| 1 | Role of microtubule dynamics in Wallerian degeneration and nerve regeneration after peripheral nerve injury显示文摘Wallerian degeneration,the progressive disintegration of distal axons and myelin that occurs after peripheral nerve injury,is essential for creating a permissive microenvironment for nerve regeneration,and involves cytoskeletal reconstruction.However,it is unclear whether microtubule dynamics play a role in this process.To address this,we treated cultured sciatic nerve explants,an in vitro model of Wallerian degeneration,with the microtubule-targeting agents paclitaxel and nocodazole.We found that paclitaxel-induced microtubule stabilization promoted axon and myelin degeneration and Schwann cell dedifferentiation,whereas nocodazole-induced microtubule destabilization inhibited these processes.Evaluation of an in vivo model of peripheral nerve injury showed that treatment with paclitaxel or nocodazole accelerated or attenuated axonal regeneration,as well as functional recovery of nerve conduction and target muscle and motor behavior,respectively.These results suggest that microtubule dynamics participate in peripheral nerve regeneration after injury by affecting Wallerian degeneration.This study was approved by the Animal Care and Use Committee of Southern Medical University,China(approval No.SMUL2015081) on October 15,2015. | Jingmin Liu Lixia Li Ying Zou Lanya Fu Xinrui Ma Haowen Zhang Yizhou Xu Jiawei Xu Jiaqi Zhang Mi Li Xiaofang Hu Zhenlin Li Xianghai Wang Hao Sun Hui Zheng Lixin Zhu Jiasong Guo | 2022 | Neural Regeneration Research2022,17,3: | 1 |

| 3 | High Microplastic Contamination in Juvenile Tri-Spine Horseshoe Crabs:A Baseline Study of Nursery Habitats in Northern Beibu Gulf,China显示文摘Microplastics(MPs)are ubiquitous in marine sedimentary environments.Their occurrence in horseshoe crabs and interactions with the sedimentary environment has not been determined.In this study,MPs,mostly microfibers,were found in all extracted gastrointestinal tract(GIT)samples of the juvenile Tachypleus tridentatus from the northern Beibu Gulf,China.The MP concentrations(4–53 items ind-1)were higher than those in most marine benthic invertebrates(<15 items ind-1)reported in Chinese waters,despite their generally low level in habitat sediments(9–1818 items kg-1).The correlation between the juveniles and sediments was not evident,but the abundance in juvenile horseshoe crab GITs decreased with ages.The findings were relevant to the life-history characteristics of the species,typically with limited dispersal capability and their spending nine years or longer time living in mangrove wetlands during low tides,with apparent ontogenetic changes in their diets.These baseline data enable a better understanding of MP availability in benthic macroinvertebrates,and the ecological risks present in the ecosystems. | WANG Xueping LO Hoi Shing FU Yijian WU Zhou QIN Danmei HUANG Xing ZHU Jingmin CHEUNG Siu Gin KWAN Kit Yue | 2022 | Journal of Ocean University of China2022,21,3: | 0 |

| 5 | Unexpected phenotypes of malformations induced in Xenopus tropicalis embryos by combined exposure to triphenyltin and 9-cis-retinoic acid显示文摘Xenopus tropicalis embryos were exposed for 48 hr to the mixtures of 5 g Sn/L triphenyltin(TPT), which is a well-known endocrine disruptor, and 0.25–5 g/L 9-cis retinoic acid(9c-RA), which is the natural ligand of retinoid X receptor. The phenotypes induced by combined exposure were more variable than those resulting from single exposure to either TPT or 9c-RA. The prominent phenotypes included underdeveloped head structures, abnormal eyes, narrow fins, enlarged proctodaeum, etc. Especially, combined exposure induced unexpected notochord malformations, which ranged from small swellings of the surface of the tails to the extension and extrusion of notochord out of the posterior tails. Compared with the 5 g Sn/L TPT-treated group, the index of fin deficiency was not affected, and the index of axis deficiency was significantly increased with increasing RA concentrations in the mixtures. | 6 | Vitamin D receptor (VDR) mediates the quiescence of activated hepatic stellate cells (aHSCs) by regulating M2 macrophage exosomal smooth muscle cell-associated protein 5 (SMAP-5)显示文摘An effective therapeutic regimen for hepatic fibrosis requires a deep understanding of the pathogenesis mechanism.Hepatic fibrosis is characterized by activated hepatic stellate cells(aHSCs)with an excessive production of extracellular matrix.Although promoted activation of HSCs by M2 macrophages has been demonstrated,the molecular mechanism involved remains ambiguous.Herein,we propose that the vitamin D receptor(VDR)involved in macrophage polarization may regulate the communication between macrophages and HSCs by changing the functions of exosomes.We confirm that activating the VDR can inhibit the effect of M2 macrophages on HSC activation.The exosomes derived from M2 macrophages can promote HSC activation,while stimulating VDR alters the protein profiles and reverses their roles in M2 macrophage exosomes.Smooth muscle cell-associated protein 5(SMAP-5)was found to be the key effector protein in promoting HSC activation by regulating autophagy flux.Building on these results,we show that a combined treatment of a VDR agonist and a macrophage-targeted exosomal secretion inhibitor achieves an excellent anti-hepatic fibrosis effect.In this study,we aim to elucidate the association between VDR and macrophages in HSC activation.The results contribute to our understanding of the pathogenesis mechanism of hepatic fibrosis,and provide potential therapeutic targets for its treatment. | Xuwentai LIU Yue WU Yanyi LI Kaiming LI Siyuan HOU Ming DING Jingmin TAN Zijing ZHU Yingqi TANG Yuming LIU Qianhui SUN Cong WANG Can ZHANG | 2023 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2023,24,3: | 0 |
