2Targeting UDP-α-D-glucose 6-dehydrogenase alters the CNS tumor immune microenvironment and inhibits glioblastoma growth显示文摘Glioblastoma(GBM,WHO grade IV glioma)is the most common and lethal malignant brain tumor in aduts with a dismal prognosis.The extracellular matrix(ECM)supports GBM progression by promoting tumor cell proliferation,migration,and immune escape.Uridine diphosphate(UDP)-glucose 6-dehydrogenase(UGDH)is the rate-limiting enzyme that catalyzes the biosynthesis of glycosaminoglycans that are the principal component of the CNS ECM.We investigated how targeting UGDH in GBM infuence$the GBM immune microenvironment,including tumor-associated microglia/macrophages(TAMs)and T cells.TAMs are the main im-mune effector cells in GBM and can directly target tumor cells if properly activated.In co-cultures of GBM cells and human primary macrophages,UGDH knockdown in GBM cells pro-moted macrophage phagocytosis and M-like polarization.In orthotropic human GBM xeno-grafts and syngeneic mouse glioma models,targeting UGDH decreased ECM deposition,increased TAM phagocytosis marker expression,reduced M2-like TAMS and inhibited tumor growth.UGDH knockdown in GBM cells also promoted cytotoxic T cell ifltration and activa-tion in orthotopic syngeneic mouse glioma models.The potent and in-human-use small mole-cule GAG synthesis inhibitor 4-methylumbelliferone(4-MU)was found to inhibit GBM cell proliferation and migration in vitro,mimic the macrophage and T-cell responses to UGDH knockdown in vitro and in vivo and inhibit growth of orthotopic murine GBM.Our study shows that UGDH supports GBM growth through multiple mechanisms and supports the development of ECM-based therapeutic strategies to simultaneously target tumor cells and their microenvi-ronment.Daqian Zhan Fatih Yalcin Ding Ma Yi Fu Shuang Wei Bachchu Lal Yunqing Li Omar Dzaye John Laterra Mingyao Ying Hernando Lopez-Bertoni Shuli Xia 2022Genes & Diseases2022,9,3:1

4Catalytic hydrotreatment of humins into cyclic hydrocarbons over solid acid supported metal catalysts in cyclohexane显示文摘Humins are common undesirable sideproducts during many acid-catalyzed reactions in renewable biomass platform conversion. However, few studies have been reported to the efficient utilization of humins.For the first time, the selective catalytic conversion of biomass-derived humins into cyclic hydrocarbons with high conversion rate and selectivity is presented using a home-made Ru/W-P-Si-O bifunctional catalyst. The multistage polymerization structure of humins was studied through controlled experiments.Results show that the CAC bond network can be efficiently depolymerized at a mild reaction temperature of 340–380 °C, catalyzed by the cooperative catalysis of nano-Ru particles and porous strong Lewis solid acid. Particularly, 95.4% conversion of humins was achieved under the optimal condition with up to 88.3%yield of cyclic hydrocarbons. The detailed composition after liquefaction was also analyzed. 5Fabrication and Properties of Aluminabased Reticulated Porous Ceramics显示文摘The primary impregnation slurry was prepared using active alumina(56.25 mass%),kaolin(15 mass%),zirconia(3.75 mass%),deionized water(25 mass%),and extra adding FS(0.2 mass%)and CMC(0.4 mass%).The effects of the active alumina particle size(d50=5.043,2.934,and 1.629μm)on the rheology and the thixotropy of the slurry were researched.It was found that the bimodal activeα-Al2O3(AMA-10)with d50=1.629μm was optimum.The secondary impregnation slurry was prepared using AMA-10,kaolin and zirconia as the main raw materials.Then the alumina-based reticulated porous ceramics were fabricated by the organic foam impregnation method combined with a secondary vacuum impregnation process.The influence of the AMA-10 content on the properties of the ceramics was studied.The residual stress of the specimens was analyzed by finite element analysis.The results show that the smaller alumina particle size and multimodal distribution are beneficial to the thixotropy of the primary impregnation slurry.The secondary vacuum impregnation technique can significantly improve the mechanical properties,the thermal shock resistance and the residual strength of the alumina-based reticulated porous ceramics.With the decrease of alumina content in the secondary impregnation slurry,the residual stress of the external layer of ceramic reinforcement gradually changes from tensile stress to compressive stress,which effectively inhibits the expansion of the surface crack,and remarkably improves the crushing strength retention ratio of alumina reticulated porous ceramics.LAO Dong JIA Wenbao WANG Yufan CHEN Ruoyu LI Shujing HEI Daqian WANG Zhonghua DING Yue ZHANG Wenhao LIU Meiqi 2020China's Refractories2020,29,2:0
