| 1 | An Effective Platform for Exploring Rotavirus Receptors by Bacterial Surface Display System显示文摘Rotavirus(RV) is a major foodborne pathogen. For RV prevention and control, it is a key to uncover the interaction mechanism between virus and its receptors. However, it is hard to specially purify the viral receptors, including histo-blood group antigens(HBGAs). Previously, the protruding domain protein(P protein) of human norovirus(genotype Ⅱ.4) was displayed on the surface of Escherichia coli, and it specifically recognized and captured the viral ligands. In order to further verify the feasibility of the system, P protein was replaced by VP8* of RV(G9 P[8]) in this study. In the system, VP8*could be correctly released by thrombin treatment with antigenicity retaining, which was confirmed by Western blot and Enzyme-Linked Immunosorbent Assays. Type A HBGAs from porcine gastric mucin(PGM) were recognized and captured by this system. From saliva mixture, the captured viral receptor bound with displayed VP8* was confirmed positive with monoclonal antibody against type A HBGAs. It indicated that the target ligands could be easily separated from the complex matrix. | 2 | Chromatin-associated SUMOylation controls the transcriptional switch between plant development and heat stress responses显示文摘The post-translational protein modification known as SUMOylation has conserved roles in the heat stress responses of various species.The functional connection between the global regulation of gene expression and chromatin-associatedSUMOylation in plant cells isunknown.Here,weuncovereda genome-wide relationship between chromatin-associated SUMOylation and transcriptional switches in Arabidopsis thaliana grown at room temperature,exposed to heat stress,and exposed to heat stress followed by recovery.The small ubiquitin-like modifier(SUMO)-associated chromatin sites,characterized by whole-genome ChIP-seq,were generally associated with active chromatin markers.In response to heat stress,chromatin-associated SUMO signals increased at promoter-transcriptional start site regions and decreased in gene bodies.RNAseq analysis supported the role of chromatin-associatedSUMOylation in transcriptional activation during rapid responses to high temperature.Changes inSUMOsignals on chromatinwere associated with the upregulation of heat-responsivegenesandthedownregulation ofgrowth-relatedgenes.Disruption of theSUMOligasegene SIZ1 abolished SUMOsignals on chromatin and attenuated rapid transcriptional responses to heat stress.The SUMO signal peaks were enriched in DNA elements recognized by distinct groups of transcription factors under different temperature conditions.These observations provide evidence that chromatin-associated SUMOylation regulates the transcriptional switch between development and heat stress response in plant cells. | Danlu Han Chen Chen Simin Xia Jun Liu Jie Shu Vi Nguyen Jianbin Lai Yuhai Cui Chengwei Yang | 2021 | Plant Communications2021,2,1: | 1 |
| 3 | Design of a novel chimeric peptide via dual blockade of CD47/SIRPαand PD-1/PD-L1 for cancer immunotherapy显示文摘Although immune checkpoint inhibition has been shown to effectively activate antitumor immunity in various tumor types,only a small subset of patients can benefit from PD-1/PD-L1 blockade.CD47 expressed on tumor cells protects them from phagocytosis through interaction with SIRPαon macrophages,while PD-L1 dampens T cell-mediated tumor killing.Therefore,dual targeting PD-L1 and CD47 may improve the efficacy of cancer immunotherapy.A chimeric peptide Pal-DMPOP was designed by conjugating the double mutation of CD47/SIRPαblocking peptide(DMP)with the truncation of PD-1/PD-L1 blocking peptide OPBP-1(8-12)and was modified by a palmitic acid tail.Pal-DMPOP can significantly enhance macrophage-mediated phagocytosis of tumor cells and activate primary T cells to secret IFN-γin vitro.Due to its superior hydrolysis-resistant activity as well as tumor tissue and lymph node targeting properties,Pal-DMPOP elicited stronger anti-tumor potency than Pal-DMP or OPBP-1(8-12)in immune-competent MC38 tumor-bearing mice.The in vivo anti-tumor activity was further validated in the colorectal CT26 tumor model.Furthermore,Pal-DMPOP mobilized macrophage and T-cell anti-tumor responses with minimal toxicity.Overall,the first bispecific CD47/SIRPαand PD-1/PD-L1 dual-blockade chimeric peptide was designed and exhibited synergistic anti-tumor efficacy via CD8^(+)T cell activation and macrophage-mediated immune response.The strategy could pave the way for designing effective therapeutic agents for cancer immunotherapy. | Zheng Hu Wanqiong Li Shaomeng Chen Danhong Chen Ran Xu Danlu Zheng Xin Yang Shuzhen Li Xiuman Zhou Xiaoshuang Niu Youmei Xiao Zhuoying He Huihao Li Juan Liu Xinghua Sui Yanfeng Gao | 2023 | Science China(Life Sciences)2023,66,10: | 1 |
| 4 | Study of the influence of virtual guard ring width on the performance of SPAD detectors in 180 nm standard CMOS technology显示文摘The influence of the virtual guard ring width(GRW)on the performance of the p-well/deep n-well single-photon avalanche diode(SPAD)in a 180 nm standard CMOS process was investigated.TCAD simulation demonstrates that the electric field strength and current density in the guard ring are obviously enhanced when GRW is decreased to 1μm.It is experimentally found that,compared with an SPAD with GRW=2μm,the dark count rate(DCR)and afterpulsing probability(AP)of the SPAD with GRW=1μm is significantly increased by 2.7 times and twofold,respectively,meanwhile,its photon detection probability(PDP)is saturated and hard to be promoted at over 2 V excess bias voltage.Although the fill factor(FF)can be enlarged by reducing GRW,the dark noise of devices is negatively affected due to the enhanced trap-assisted tunneling(TAT)effect in the 1μm guard ring region.By comparison,the SPAD with GRW=2μm can achieve a better trade-off between the FF and noise performance.Our study provides a design guideline for guard rings to realize a low-noise SPAD for large-array applications. | Danlu Liu Ming Li Tang Xu Jie Dong Yuming Fang Yue Xu | 2023 | Journal of Semiconductors2023,44,11: | 0 |
