1ROS-responsive cyclodextrin nanoplatform for combined photodynamic therapy and chemotherapy of cancer显示文摘Cyclodextrin(CD) has special spatial structure and well biological safety,so it has been widely used for constructing CD-based na noplatforms.Through functionalization,cyclodextrin can form various stimulusresponse nanoplatforms,such as pH,temperature,redox,light and magnetic fields.In this study,we designed a highly sensitive reactive oxygen species(ROS)-responsive polymer PCP which encapsulated doxorubicin(DOX) and purpurin 18(P18) to achieve the syne rgy of photodynamic and chemotherapy.The high content of reactive oxygen species(ROS) in the tumor microenvironment(TME) triggers the cleavage of the borate bond of MPEG-CD-PHB(PCP),thereby promoting the re lease of drugs.When irradiated with nea rinfrared laser,the photosensitizer P18 released by polymer micelles can produce reactive oxygen species to promote cell apoptosis.Compared with monotherapy,a series of experiments confirmed that our micelles had enhanced anti-cancer activity.This work was beneficial to the design of ROS-responsive materials and provides an effective strategy for the application of collaborative anti-tumor therapy.Die Jia Xianbin Ma Yi Lu Xinyi Li Shengxin Hou Yuan Gao Peng Xue Yuejun Kang Zhigang Xu 2021Chinese Chemical Letters2021,32,1:4
2The design and synthesis of dextran-doxorubicin prodrug-based pH-sensitive drug delivery system for improving chemotherapy efficacy显示文摘Tumor cells show acidic conditions compared with normal cells,which further inspires scientist to build nanocarrier responsive to tumor microenvironment(TME)for enhancing tumor therapeutic efficacy.Here,we report a pH-sensitive and biocompatible polyprodrug based on dextran-doxorubicin(DOX)prodrug(DOXDT)for enhanced chemotherapy.Highdensity DOX component was covalently decorated on the nanocarrier and the drug molecules could be effectively released in the acidic tumor tissue/cells,improving chemotherapy efficacy.Specifically,a dextran-based copolymer was preliminarily prepared by one-step atom transfer radical polymerization(ATRP);then DOX was conjugated on the copolymer component via pH-responsive hydrazone bond.The structure of DOXDT can be well-controlled.The resulting DOXDT was able to further self-assemble into nanoscale micelles with a hydration diameter of about 32.4 nm,which presented excellent micellar stability.Compared to lipid-based drug delivery system,the DOXDT prodrug showed higher drug load capacity up to 23.6%.In addition,excellent stability and smaller size of the nanocarrier contributed to better tissue permeability and tumor suppressive effects in vivo.Hence,this amphipathic DOXDT prodrug is promising in the development of translational DOX formulations,which would be widely applied in cancer therapy.Xiaoli Zhang Tian Zhang Xianbin Ma Yajun Wang Yi Lu Die Jia Xiaohua Huang Jiucun Chen Zhigang Xu Feiqiu Wena 2020Asian Journal of Pharmaceutical Sciences2020,15,5:3
3Cylindrical polymer brushes-anisotropic unimolecular micelle drug delivery system for enhancing the effectiveness of chemotherapy显示文摘Polymer systems can be designed into different structures and morphologies according to their physical and chemical performance requirements,and are considered as one of the most promising controlled delivery systems that can effectively improve the cancer therapeutic index.However,the majority of the polymer delivery systems are designed to be simple spherical nanostructures.To explore morphology/size-oriented delivery performance optimization,here,we synthesized three novel cylindrical polymer brushes(CPBs)by atom transfer radical polymerization(ATRP),which were cellulose-g-(CPT-b-OEGMA)(CCO)with different lengths(~86,~40,and~21 nm).The CPBs are composed of bio-degradable cellulose as the carrier,poly(ethylene glycol)methyl ether methacrylate(OEGMA)as hydrophily block,and glutathione(GSH)-responsive hydrophobic camptothecin(CPT)monomer as loaded anticancer drug.By controlling the chain length of the initiator,three kinds of polymeric prodrugs with different lengths(CCO-1,CCO-2,and CCO-3)could be self-organized into unimolecular micelles in water.We carried out comparative studies of three polymers,whose results verified that the shorter CPBs exhibited higher drug release efficiency,more cellular uptake,and enhanced tumor permeability,accompanied by shortened blood circulation time and lower tumor accumulation.As evidenced by in vivo experiments,the shorter CPBs exhibited higher anti-tumor efficiency,revealing that the size advantage has a higher priority than the anisotropic structure advantage.This provided vital information as to design an anisotropic polymer-based drug delivery system for cancer therapy.Shuang Bai Die Jia Xianbin Ma Mengyun Liang Peng Xue Yuejun Kang Zhigang Xu 2021Bioactive Materials2021,6,9:2
4The novel cereblon modulator CC-885 inhibits mitophagy via selective degradation of BNIP3L显示文摘Mitophagy is a degradative pathway that mediates the degradation of the entire mitochondria,and defects in this process are implicated in many diseases including cancer.In mammals,mitophagy is mediated by BNIP3L(also known as NIX)that is a dual regulator of mitochondrial turnover and programmed cell death pathways.Acute myeloid leukemia(AML)cells with deficiency of BNIP3L are more sensitive to mitochondria-targeting drugs.But small molecular inhibitors for BNIP3L are currently not available.Some immunomodulatory drugs(IMiDs)have been proved by FDA for hematologic malignancies,however,the underlining molecular mechanisms are still elusive,which hindered the applications of BNIP3L inhibition for AML treatment.In this study we carried out MS-based quantitative proteomics analysis to identify the potential neosubstrates of a novel thalidomide derivative CC-885 in A549 cells.In total,we quantified 5029 proteins with 36 downregulated in CRBN+/+cell after CC-885 administration.Bioinformatic analysis showed that macromitophagy pathway was enriched in the negative pathway after CC-885 treatment.We further found that CC-885 caused both dose-and time-dependent degradation of BNIP3L in CRBN+/+,but not CRBN−/−cell.Thus,our data uncover a novel role of CC-885 in the regulation of mitophagy by targeting BNIP3L for CRL4CRBN E3 ligase-dependent ubiquitination and degradation,suggesting that CC-885 could be used as a selective BNIP3L degradator for the further investigation.Furthermore,we demonstrated that CC-885 could enhance AML cell sensitivity to the mitochondria-targeting drug rotenone,suggesting that combining CC-885 and mitochondria-targeting drugs may be a therapeutic strategy for AML patients.Bing-bing Hao Xiao-jing Li Xing-long Jia Yu-xing Wang Lin-hui Zhai Duan-zhuo Li Jie Liu Die Zhang Yu-lu Chen Yong-hu Xu Sang-kyu Lee Guo-feng Xu Xiao-hua Chen Yong-jun Dang Bin Liu Min-jia Tan 2020Acta Pharmacologica Sinica2020,41,9:1

6Data augmentation for bias correction in mapping PM_(2.5) based on satellite retrievals and ground observations显示文摘As most air quality monitoring sites are in urban areas worldwide,machine learning models may produce substantial estimation bias in rural areas when deriving spatiotemporal distributions of air pollutants.The bias stems from the issue of dataset shift,as the density distributions of predictor variables differ greatly between urban and rural areas.We propose a data-augmentation approach based on the multiple imputation by chained equations(MICE-DA)to remedy the dataset shift problem.Compared with the benchmark models,MICE-DA exhibits superior predictive performance in deriving the spatiotemporal distributions of hourly PM2.5 in the megacity(Chengdu)at the foot of the Tibetan Plateau,especially for correcting the estimation bias,with the mean bias decreasing from-3.4µg/m3 to-1.6µg/m3.As a complement to the holdout validation,the semi-variance results show that MICE-DA decently preserves the spatial autocorrelation pattern of PM2.5 over the study area.The essence of MICE-DA is strengthening the correlation between PM2.5 and aerosol optical depth(AOD)during the data augmentation.Consequently,the importance of AOD is largely enhanced for predicting PM2.5,and the summed relative importance value of the two satellite-retrieved AOD variables increases from 5.5%to 18.4%.This study resolved the puzzle that AOD exhibited relatively lower importance in local or regional studies.The results of this study can advance the utilization of satellite remote sensing in modeling air quality while drawing more attention to the common dataset shift problem in data-driven environmental research.Tan Mi Die Tang Jianbo Fu Wen Zeng Michael L.Grieneisen Zihang Zhou Fengju Jia Fumo Yang Yu Zhan 2024Geoscience Frontiers2024,15,1:0
