| 1 | In vitro genotoxicity evaluation and metabolic study of residual glutaraldehyde in animal-derived biomaterials显示文摘Glutaraldehyde(GA)is an important additive that is mainly used in animal-derived biomaterials to improve their mechanical and antimicrobial capacities.However,GA chemical toxicity and the metabolic mechanism remain relatively unknown.Therefore,residual GA has always been a major health risk consideration for animal-derived medical devices.In this study,extracts of three biopatches were tested via the GA determination test and mouse lymphoma assay(MLA).The results showed that dissolved GA was a potential mutagen,which could induce significant cytotoxic and mutagenic effects in mouse lymphoma cells.These toxic reactions were relieved by the S9 metabolic activation(MA)system.Furthermore,we confirmed that GA concentration decreased and glutaric acid was generated during the catalytic process.We revealed GA could be oxidized via cytochrome P450 which was the main metabolic factor of S9.We found that even though GA was possibly responsible for positive reactions of animal-derived biomaterials’biocompatibility evaluation,it may not represent the real situation occurring in human bodies,owing to the presence of various detoxification mechanisms including the S9 system.Overall,in order to achieve a general balance between risk management and practical application,rational decisions based on comprehensive analyses must be considered. | Jianfeng Shi Huan Lian Yuanli Huang Danmei Zhao Han Wang Chunren Wang Jingli Li Linnan Ke | 2020 | Regenerative Biomaterials2020,7,6: | 1 |

| 3 | CMLP: Exploiting Caches at Multiple Levels of Proxies to Enhance Seamless Mobility Support in Information-Centric Networks显示文摘The recent evolution of the Internet towards 'Information-centric' transfer modes has renewed the interest in exploiting proxies to enhance seamless mobility. In this work, we focus on the case of multiple levels of proxies in ICN architectures, in which content requests from mobile subscribers and the corresponding items are proactively cached to these proxies at different levels. Specifically, we present a multiple-level proactive caching model that selects the appropriate subset of proxies at different levels and supports distributed online decision procedures in terms of the tradeoff between delay and cache cost. We show via extensive simulations the reduction of up to 31.63% in the total cost relative to Full Caching, in which caching in all 1-level neighbor proxies is performed, and up to 84.21% relative to No Caching, in which no caching is used. | 4 | High Microplastic Contamination in Juvenile Tri-Spine Horseshoe Crabs:A Baseline Study of Nursery Habitats in Northern Beibu Gulf,China显示文摘Microplastics(MPs)are ubiquitous in marine sedimentary environments.Their occurrence in horseshoe crabs and interactions with the sedimentary environment has not been determined.In this study,MPs,mostly microfibers,were found in all extracted gastrointestinal tract(GIT)samples of the juvenile Tachypleus tridentatus from the northern Beibu Gulf,China.The MP concentrations(4–53 items ind-1)were higher than those in most marine benthic invertebrates(<15 items ind-1)reported in Chinese waters,despite their generally low level in habitat sediments(9–1818 items kg-1).The correlation between the juveniles and sediments was not evident,but the abundance in juvenile horseshoe crab GITs decreased with ages.The findings were relevant to the life-history characteristics of the species,typically with limited dispersal capability and their spending nine years or longer time living in mangrove wetlands during low tides,with apparent ontogenetic changes in their diets.These baseline data enable a better understanding of MP availability in benthic macroinvertebrates,and the ecological risks present in the ecosystems. | WANG Xueping LO Hoi Shing FU Yijian WU Zhou QIN Danmei HUANG Xing ZHU Jingmin CHEUNG Siu Gin KWAN Kit Yue | 2022 | Journal of Ocean University of China2022,21,3: | 0 |
| 5 | Interaction of Hepatitis B Virus X Protein with the Pregnane X Receptor Enhances the Synergistic Effects of Aflatoxin B1 and Hepatitis B Virus on Promoting Hepatocarcinogenesis显示文摘Background and Aims:Hepatitis B virus(HBV)infection has been found to increase hepatocellular sensitivity to carcinogenic xenobiotics,by unknown mechanisms,in the generation of hepatocellular carcinoma.The pregnane X receptor(PXR)is a key regulator of the body’s defense against xenobiotics,including xenobiotic carcinogens and clinical drugs.In this study,we aimed to investigate the molecular mechanisms of HBV X protein(HBx)-PXR signaling in the synergistic effects of chemical carcinogens in HBV-associated hepatocarcinogenesis.Methods:The expression profile of PXR-cytochrome p4503A4(CYP3A4)signaling was determined by PCR,western blotting,and tissue microarray.Cell viability and aflatoxin B1(AFB1)cytotoxicity were measured using the cell counting kit-8 assay.Target gene expression was evaluated using transient transfection and real time-PCR.The genotoxicity of AFB1 was assessed in newborn mice with a single dose of AFB1.Results:HBx enhanced the hepatotoxicity of AFB1 by activating CYP3A4 and reducing glutathione Stransferase Mu 1(GSTM1)in cell lines.Activation of PXR by pregnenolone 16α-carbonitrile increased AFB1-induced liver tumor incidence by up-regulating oncogenic KRAS to enhance interleukin(IL)-11:IL-11 receptor subunit alpha-1(IL11RA-1)-mediated inflammation in an HBx transgenic model.Conclusions:Our finding regarding AFB1 toxicity enhancement by an HBx-PXR-CYP3A4/GSTM1-KRASIL11:IL11RA signaling axis provides a rational explanation for the synergistic effects of chemical carcinogens in HBV infection-associated hepatocarcinogenesis. | Yongdong Niu Shaohua Fan Qin Luo Liming Chen Danmei Huang Wenjun Chang Wenxin Qin Ganggang Shi | 2021 | Journal of Clinical and Translational Hepatology2021,9,4: | 0 |
