1Hydrogen sulfide-linked persulfidation of ABI4 controls ABA responses through the transactivation of MAPKKK18 in Arabidopsis显示文摘Hydrogen sulfide(H2S)is a signaling molecule that regulates plant hormone and stress responses.The phytohormone abscisic acid(ABA)plays an important role in plant adaptation to unfavorable environmental conditions and induces the persulfidation of L-CYSTEINE DESULFHYDRASE1(DES1)and the production of H2S in guard cells.However,it remains largely unclear how H2S and protein persulfidation participate in the relay of ABA signals.In this study,we discovered that ABSCISIC ACID INSENSITIVE 4(ABI4)acts downstream of DES1 in the control of ABA responses in Arabidopsis.ABI4 undergoes persulfidation at Cys250 that is triggered in a time-dependent manner by ABA,and loss of DES1 function impairs this process.Cys250 and its persulfidation are essential for ABI4 function in the regulation of plant responses to ABA and the H2S donor NaHS during germination,seedling establishment,and stomatal closure,which are abolished in the ABI4Cys250Ala mutated variant.Introduction of the ABI4Cys250Ala variant into the abi4 des1 mutant did not rescue its hyposensitivity to ABA.Cys250 is critical for the binding of ABI4 to its cognate motif in the promoter of Mitogen-Activated Protein Kinase Kinase Kinase 18(MAPKKK18),which propagates the MAPK signaling cascade induced by ABA.Furthermore,the DES1-mediated persulfidation of ABI4 enhances the transactivation activity of ABI4 toward MAPKKK18,and ABI4 can bind the DES1 promoter,forming a regulatory loop.Taken together,these findings advance our understanding of a post-translational regulatory mechanism and suggest that ABI4 functions as an integrator of ABA and MAPK signals through a process in which DES1-produced H2S persulfidates ABI4 at Cys250.Mingjian Zhou Jing Zhang Jie Shen Heng Zhou Didi Zhao Cecilia Gotor Luis CRomero Ling Fu Zongmin Li Jing Yang Wenbiao Shen Xingxing Yuan Yanjie Xie 2021Molecular Plant2021,14,6:5
2Improved electrochemical performances of yttrium oxyfluoride-coated Li[Li0.2Mn0.54Ni0.13Co0.13]O2 for lithium ion batteries显示文摘The Li-rich layered oxides show a higher discharge capacity over 250 m Ah/g and have been developed into a promising positive material for lithium ion batteries. A rare earth metal oxyfluoride YOF-coated Li[Li_(0.2)Mn_(0.54)Ni_(0.13)Co_(0.13)]O_2 composites have been synthesized by a simple wet chemical method. Crystal structure, micro-morphology and element valence of the pristine and YOF-coated Li[Li_(0.2)Mn_(0.54)Ni_(0.13)Co_(0.13)]O_2 materials are characterized by XRD, SEM, TEM, and XPS. The results indicate that all materials exhibit a typical layered structure, and are made up of small and homogenous particles ranging from 100 nm to 200 nm. In addition, YOF layer with a thickness of approximately 3–8 nm is precisely coated on the surface of the Li[Li_(0.2)Mn_(0.54)Ni_(0.13)Co_(0.13)]O_2. Constant current charge/discharge tests at various current densities show that the electrochemical performance of 2 wt% YOF-coated Li[Li_(0.2)Mn_(0.54)Ni_(0.13)Co_(0.13)]O_2 has been improved significantly. 2 wt% YOF-coated Li[Li_(0.2)Mn_(0.54)Ni_(0.13)Co_(0.13)]O_2 delivers the highest discharge capacity of 250.4 m Ah/g at 20 m A/g among all the samples, and capacity retention of 87% after 100 charge/discharge cycles at 200 m A/g while that of the pristine one is only81.6%. 3Analysis of the correlation between the longitudinal trajectory of SOFA scores and prognosis in patients with sepsis at 72 hour after admission based on group trajectory modeling显示文摘Background:To identify the distinct trajectories of the Sequential Organ Failure Assessment(SOFA)scores at 72 h for patients with sepsis in the Medical Information Mart for Intensive Care(MIMIC)-IV database and determine their effects on mortality and adverse clinical outcomes.Methods:A retrospective cohort study was carried out involving patients with sepsis from the MIMIC-IV database.Group-based trajectory modeling(GBTM)was used to identify the distinct trajectory groups for the SOFA scores in patients with sepsis in the intensive care unit(ICU).The Cox proportional hazards regression model was used to investigate the relationship between the longitudinal change trajectory of the SOFA score and mortality and adverse clinical outcomes.Results:A total of 16,743 patients with sepsis were included in the cohort.The median survival age was 66 years(interquartile range:54-76 years).The 7-day and 28-day in-hospital mortality were 6.0%and 17.6%,respectively.Five different trajectories of SOFA scores according to the model fitting standard were determined:group 1(32.8%),group 2(30.0%),group 3(17.6%),group 4(14.0%)and group 5(5.7%).Univariate and multivariate Cox regression analyses showed that,for different clinical outcomes,trajectory group 1 was used as the reference,while trajectory groups 2-5 were all risk factors associated with the outcome(P<0.001).Subgroup analysis revealed an interaction between the two covariates of age and mechanical ventilation and the different trajectory groups of patients’SOFA scores(P<0.05).Conclusion:This approach may help identify various groups of patients with sepsis,who may be at different levels of risk for adverse health outcomes,and provide subgroups with clinical importance.Rui Yang Didi Han Luming Zhang Tao Huang Fengshuo Xu Shuai Zheng Haiyan Yin Jun Lyu 2022Journal of Intensive Medicine2022,2,1:1
4DC-CIK细胞的生物学活性及抗淋巴瘤细胞的作用(英文)显示文摘Objective: To investigate the proliferation capabilities, immunophenotype changes, level of secreted cytokines and activities against lymphoma cells under the condition that cytokine-induced killer (CIK) cells co-cultured with dendritic cells (DC) in vitro. Methods: DC and CIK cells were induced from peripheral blood mononuclear cells of healthy volunteers. They were co-cultured meanwhile CIK cells were cultured alone as controls. Increased number of cells were counted by tapan-blue staining, killing activities were detected by MTT assay, immunophenotype changes were analyzed by flow cytometry, the IL-12 and INF-γ levels of the cultured supernatants were detected by ELISA kits. Results: The proliferation capabilities of DC-CIK cells were significantly higher than that of CIK cells (P < 0.05). Under the same condition, the ratio of double positive cells such as CD3+ CD8+, CD3+ CD56+ in CIK cells was significantly enhanced by co-cultured with DC cells (P < 0.05). The level of IL-12 and INF-γ secreted in supernatants was increased noticeably by co-cultured DC-CIK cells on day 3 compared to CIK cells which were cultured alone (P < 0.01 and P < 0.05). Within the effector-target ratio range between 5:1 to 40:1, the activi-ties against lymphoma cells of DC-CIK cells were much higher than that of CIK cells (P < 0.05), and this effect was showed a positive correlation with the effector-target ratio. Conclusion: The proliferation capabilities, the level of secreted cytokines and the activities against lymphoma cells of DC-CIK cells were significantly higher than those of CIK cells. 5Associations of HDL-C/LDL-C with myocardial infarction,all-cause mortality,haemorrhagic stroke and ischaemic stroke:a longitudinal study based on 384093 participants from the UK Biobank显示文摘Objective To explore the correlations of high-density lipoprotein cholesterol(HDL-C)/low-density lipoprotein cholesterol(LDL-C)with myocardial infarction(MI),all-cause mortality,haemorrhagic stroke and ischaemic stroke,as well as the joint association of genetic susceptibility and HDL-C/LDL-C with the MI risk.Methods and results This study selected 384093 participants from the UK Biobank(UKB)database.First,restricted cubic splines indicated non-linear associations of HDL-C/LDL-C with MI,ischaemic stroke and all-cause mortality.Second,a Cox proportional-hazards model indicated that compared with HDL-C/LDL-C=0.4-0.6,HDL-C/LDL-C<0.4 and>0.6 were correlated with all-cause mortality(HR=0.97 for HDL-C/LDL-C<0.4,95%CI=0.939 to 0.999,p<0.05;HR=1.21 for HDL-C/LDL-C>0.6,95%CI=1.16 to 1.26,p<0.001)after full multivariable adjustment.HDL-C/LDL-C<0.4 was correlated with a higher MI risk(HR=1.36,95%CI=1.28 to 1.44,p<0.05)and ischaemic stroke(HR=1.12,95%CI=1.02 to 1.22,p<0.05)after full multivariable adjustment.HDL-C/LDL-C>0.6 was associated with higher risk haemorrhagic stroke risk after full multivariable adjustment(HR=1.25,95%CI=1.03 to 1.52,p<0.05).Third,after calculating the coronary heart disease Genetic Risk Score(CHD-GRS)of each participant,the Cox proportional-hazards model indicated that compared with low CHD-GRS and HDL-C/LDL-C=0.4-0.6,participants with a combination of high CHD-GRS and HDL-C/LDL-C<0.4 were associated with the highest MI risk(HR=2.45,95%CI=2.15 to 2.8,p<0.001).Participants with HDL-C/LDL-C<0.4 were correlated with a higher MI risk regardless of whether they had a high,intermediate or low CHD-GRS.Conclusion In UKB participants,HDL-C/LDL-C ratio of 0.4-0.6 was correlated with lower MI risk,all-cause mortality,haemorrhagic stroke and ischaemic stroke.Participants with HDL-C/LDL-C<0.4 were correlated with a higher MI risk regardless of whether they had a high,intermediate or low CHD-GRS.The clinical significance and impact of HDL-C/LDL-C need to be further verified in future studies.Shiqi Yuan Xiaxuan Huang Wen Ma Rui Yang Fengshuo Xu Didi Han Tao Huang MIn Peng Anding Xu Jun Lyu 2023Stroke & Vascular Neurology2023,8,2:1
