| 3 | Asian Hydroclimate Changes and Mechanisms in the Preboreal from an Annually-laminated Stalagmite, Daoguan Cave, Southern China显示文摘One-year-resolved and annually-counted stalagmite multi-proxies(δ^(18)O, δ^(13)C, and layer width) from Daoguan Cave, Guizhou Province revealed detailed variability regarding the Asian Summer Monsoon(ASM) and local humidity across Bond events(BE) in the Preboreal. During BEs 8 and 7, 1.5‰ enrichments in δ^(18)O values were generally consistent with high-to low-latitude climate changes. In detail, the decadal-scale minor δ^(18)O oscillations in BE8 were broadly less than the mean value, in contrast to the significant changes in local soil moisture derived from the δ^(13)C values and layer records. In the mid-BE7, δ^(18)O variability was generally above the average level, and higher-amplitude variations were observed in the three proxy indicators. Wavelet analysis on the total δ^(18)O time series and across the specific time windows of BEs 8 and 7 identified periodicities of about 130, 60, and 20-a, respectively. Exceptionally strong in BE7, the 60-a cycle, pervasively observed in instrumental studies, became prominent starting at 11.4 kaBP. Thus, glacial background conditions are important for suppressing the ASM intensity in BE8, while during BE7, tropical hydrological circulations were potentially actively involved. | 11 | HIV-1 viral cores enter the nucleus collectively through the nuclear endocytosis-like pathway显示文摘It is recognized that HIV-1 capsid cores are disassembled in the cytoplasm, releasing their genomes into the nucleus through nuclear pores, but there is also evidence showing the capsid(CA) exists in the nucleus. Whether HIV-1 enters the nucleus and how it enters the nucleus through the undersized nuclear pore remains mysterious. Based on multicolor labeling and real-time imaging of the viral and cellular components, our observations via light and electron microscopy suggest that HIV-1 selectively gathered at the microtubule organization center(MTOC), leading the nearby nuclear envelope(NE) to undergo deformation,invagination and restoration to form a nuclear vesicle in which the viral particles were wrapped;then, the inner membrane of the nuclear vesicle ruptured to release HIV-1 into the nucleus. | 17 | Altered hACE2 binding affinity and S1/S2 cleavage efficiency of SARS-CoV-2 spike protein mutants affect viral cell entry显示文摘SARS-CoV-2 variants are constantly emerging,hampering public health measures in controlling the number of infections.While it is well established that mutations in spike proteins observed for the different variants directly affect virus entry into host cells,there remains a need for further expansion of systematic and multifaceted comparisons.Here,we comprehensively studied the effect of spike protein mutations on spike expression and proteolytic activation,binding affinity,viral entry efficiency and host cell tropism of eight variants of concern(VOC)and variants of interest(VOI).We found that both the full-length spike and its receptor-binding domain(RBD)of Omicron bind to hACE2 with an affinity similar to that of the wild-type.In addition,Alpha,Beta,Delta and Lambda pseudoviruses gained significantly enhanced cell entry ability compared to the wild-type,while the Omicron pseudoviruses showed a slightly increased cell entry,suggesting the vastly increased rate of transmission observed for Omicron variant is not associated with its affinity to hACE2.We also found that the spikes of Omicron and Mu showed lower S1/S2 cleavage efficiency and inefficiently utilized TMPRSS2 to enter host cells than others,suggesting that they prefer the endocytosis pathway to enter host cells.Furthermore,all variants'pseudoviruses we tested gained the ability to enter the animal ACE2-expressing cells.Especially the infection potential of rats and mice showed significantly increased,strongly suggesting that rodents possibly become a reservoir for viral evolution.The insights gained from this study provide valuable guidance for a targeted approach to epidemic control,and contribute to a better understanding of SARS-CoV-2 evolution. | Ke Wang Yu Pan Dianbing Wang Ye Yuan Min Li Yuanyuan Chen Lijun Bi Xian-En Zhang | 2023 | Virologica Sinica2023,38,4: | 0 |
