure activities in the hyper-endemic area.These findings support local education and allow people to protect themselves from scrub typhus,especially in areas with limitations in diagnostic capacity. | Hanh Thi Duc Tran Jan Hattendorf Hung Manh Do Thanh Tien Hoang Hang Thi Hai Hoang Hoa Ngoc Lam Mai Kim Huynh Lan Thi Hoang Vu Jakob Zinsstag Daniel Henry Paris Esther Schelling | 2021 | Infectious Diseases of Poverty2021，10，4： | 1 |

| 18 | Silk fibroin as an additive for cell-free protein synthesis显示文摘Cell-free systems contain many proteins and metabolites required for complex functions such as transcription and translation or multi-step metabolic conversions.Research into expanding the delivery of these systems by drying or by embedding into other materials is enabling new applications in sensing,point-of-need manufacturing,and responsive materials.Meanwhile,silk fibroin from the silk worm,Bombyx mori,has received attention as a protective additive for dried enzyme formulations and as a material to build biocompatible hydrogels for controlled localization or delivery of biomolecular cargoes.In this work,we explore the effects of silk fibroin as an additive in cell-free protein synthesis(CFPS)reactions.Impacts of silk fibroin on CFPS activity and stability after drying,as well as the potential for incorporation of CFPS into hydrogels of crosslinked silk fibroin are assessed.We find that simple addition of silk fibroin increased productivity of the CFPS reactions by up to 42%,which we attribute to macromolecular crowding effects.However,we did not find evidence that silk fibroin provides a protective effects after drying as previously described for purified enzymes.Further,the enzymatic crosslinking transformations of silk fibroin typically used to form hydrogels are inhibited in the presence of the CFPS reaction mixture.Crosslinking attempts did not impact CFPS activity,but did yield localized protein aggregates rather than a hydrogel.We discuss the mechanisms at play in these results and how the silk fibroin-CFPS system might be improved for the design of cell-free devices. | Marilyn S.Lee Chia-Suei Hung Daniel A.Phillips Chelsea C.Buck Maneesh K.Gupta Matthew W.Lux | 2020 | Synthetic and Systems Biotechnology2020，5，3： | 0 |
