| 1 | Diagnostic value of glypican-3 in serum and liver for primary hepatocellular carcinoma显示文摘AIM:To evaluate the diagnostic value of glypican-3(GPC3) in serum and liver for primary hepatocellular carcinoma(HCC).METHODS:Serum levels of GPC3 and α-fetoprotein(AFP) were measured in 75 patients with primary HCC and 32 patients with liver cirrhosis.Expression of GPC3 and AFP in 58 HCC and 12 cirrhotic specimens was detected with immunohistochemical staining.RESULTS:When the cut-off value of serum GPC3 was set at 300 ng/L,its sensitivity and specificity for HCC were 47.0% and 93.5%,respectively.Among the 14 patients with HCC at stage according to the Barcelona Clinic Liver Cancer staging system,the serum GPC3 level was higher than 300 ng/L in 50%(7/14) patients,the serum AFP level was not ≥ 400 μg/L in any patient.Combined serum AFP and GPC3 significantly increased the sensitivity to the diagnosis of HCC.The GPC3 expression was detected in cytoplasm of HCC cells but not in hepatocytes and bile ducts of benign tumors.Among the 58 HCC patients,the GPC3 was expressed in 100%(28/28) patients with their serum AFP level ≥ 400 μg/L,and in 90%(27/30) patients with their AFP level < 400 μg/L,respectively.The GPC3 was weakly or negatively expressed in all paracarcinomatous and cirrhotic tissue samples.AFP positive HCC cells were only found in 1 out of the 58 HCC patients.CONCLUSION:GPC3 protein is a sensitive and specific serum marker for diagnosis of early HCC.Its expression in liver tissues can be used to discriminate tumor cells from benign hepatic cells. | Hui Liu Li-Jie Zhang Peng Li Yun Zhai Yu-Fen Tan Ning Li Hui-Guo Ding Chun-Feng Qu | 2010 | World Journal of Gastroenterology2010，16，35： | 57 |
| 2 | Transplantation of collagen scaffold with autologous bone marrow mononuclear cells promotes functional endometrium reconstruction via downregulating ΔNp63 expression in Asherman's syndrome显示文摘Asherman's syndrome(AS) is a common disease that presents endometrial regeneration disorder. However, little is known about its molecular features of this aregenerative endometrium in AS and how to reconstruct the functioning endometrium for the patients with AS. Here, we report that ΔNp63 is significantly upregulated in residual epithelial cells of the impaired endometrium in AS; the upregulated-ΔNp63 induces endometrial quiescence and alteration of stemness. Importantly, we demonstrate that engrafting high density of autologous bone marrow mononuclear cells(BMNCs) loaded in collagen scaffold onto the uterine lining of patients with AS downregulates ΔNp63 expression, reverses ΔNp63-induced pathological changes, normalizes the stemness alterations and restores endometrial regeneration. Finally, five patients achieved successful pregnancies and live births. Therefore, we conclude that ΔNp63 is a crucial therapeutic target for AS. | 3 | Effect of ultrasound-guided acupotomy vs electro-acupuncture on knee osteoarthritis:a randomized controlled study显示文摘OBJECTIVE:To investigate the effect of acupotomy on knee osteoarthritis(KOA),compared to electro-acupuncture(EA).METHODS:Sixty KOA patients were randomly divided into 2 groups:ultrasound-guided acupotomy group and EA group;each had 3 weeks' therapy.After the treatment,by contrast before and after therapy,by comparing curative effects among groups,we looked into disease improvement degree through activities of daily living score(ADL),hospital for special surgery index(HSS),visual analogue scales score(VAS) and knee joint's infrared thermal imaging detection.RESULTS:Graded by ADL,the excellent rate in acupotomy group was much higher than EA group;both acupotomy group and EA group had obviouschanges in HSS index before and after the therapy(P < 0.01).And there was remarkable difference in HSS index variation between the groups(P < 0.01).Acupotomy group and EA group showed big difference in pain index before and after treatment(P <0.01).Apparent difference also existed in the comparison among groups(P < 0.01).Both acupotomy group and EA group had apparent changes in infrared thermal imaging detection before and after the treatment(P < 0.01).CONCLUSION:Acupotomy and EA both have significant effects in KOA treatment;the former is better than the latter in relieving pain and improving knee functions. | Ding Yu Wang Yuexiang Shi Xian Luo Yun Gao Yuhong Pan Jingkun | 2016 | Journal of Traditional Chinese Medicine2016，36，4： | 21 |

| 5 | Comparison of Undernutrition Prevalence of Children under 5 Years in China between 2002 and 2013显示文摘Objective To describe the undernutrition status of children under 5-year in China, and study the trend between 2002 and 2013. Methods The study was based on two national surveys. Undernutrition was determined against WHO's 2006 growth standards. The prevalence in 2013 and 2002 was weighted by China sixth National Population Census(2010). The relationship between undernutrition and gender/age groups/different areas use weighted logistic regression. Results The results indicated the overall prevalence of stunting, underweight, and wasting of Chinese children under 5-year was 8.1%, 2.4%, and 1.9% in 2013, respectively. The prevalence of stunting was higher for children aged 12-47 month, while underweight was higher for children aged 48-59 month. The prevalence of undernutrition was higher in rural areas than in urban areas, especially in poor rural areas. There was a decline of stunting, underweight, and wasting between 2002 and 2013 among the children, with greater reduction in rural areas than in urban areas. Conclusion The prevalence of undernutrition of children under 5-year remains high in rural areas especially in poor rural areas in China. | 6 | Sirtuin 3 deficiency exacerbates diabetic cardiomyopathy via necroptosis enhancement and NLRP3 activation显示文摘Sirtuin 3(SIRT3)is a potential therapeutic target for cardiovascular,metabolic,and other aging-related diseases.In this study,we investigated the role of SIRT3 in diabetic cardiomyopathy(DCM).Mice were injected with streptozotocin(STZ,60 mg/kg,ip)to induce diabetes mellitus.Our proteomics analysis revealed that SIRT3 expression in the myocardium of diabetic mice was lower than that of control mice,as subsequently confirmed by real-time PCR and Western blotting.To explore the role of SIRT3 in DCM,SIRT3-knockout mice and 129S1/SvImJ wild-type mice were injected with STZ.We found that diabetic mice with SIRT3 deficiency exhibited aggravated cardiac dysfunction,increased lactate dehydrogenase(LDH)level in the serum,decreased adenosine triphosphate(ATP)level in the myocardium,exacerbated myocardial injury,and promoted myocardial reactive oxygen species(ROS)accumulation.Neonatal rat cardiomyocytes were transfected with SIRT3 siRNA,then exposed to high glucose(HG,25.5 mM).We found that downregulation of SIRT3 further increased LDH release,decreased ATP level,suppressed the mitochondrial membrane potential,and elevated oxidative stress in HG-treated cardiomyocytes.SIRT3 deficiency further raised expression of necroptosis-related proteins including receptor-interacting protein kinase 1(RIPK1),RIPK3,and cleaved caspase 3,and upregulated the expression of inflammation-related proteins including NLR family pyrin domain-containing protein 3(NLRP3),caspase 1 p20,and interleukin-1βboth in vitro and in vivo.Collectively,SIRT3 deficiency aggravated hyperglycemia-induced mitochondrial damage,increased ROS accumulation,promoted necroptosis,possibly activated the NLRP3 inflammasome,and ultimately exacerbated DCM in the mice.These results suggest that SIRT3 can be a molecular intervention target for the prevention and treatment of DCM. | Shu Song Yue Ding Guo-liang Dai Yue Zhang Meng-ting Xu Jie-ru Shen Ting-ting Chen Yun Chen Guo-liang Meng | 2021 | Acta Pharmacologica Sinica2021，42，2： | 14 |
| 7 | Evaluation of health benefit using Ben MAP-CE with an integrated scheme of model and monitor data during Guangzhou Asian Games显示文摘Guangzhou is the capital and largest city(land area:7287 km2)of Guangdong province in South China.The air quality in Guangzhou typically worsens in November due to unfavorable meteorological conditions for pollutant dispersion.During the Guangzhou Asian Games in November 2010,the Guangzhou government carried out a number of emission control measures that significantly improved the air quality.In this paper,we estimated the acute health outcome changes related to the air quality improvement during the 2010 Guangzhou Asian Games using a next-generation,fully-integrated assessment system for air quality and health benefits.This advanced system generates air quality data by fusing model and monitoring data instead of using monitoring data alone,which provides more reliable results.The air quality estimates retain the spatial distribution of model results while calibrating the value with observations.The results show that the mean PM_2.5concentration in November 2010 decreased by 3.5μg/m^3 compared to that in 2009 due to the emission control measures.From the analysis,we estimate that the air quality improvement avoided 106 premature deaths,1869 cases of hospital admission,and 20,026 cases of outpatient visits.The overall cost benefit of the improved air quality is estimated to be 165 million CNY,with the avoided premature death contributing 90%of this figure.The research demonstrates that Ben MAP-CE is capable of assessing the health and cost benefits of air pollution control for sound policy making. | Dian Ding Yun Zhu Carey Jang Che-Jen Lin Shuxiao Wang Joshua Fu Jian Gao Shuang Deng Junping Xie Xuezhen Qiu | 2016 | Journal of Environmental Sciences2016，28，4： | 13 |

| 10 | Factors Associated with Thrombolysis Outcome in Ischemic Stroke Patients with Atrial Fibrillation显示文摘The outcome of early intravenous thrombolysis for ischemic stroke in patients with atrial fibrillation(AF)is worse than that without thrombosis. How to increase the efficacy of intravenous thrombolysis for AF-related ischemic stroke remains largely unknown. In this study, we investigated factors that influence the effect of intravenous thrombolysis in these patients. Our results showed that thrombolysis was independently associated with a favorable outcome(P \ 0.001) and did not influence the mortality of AF-related ischemic stroke, although it increased the risk of hemorrhage within 24 h after treatment. Risk factors for a poor outcome at admission were:heart failure(P = 0.045); high systolic pressure(P = 0.039); high blood glucose(P = 0.030); and a high National Institutes of Health Stroke Scale(NIHSS) score(P \ 0.001). Moreover, high systolic pressure at admission(P = 0.007), high blood glucose(P = 0.027), and a high NIHSS score(P \ 0.001) were independent risk factors for mortality at 3 months. Besides thrombolysis, a high NIHSS score(P = 0.006) and warfarin taken within 48 h before stroke onset(P = 0.032) were also independent risk factors for symptomatic hemorrhage within 24 h after treatment. | 12 | Artificial intelligence system of faster region-based convolutional neural network surpassing senior radiologists in evaluation of metastatic lymph nodes of rectal cancer显示文摘Background: An artificial intelligence system of Faster Region-based Convolutional Neural Network (Faster R-CNN) is newly developed for the diagnosis of metastatic lymph node (LN) in rectal cancer patients. The primary objective of this study was to comprehensively verify its accuracy in clinical use. Methods: Four hundred fourteen patients with rectal cancer discharged between January 2013 and March 2015 were collected from 6 clinical centers, and the magnetic resonance imaging data for pelvic metastatic LNs of each patient was identified by Faster R-CNN. Faster R-CNN based diagnoses were compared with radiologist based diagnoses and pathologist based diagnoses for methodological verification, using correlation analyses and consistency check. For clinical verification, the patients were retrospectively followed up by telephone for 36 months, with post-operative recurrence of rectal cancer as a clinical outcome;recurrence-free survivals of the patients were compared among different diagnostic groups, by methods of Kaplan-Meier and Cox hazards regression model. Results: Significant correlations were observed between any 2 factors among the numbers of metastatic LNs separately diagnosed by radiologists, Faster R-CNN and pathologists, as evidenced by rradiologist-Faster R-CNN of 0.912, rPathologist-radiologist of 0.134, and rPathologist-Faster R-CNN of 0.448 respectively. The value of kappa coefficient in N staging between Faster R-CNN and pathologists was 0.573, and this value between radiologists and pathologists was 0.473. The 3 groups of Faster R-CNN, radiologists and pathologists showed no significant differences in the recurrence-free survival time for stage N0 and N1 patients, but significant differences were found for stage N2 patients. | 13 | Design and demonstration of a next-generation air quality attainment assessment system for PM_(2.5)and O_3显示文摘Due to the increasingly stringent standards, it is important to assess whether the proposed emission reduction will result in ambient concentrations that meet the standards. The Software for Model Attainment Test—Community Edition(SMAT-CE) is developed for demonstrating attainment of air quality standards of O3 and PM2.5. SMAT-CE improves computational efficiency and provides a number of advanced visualization and analytical functionalities on an integrated GIS platform. SMAT-CE incorporates historical measurements of air quality parameters and simulated air pollutant concentrations under a number of emission inventory scenarios to project the level of compliance to air quality standards in a targeted future year. | 14 | Deep neural network-assisted computed tomography diagnosis of metastatic lymph nodes from gastric cancer显示文摘Background:Artificial intelligence-assisted image recognition technology is currently able to detect the target area of an image and fetch information to make classifications according to target features.This study aimed to use deep neural netAVorks for computed tomography(CT)diagnosis of perigastric metastatic lymph nodes(PGMLNs)to simulate the recognition of lymph nodes by radiologists,and to acquire more accurate identification results.Methods:A total of 1371 images of suspected lymph node metastasis from enhanced abdominal CT scans were identified and labeled by radiologists and were used with 18,780 original images for faster region-based convolutional neural networks(FR-CNN)deep learning.The identification results of 6000 random CT images from 100 gastric cancer patients by the FR-CNN were compared with results obtained from radiologists in terms of their identification accuracy.Similarly,1004 CT images with metastatic lymph nodes that had been post-operatively confirmed by pathological examination and 11,340 original images were used in the identification and learning processes described above.The same 6000 gastric cancer CT images were used for the verification,according to which the diagnosis results were analyzed.Results:In the initial group,precision-recall curves were generated based on the precision rates,the recall rates of nodule classes of the training set and the validation set;the mean average precision(mAP)value was 0.5019.To verify the results of the initial learning group,the receiver operating characteristic curves was generated,and the corresponding area under the curve(AUC)value was calculated as 0.8995.After the second phase of precise learning,all the indicators were improved,and the mAP and AUC values were 0.7801 and 0.9541,respectively.Conclusion:Through deep learning,FR-CNN achieved high judgment effectiveness and recognition accuracy for CT diagnosis of PGMLNs. | Yuan Gao Zheng-Dong Zhang Shuo Li Yu-Ting Guo Qing-Yao Wu Shu-Hao Liu Shu-Jian Yang Lei Ding Bao-Chun Zhao Shuai Li Yun Lu | 2019 | Chinese Medical Journal2019，，23： | 11 |

A new method of electrostatic precipitation of the corona discharges with spraying water(CDSW) was introduced. A new electrode configuration and the circulation spraying of water were employed in the method. The efficient electrostatic precipitation for adhesive and fine dusts can be accomplished without any drain water during a long operating period. The fundamental structure, discharge characteristics, mechanism of spraying and precipitation principle of the electrostatic precipitation using CDSW were described and analyzed. The V I characteristics, spraying state, supplying water quantity, influence of temperature and clean of the electrodes were researched in series experiments. The treating effects of circulating spraying using the corona plasma at the same time of electrostatic precipitation were investigated. | 16 | Expression of VEGFR2 and NRP-1 in non-small cell lung cancer and their clinical significance显示文摘Objective: Vascular-targeted therapy is gradually becoming more appealing for patients with lung cancer. It is unclear whether vascular endothelial growth factor receptor 2(VEGFR2) and neuropilin-1(NRP-1) can be biomarkers for clinical treatment. We aimed to investigate the expression levels of VEGFR2 and NRP-1 in human non-small cell lung cancer(NSCLC) and their clinical significance by observing patient prognosis. Methods: VEGFR2 and NRP-1 were assessed by immunohistochemistry(IHC) in 40 patients with NSCLC and in 10 patients with benign lesions of lung; kinase insert domain receptor(KDR) and NRP-1 copy number gain(CNG) was assessed by fluorescence in situ hybridization(FISH). The distributions of overall survival(OS) and progression-free survival(PFS) were estimated using the Kaplan-Meier method and compared between groups by log-rank test.Results: Rates of positive immunostaining for VEGFR2 and NRP-1 were 58% and 55%, respectively. KDR and NRP-1 CNG(+) were detected in 32.5% and 30% of tumors, respectively. Levels of both VEGFR2 and NRP-1 in lung tumors were significantly different than in the control tissue(χ2=11.22, P=0.001; χ2=9.82, P=0.001, respectively); similar results were obtained using CNGs(χ2=4.39, P=0.036; χ2=3.95, P=0.046, respectively). Statistically significant correlations were observed with histological grade, clinical TNM stage and the lymph node status(P<0.05), but not age, gender or pathology type(P>0.05). VEGFR2 showed a strong correlation with NRP-1(Rs=0.68, P=0.00); similar results were observed with KDR and NRP-1 CNG(Rs=0.32, P=0.04). Significant differences in OS and PFS were observed between the groups with higher VEGFR2 and NRP-1 and those with lower expression(P<0.05). Conclusions: According to these data, VEGFR2 and NRP-1 are highly expressed in NSCLC. | 19 | Embryonic endothelial evolution towards first hematopoietic stem cells revealed by single-cell transcriptomic and functional analyses显示文摘Hematopoietic stem cells(HSCs)in adults are believed to be born from hemogenic endothelial cells(HECs)in mid-gestational embryos.Due to the rare and transient nature,the HSC-competent HECs have never been stringently identified and accurately captured,let alone their genuine vascular precursors.Here,we first used high-precision single-cell transcriptomics to unbiasedly examine the relevant EC populations at continuous developmental stages with intervals of 0.5 days from embryonic day(E)9.5 to E11.0.As a consequence,we transcriptomically identified two molecularly different arterial EC populations and putative HSCprimed HECs,whose number peaked at El 0.0 and sharply decreased thereafter,in the dorsal aorta of the aorta-gonadmesonephros(AGM)region.Combining computational prediction and in vivo functional validation,we precisely captured HSCcompetent HECs by the newly constructed Neurl3-EGFP reporter mouse model,and realized the enrichment further by a combination of surface markers(Procr+Kit+CD44+,PK44).Surprisingly,the endothelial-hematopoietic dual potential was rarely but reliably witnessed in the cultures of single HECs.Noteworthy,primitive vascular ECs from E8.0 experienced two-step fate choices to become HSC-primed HECs,namely an Initial arterial fate choice followed by a hemogenic fate conversion.This finding resolves several previously observed contradictions.Taken together,comprehensive understanding of endothelial evolutions and molecular programs underlying HSC-primed HEC specification in vivo will facilitate future investigations directing HSC production in vitro. | Siyuan Hou Zongcheng Li Xiaona Zheng Yun Gao Ji Dong Yanli Ni Xiaobo Wang Yunqiao Li Xiaochen Ding Zhilin Chang Shuaili Li Yuqiong Hu Xiaoying Fan Yu Hou Lu Wen Bing Liu Fuchou Tang Yu Lan | 2020 | Cell Research2020，30，5： | 8 |
