1Genetic analysis and gene mapping of a narrow leaf mutant in rice (Oryza sativa L.)显示文摘A narrow leaf mutant was obtained after T-DNA transformation conducted on a rice variety Zhonghua 11. Several abnormal morphological characteristics, including semi-dwarf, delayed flowering time, narrow and inward rolling leaves, and lower seed-setting, were observed. The rate of net photosynthesis (un-der saturate light) of flag leaves in the mutant was significantly lower than that of the wild type. More-over, the leaf transpiration rate and stomatal conductance in the mutant flag leaf were lower than those of the wild type at the grain filling stage. It was found that the mutant phenotype was not caused by the T-DNA insertion. Genetic analysis showed that the mutant was controlled by a single recessive gene, designated as nal3(t). A genetic linkage map was constructed using a large F2 mapping population de-rived from a cross between nal3(t) and an indica variety Longtefu B with 6 polymorphic markers on chromosome 12 identified from 366 SSR markers by the BAS method. Gene nal3(t) was mapped be-tween the markers RM7018 and RM3331. 2OspTAC2 encodes a pentatricopeptide repeat protein and regulates rice chloroplast development显示文摘Functional chloroplast generation depends on the precise coordination of gene expression between the plastid and the nucleus and is essential for plant growth and development. In this study, a rice(Oryza sativa) mutant that exhibited albino and seedling-lethal phenotypes was isolated from a60Co-irradiated rice population. The mutant gene was identified as an ortholog of the Arabidopsis plastid transcriptionally active chromosome protein 2(p TAC2) gene, and the mutant strain was designated osptac2. Sequence and transcription analyses showed that Osp TAC2 encodes a putative chloroplast protein consisting of 10 pentratricopeptide repeat(PPR) domains and a C-terminal small Mut S-related(SMR) domain. Cytological observations via microscopy showed that the Osp TAC2-green fluorescent fusion protein is localized in the chloroplasts. Transmission electron microscopy revealed that the chloroplast of the osptac2 mutant lacks an organized thylakoid membrane. The transcript levels of all investigated PEP(plastid-encoded RNA polymerase)-dependent genes were dramatically reduced in the osptac2 mutant, whereas the transcript levels of NEP(nuclear-encoded polymerase)-dependent genes were increased. 3Characterization and molecular cloning of a serine hydroxymethyltransferase 1(OsSHM_1) in rice显示文摘Serine hydroxymethyltransferase(SHMT) is important for one carbon metabolism and photorespiration in higher plants for its participation in plant growth and development,and resistance to biotic and abiotic stresses. A rice serine hydroxymethyltransferase gene, Os SHM1, an ortholog of Arabidopsis SHM1, was isolated using map-based cloning. The osshm1 mutant had chlorotic lesions and a considerably smaller,lethal phenotype under natural ambient CO2 concentrations,but could be restored to wild type with normal growth under elevated CO2levels(0.5% CO2), showing a typical photorespiratory phenotype. The data from antioxidant enzymes activity measurement suggested that osshm1 was subjected to signi fi cant oxidative stress. Also, Os SHM1 was expressed in allorgans tested(root, culm, leaf, and young panicle) but predominantly in leaves. Os SHM1 protein is localized to the mitochondria. 6Magnetic Nanomotor-Based Maneuverable SERS Probe显示文摘Surface-enhanced Raman spectroscopy(SERS)is a powerful sensing technique capable of capturing ultrasensitive fingerprint signal of analytes with extremely low concentration.However,conventional SERS probes are passive nanoparticles which are usually massively applied for biochemical sensing,lacking controllability and adaptability for precise and targeted sensing at a small scale.Herein,we report a“rod-like”magnetic nanomotor-based SERS probe(MNM-SP)that integrates a mobile and controllable platform of micro-/nanomotors with a SERS sensing technique.The“rod-like”structure is prepared by coating a thin layer of silica onto the self-assembled magnetic nanoparticles.Afterwards,SERS hotspots of silver nanoparticles(AgNPs)are decorated as detecting nanoprobes.The MNM-SPs can be navigated on-demand to avoid obstacles and target sensing sites by the guidance of an external gradient magnetic field.Through applying a rotating magnetic field,the MNM-SPs can actively rotate to efficiently stir and mix surrounding fluid and thus contact with analytes quickly for SERS sensing.Innovatively,we demonstrate the self-cleaning capability of the MNM-SPs which can be used to overcome the contamination problem of traditional single-use SERS probes.Furthermore,the MNM-SPs could precisely approach the targeted single cell and then enter into the cell by endocytosis.It is worth mentioning that by the effective mixing of intracellular biocomponents,much more informative Raman signals with improved signal-to-noise ratio can be captured after active rotation.Therefore,the demonstrated magnetically activated MNM-SPs that are endowed with SERS sensing capability pave way to the future development of smart sensing probes with maneuverability for biochemical analysis at the micro-/nanoscale.Yong Wang Yuhuan Liu Yang Li Dandan Xu Xi Pan Yuduo Chen Dekai Zhou Bo Wang Huanhuan Feng Xing Ma 2020Research2020,,1:1
75′-tiRNA-Gln inhibits hepatocellular carcinoma progression by repressing translation through the interaction with eukaryotic initiation factor 4A-I显示文摘tRNA-derived small RNAs(tsRNAs)are novel non-coding RNAs that are involved in the occurrence and progression of diverse diseases.However,their exact presence and function in hepatocellular carcinoma(HCC)remain unclear.Here,differentially expressed tsRNAs in HCC were profiled.A novel tsRNA,tRNAGln-TTG derived 5′-tiRNA-Gln,is significantly downregulated,and its expression level is correlated with progression in patients.In HCC cells,5′-tiRNA-Gln overexpression impaired the proliferation,migration,and invasion in vitro and in vivo,while 5′-tiRNA-Gln knockdown yielded opposite results.5′-tiRNA-Gln exerted its function by binding eukaryotic initiation factor 4A-I(EIF4A1),which unwinds complex RNA secondary structures during translation initiation,causing the partial inhibition of translation.The suppressed downregulated proteins include ARAF,MEK1/2 and STAT3,causing the impaired signaling pathway related to HCC progression.Furthermore,based on the construction of a mutant 5′-tiRNA-Gln,the sequence of forming intramolecular G-quadruplex structure is crucial for 5′-tiRNA-Gln to strongly bind EIF4A1 and repress translation.Clinically,5′-tiRNA-Gln expression level is negatively correlated with ARAF,MEK1/2,and STAT3 in HCC tissues.Collectively,these findings reveal that 5′-tiRNA-Gln interacts with EIF4A1 to reduce related mRNA binding through the intramolecular Gquadruplex structure,and this process partially inhibits translation and HCC progression.Chengdong Wu Dekai Liu Lufei Zhang Jingjie Wang Yuan Ding Zhongquan Sun Weilin Wang 2023Frontiers of Medicine2023,17,3:0
8Potential application of functional micro-nano structures in petroleum显示文摘This paper takes micro-nano motors and metamaterials as examples to introduce the basic concept and development of functional micro nano structures, and analyzes the application potential of the micro-nano structure design and manufacturing technology in the petroleum industry. The functional micro-nano structure is the structure and device with special functions prepared to achieve a specific goal. New functional micro-nano structures are classified into mobile type(e.g. micro-nano motors) and fixed type(e.g. metamaterials), and 3 D printing technology is a developed method of manufacturing. 9Robust Watermarking Algorithm for Medical Volume Data Based on Polar Cosine Transform and 3D-DCT显示文摘The amount of 3D data stored and transmitted in the Internet of Medical Things(IoMT)is increasing,making protecting these medical data increasingly prominent.However,there are relatively few researches on 3D data watermarking.Moreover,due to the particularity of medical data,strict data quality should be considered while protecting data security.To solve the problem,in the field of medical volume data,we proposed a robust watermarking algorithm based on Polar Cosine Transform and 3D-Discrete Cosine Transform(PCT and 3D-DCT).Each slice of the volume data was transformed by PCT to obtain feature row vector,and then the reshaped three-dimensional feature matrix was transformed by 3D-DCT.Based on the contour information of the volume data and the detail information of the inner slice,the visual feature vector was obtained by applying the per-ceptual hash.In addition,the watermark was encrypted by a multi-sensitive initial value Sine and Piecewise linear chaotic Mapping(SPM)system,and embedded as a zero watermark.The key was stored in a third party.Under the same experimental conditions,when the volume data is rotated by 80 degrees,cut 25%along the Z axis,and the JPEG compression quality is 1%,the Normalized Correlation Coefficient(NC)of the extracted watermark is 0.80,0.89,and 1.00 respectively,which are significantly higher than the comparison algorithm.Pengju Zhang Jingbing Li Uzair Aslam Bhatti Jing Liu Yen-wei Chen Dekai Li Lei Cao 2023Computers, Materials & Continua2023,,6:0

11LHD3 Encoding a J-Domain Protein Controls Heading Date in Rice显示文摘Heading date is one of the most important agronomic traits of rice,which critically affects rice ecogeographical adaptation,yield and quality.In this study,a late heading date 3(lhd3)mutant was screened from the^(60)Co-γirradiation mutant library.The lhd3 delayed heading date in rice under both short day and long day conditions.Map-based cloning combined with Mutmap strategy was adopted to isolate the causal LHD3 gene.The LHD3 gene encodes a DNA_J domain protein,which was ubiquitously expressed in various plant organs,and dominant expressed in stems and leaves.Subcellular localization analysis showed that LHD3 was localized to nucleus,indicating that LHD3 may interact with other elements to regulate the expression of flowering genes.The transcriptions of the heading activators Ehd1,Hd3a and RFT1 significantly decreased in the lhd3 mutant,suggesting that LHD3 may control the heading date through the Ehd1-Hd3a/RFT1 photoperiodic flowering pathway.The variation and haplotype analyses of the genomic region of LHD3 showed that there were 7 haplotypes in the LHD3 region from 4702 accessions.The haplotypes of LHD3 can be divided into two classes:class a and class b,and the heading dates of these two classes were significantly different.Further study showed that two single nucleotide polymorphisms(SNPs),SNP10(G2100C)in Hap II and SNP3(C861T)in Hap VII,may be the functional sites causing early and late heading in accessions.Nucleotide diversity analysis showed LHD3 had been selected in the indica population,rather than in the japonica population.Therefore,the present study sheds light on the regulation of LHD3 on heading date in rice and suggests that LHD3 is a novel promising new target for rice molecular design and breeding improvement.LIU Qiao QIU Linlin HUA Yangguang LI Jing PANG Bo ZHAI Yufeng WANG Dekai 2023Rice science2023,30,5:0
